Neutrophyl function in children with chondrodystrophia calcificans congenita: evidence for defective chemotaxis.

Some neutrophil functions (superoxide anion production, bactericidal activity and chemotaxis) have been examined in four children with Chondrodystrophia Calcificans Congenita. Superoxide anion production and bactericidal activity were in the normal range, but a defect in cellular chemotaxis has been proved. This finding account for predisposition to infection which is frequently present in these patients, and may include this disease among those syndromes in which a deficiency of the immune system is associated with growth failure.

[Drug-induced changes in chemotaxis under normal conditions and in rheumatoid arthritis].

The effects of agents used in RA treatment, various drugs, RF, rheumatoid nodule and synovial fluid was studied on chemotaxis of PMNs. NSAIDS, corticosteroids, theophylline, colchicine, SOD, RF, rheumatoid nodule and synovial fluid were found to inhibit the chemotactic responsiveness while AMPc, GMPc, PGEI and immunodulator drugs enhanced chemotaxis. The results support the hypothesis that drugs tested may modulate chemotactic function by affecting cellular microtubules assembly and/or GMPc accumulation.

CXCR6 sustains TRM-driven psoriasis relapse by CXCL16 chemotaxis and curcumol targeting.

INTRODUCTION: Psoriasis is a chronic inflammatory skin disease characterized by a high relapse rate, primarily driven by tissue-resident memory T cells (TRM). OBJECTIVES: This study aims to elucidate that the CXCR6/CXCL16 axis is a key regulator of TRM persistence and psoriasis relapse, and demonstrates that curcumol can mitigate psoriasis relapse via this axis and TRM modulation. METHODS: By integrating single-cell RNA sequencing, spatial transcriptomics, and a CXCR6-knockout mouse model, we analyzed the characteristics of TRM cell subsets and the functional role of CXCR6 in psoriatic lesio

Lyn Phosphorylates and Controls ROR1 Surface Dynamics During Chemotaxis of CLL Cells.

Chronic lymphocytic leukemia (CLL) and mantle cell lymphoma (MCL) are malignancies characterized by the dependence on B-cell receptor (BCR) signaling and by the high expression of ROR1, the cell surface receptor for Wnt-5a. Both, BCR and ROR1 are therapeutic targets in these diseases and the understanding of their mutual cross talk is thus of direct therapeutic relevance. In this study we analyzed the role of Lyn, a kinase from the Src family participating in BCR signaling, as a mediator of the BCR-ROR1 crosstalk. We confirm the functional interaction between Lyn and ROR1 and demonstrate that

More Than Just Attractive: How CCL2 Influences Myeloid Cell Behavior Beyond Chemotaxis.

Monocyte chemoattractant protein-1 (MCP-1/CCL2) is renowned for its ability to drive the chemotaxis of myeloid and lymphoid cells. It orchestrates the migration of these cell types both during physiological immune defense and in pathological circumstances, such as autoimmune diseases including rheumatoid arthritis and multiple sclerosis, inflammatory diseases including atherosclerosis, as well as infectious diseases, obesity, diabetes, and various types of cancer. However, new data suggest that the scope of CCL2's functions may extend beyond its original characterization as a chemoattractant. …

CIP4 controls CCL19-driven cell steering and chemotaxis in chronic lymphocytic leukemia.

Solid tumor dissemination relies on the reprogramming of molecular pathways controlling chemotaxis. Whether the motility of nonsolid tumors such as leukemia depends on the deregulated expression of molecules decoding chemotactic signals remains an open question. We identify here the membrane remodeling F-BAR adapter protein Cdc42-interacting protein 4 (CIP4) as a key regulator of chemotaxis in chronic lymphocytic leukemia (CLL). CIP4 is expressed at abnormally high levels in CLL cells, where it is required for CCL19-induced chemotaxis. Upon CCL19 stimulation of CLL cells, CIP4 associates with …

Chemotaxis-promoting and adhesion properties of human tonsillar follicular dendritic cell clusters.

Lymph follicles are globular and compact due to aggregation of lymphoid cells on follicular dendritic cells (FDC). To probe the mechanisms underlying this accumulation of cells, we analyse here the role played by FDCs in attracting and binding cells. FDCs prepared from human tonsils by mild separation techniques appeared in the form of clusters (FDC clusters), where, via cytoplasmic extensions, they enveloped lymphoid cells. Using Boyden's chambers, we demonstrated that these FDC clusters produced one or more chemoattractants capable of inducing chemotaxis of lymphoid cells. Supernatants of FD…

Beta-endorphin and met-enkephalin stimulate human peripheral blood mononuclear cell chemotaxis.

