Chicken Infectious Anemia Virus Markedly Enhances the Pathogenicity of Infectious Bronchitis Virus-Infected Chickens.

Chicken infectious anemia virus (CIAV) and infectious bronchitis virus (IBV) represent two of the most economically significant pathogens in the global poultry production. In recent years, coinfections involving CIAV and IBV have become increasingly prevalent in clinical scenarios across Southern China and globally. Nevertheless, the underlying synergistic pathogenic mechanisms remain largely elusive. This study establishes a coinfection model using specific pathogen-free (SPF) chickens infected with field-derived strains CIAV GDHY230813 and IBV CK/CK/GX/LA/071423 to systematically investigate…

Genomic Sequence and Pathogenicity of the Chicken Anemia Virus Isolated From Chicken in Yunnan Province, China.

Chicken anemia virus (CAV), which has been reported in many countries, causes severe anemia and immunosuppression in chickens. In this study, a CAV strain YN04 belonging to genotype A was first identified from infected chickens in Yunnan province, China. Moreover, the animal infection experiments further confirmed that the strain YN04 is a highly pathogenic strain, which can cause 86.67% mortality in chickens in the infection group. The mean death time of infected chickens was 13.1 days post infection (dpi). CAV infection induced severe anemia with significant decrease in packed cell volume (P…

Novel Gyroviruses, including Chicken Anaemia Virus, in Clinical and Chicken Samples from South Africa.

Introduction. Chicken anaemia virus, CAV, was until recently the only member of the Gyrovirus genus. 6 novel gyroviruses, AGV2, HGyV1, and GyV3-6, have since been discovered in human and chicken samples. Methods. PCR amplification of the VP2 gene was used to detect AGV2/HGyV1, GyV3, and CAV in a range of clinical samples including stool, respiratory, CSF, and HIV-positive plasma. Screening of fresh local chicken meat was also performed. Results. AGV2/HGyV1 or GyV3 was detected in stools from healthy children (17/49, 34.7%) and patients with diarrhoea (22/149, 14.8%). 1.2% (3/246) nasopharyngea

Pathological and immunohistochemical study of chickens with co-infection of Marek's disease virus and chicken anaemia virus.

Chicken anaemia virus (CAV) is the most important confounding pathogen in Marek's disease virus (MDV) infection. The effect of CAV co-infection at 4 weeks of age after inoculation of virulent MDV (vMDV, KS strain) or very virulent MDV (vvMDV, Md/5 strain) in 1-day-old chicks was investigated by pathological and immunohistochemical studies. CAV increased the mortality rates induced by vMDV or vvMDV. The packed cell volume was reduced significantly in vMDV-CAV infection; however, no reduction or non-significant reduction was observed in vMDV infection. Bone marrow hypoplasia was related to CAV c

Lethality and bone alterations in chicken embryos and newly hatched chickens given bone-active agents.

Studies were undertaken to assess the chicken embryo and newly hatched chicken as models for studying the effects of bone-active agents. Initially, 1,25-dihydroxycholecaliferol (1,25[OH]2D3), sodium fluoride (NaF), parathyroid extract, epidermal growth factor, and prostaglandin E2, were tested for lethality over a broad dose range. One or 3 injections of 1,25(OH)2D3 into the yolk sac of chicken embryos resulted in death of embryos given greater than or equal to 0.1 ng/injection, whereas 0.01 ng was tolerated by the embryos. Administering 1,25(OH)2D3 intraperitoneally to newly hatched chickens …

"Natural" antibodies to chicken MHC antigens are present in mice, rats, humans, alligators and allogeneic chickens.

In the absence of any deliberate immunization, mice, rats, humans and alligators all have detectable titers of antibody against chicken red blood cells (CRBC's). Remarkably, this antibody is directed predominantly against private or public determinants of MHC proteins on the CRBC's, and little or no antibody is directed against species-specific determinants on MHC or other proteins, including other polymorphic blood group antigens. In chickens, "natural" antibody can be detected against CRBC's from all chickens differing at the MHC locus, but "natural" antibodies against other polymorphic anti…

Exploiting RAG1-deficient chickens to construct a single-cell transcriptional atlas of chicken heterophils during inflammation.