The neuropeptides beta-endorphin and met-enkephalin are potent analgesics and have a broad spectrum of biologic activities including the recently described alterations of lymphocyte proliferation and antibody production. The current study demonstrates that beta-endorphin and met-enkephalin stimulate human mononuclear cell chemotaxis, as measured by the in vitro leading front assay for migration. The response to both beta-endorphin and met-enkephalin was bimodal, with peak activities occurring at 10(-12) M and 10(-8) M. The distance migrated in response to optimal concentrations of beta-endorph…

Severe combined immunodeficiency associated with hyperimmunoglobulinaemia E, eosinophilia and impaired neutrophil chemotaxis.

A boy with severe combined immunodeficiency was found to have coexistent hyperimmunoglobulinaemia E, eosinophilia and impaired neutrophil chemotaxis. Based on the literature, a deficient regulatory function of the T-cell system is proposed as the basic defect leading to the observed impairment of the cellular and humoral immunity and, probably through hyper-immunoglobulinaemia E, to defective neutrophil chemotaxis.

A biohybrid nanovesicle hijacks inflammatory chemotaxis to deliver colchicine for myocardial infarction therapy.

INTRODUCTION: Myocardial infarction (MI) triggers an excessive inflammatory response that drives adverse cardiac remodeling. Although colchicine has shown clinical promise, its narrow therapeutic window and lack of target specificity limit its efficacy. To address these limitations, we developed a biohybrid nanovesicle (MM-LP@COL) that encapsulates colchicine, aiming to leverage natural inflammatory tropism for site-specific drug delivery. METHODS: MM-LP@COL nanovesicles were fabricated by fusing macrophage membranes with liposomes. Physicochemical characterization was performed using DLS, TE

Timd4+ cardiac resident macrophages promote neutrophil chemotaxis and exacerbate sepsis-associated myocardial injury.

This study investigated the role of T-cell immunoglobulin and mucin domain-containing protein 4 (Timd4) in sepsis-associated myocardial injury. A murine sepsis model was established by cecal ligation and puncture. Timd4 was modulated in vivo by adeno-associated virus serotype 9-mediated overexpression or prophylactic administration of a Timd4-neutralizing antibody. Cardiac function, macrophage phagocytosis, chemokine expression, and neutrophil infiltration were evaluated. Sepsis reduced Timd4 expression and the proportion of Timd4-positive cardiac-resident macrophages (CRMs) in the heart. Timd

Cholangiocarcinoma-derived SPP1 drives tumor progression by inducing macrophage chemotaxis and M2 polarization.

Cholangiocarcinoma, a hepatobiliary malignancy, has a poor prognosis and treatment resistance. This study integrated spatial transcriptome and conditional knockdown models to explore the tumor microenvironment (TME) regulatory mechanism. We found that macrophages were located at the boundary between cholangiocarcinoma and normal tissue. Cholangiocarcinoma - secreted SPP1 induced blood monocytes to migrate toward the tumor tissue and polarize into M2 macrophages through the CD44 - PI3K - AKT pathway. In vitro and in vivo experiments confirmed this process. Macrophages polarized by SPP1 promoted

C5a enhances inflammation and chemotaxis of γδ T cells in malignant pleural effusion.

BACKGROUND: The inhibitory effect of γδT17 cells on the formation of murine malignant pleural effusions (MPE) has been established. However, there is limited understanding regarding the phenotypic characterization of γδ T cells in MPE patients and their recruitment to the pleural cavity. METHODS: We quantified γδ T cell prevalence in pleural effusions and corresponding peripheral blood from malignant and benign patients using immunohistochemistry and flow cytometry. The expression of effector memory phenotype, stimulatory/inhibitory/chemokine receptors and cytokines on γδ T cells in MPE was a

Involvement of cyclic nucleotide-gated channels in soybean cyst nematode chemotaxis and thermotaxis.

The soybean cyst nematode (SCN) is one of the most damaging pests affecting soybean production. SCN displays important host recognition behaviors, such as hatching and infection, by recognizing several compounds produced by the host. Therefore, controlling SCN behaviors such as chemotaxis and thermotaxis is an attractive pest control strategy. In this study, we found that cyclic nucleotide-gated channels (CNG channels) regulate SCN chemotaxis and thermotaxis and Hg-tax-2, a gene encoding a CNG channel, is an important regulator of SCN behavior. Gene silencing of Hg-tax-2 and treatment with a C…

β-Defensin 19/119 mediates sperm chemotaxis and is associated with idiopathic infertility.

Sperm chemotaxis is required for guiding sperm toward the egg. However, the molecular identity of physiological chemoattractant and its involvement in infertility remain elusive. Here, we identify DEFB19/119 (mouse/human orthologs) as a physiological sperm chemoattractant. The epithelia of the female reproductive tract and the cumulus-oocyte complex secrete DEFB19/119 that elicits calcium mobilization via the CatSper channel and induces sperm chemotaxis in capacitated sperm. Manipulating the level of DEFB19 in mice determines the number of sperm arriving at the fertilization site. Importantly,…