INTRODUCTION: Avian heterophils naturally lack myeloperoxidase (MPO), making chicken a unique evolutionary model for investigating MPO-independent innate immune strategies. However, the molecular programs governing heterophil maturation and activation remain largely unexplored. METHODS: Using single-cell RNA sequencing in a RAG1-deficient chicken model, we mapped heterophil maturation during emergency granulopoiesis and identified eight transcriptionally distinct subsets (H0-H7). We inferred a developmental trajectory spanning transcriptionally distinct heterophil subsets, from early subsets

Synergy of Subgroup J Avian Leukosis Virus and Chicken Infectious Anemia Virus Enhances the Pathogenicity in Chickens.

Subgroup J avian leukemia virus (ALV-J) and chicken infectious anemia virus (CIAV) are widely acknowledged as significant immunosuppressive pathogens that commonly co-infect chickens, causing substantial economic losses in the poultry industry. However, whether co-infection of ALV-J and CIAV have synergistic pathogenicity remains uncertain. To explore their synergistic pathogenesis, we established a co-infection model of ALV-J and CIAV in HD11 cells and specific-pathogen-free (SPF) chickens. We discovered that ALV-J and CIAV can synergistically promote the secretion of IL-6, IL-10, IFN-α, and …

Newcastle Disease Virus Vectored Chicken Infectious Anaemia Vaccine Induces Robust Immune Response in Chickens.

Newcastle disease virus (NDV) strain R2B, with an altered fusion protein cleavage site, was used as a viral vector to deliver the immunogenic genes VP2 and VP1 of chicken infectious anaemia virus (CIAV) to generate a bivalent vaccine candidate against these diseases in chickens. The immunogenic genes of CIAV were expressed as a single transcriptional unit from the NDV backbone and the two CIA viral proteins were obtained as separate entities using a self-cleaving foot-and-mouth disease virus 2A protease sequence between them. The recombinant virus (rR2B-FPCS-CAV) had similar growth kinetics as…

Recombinant chicken IL-6 does not activate heterophils isolated from day-old chickens in vitro.

Pro-inflammatory cytokines are produced as part of innate immunity. Increased resistance to extraintestinal Salmonella enteritidis (SE) has been associated with an increase in heterophil pro-inflammatory cytokine gene expression. Invasion of chicken epithelial cells by SE induces an 8- to 10-fold increase in interleukin (IL) -6 production. Infection with SE induces an influx of heterophils to the site of infection; therefore, we hypothesize heterophils would be responsive to IL-6. The objective was to determine the effects of COS cell-derived recombinant chicken interleukin 6 (rChIL-6) on in v

Optimal treatment of Ascaridia galli-infected chickens with salts of trace elements and a kinetic model for chicken growth.

Data from seven experiments with Ascaridia galli-infected chickens have been considered. The results of treatment with neutral and basic copper, zinc and copper-zinc salts and inorganic and organic manganese compounds have been compared. An optimal therapy, containing a pure Cu basic salt (Cu2(OH)3Cl) and an organic Mn compound (2Gly.MnCl2.2H2O), is proposed to correct mineral deficiencies and pathological symptoms and to ensure lower mortality and higher gains in body weight. A mathematical model has been proposed for the growth of a healthy chicken. The relative rates for two growth stages h…

Bacterial consortium construction for inhibition of Salmonella in chickens.

Salmonella enterica is one of the leading causes of bacterial enteritis in humans with consumption of contaminated poultry products being the leading cause of Salmonella infection. Salmonella is often considered a commensal organism within the poultry gut due to its asymptomatic carriage in adult birds making it difficult to clear by the host immune system once established. Understanding the niche Salmonella occupies will aid in the determination or selection for probiotic organisms with overlapping niche occupation metabolically or spatially for inhibition or exclusion of Salmonella within th

A novel spontaneous cytotoxic T-cell lymphoma in chickens.

Chicken lymphomas are generally classified into three virus-induced tumors: Marek's disease (MD), lymphoid leukosis, and reticuloendotheliosis, with MD being the most common T-cell lymphoma in commercial poultry. In this study, we describe 23 cases of a novel cytotoxic T-cell lymphoma affecting layer chickens aged >1 year in Japan that is distinct from previously known chicken lymphomas. These cases were initially misidentified as MD during routine poultry inspection but were later confirmed as a distinct and novel form of lymphoma. The average incidence of this lymphoma was 0.016%. Histologic…
