Tympanic membrane regeneration using a water-soluble chitosan patch.

Chronic otitis media or tympanic membrane (TM) perforation is one of the most common otologic diseases. Surgical tympanoplasty remains the best treatment option despite the fact that paper patches are frequently used. Although paper patches are not biocompatible or effective, tympanoplasty is an expensive, complex surgery. Tissue engineering techniques offer a new treatment strategy for TM regeneration. In this study, novel tissue-engineered artificial eardrums were fabricated from water-soluble chitosan, which is known to be a good wound-healing biomaterial. The characteristics, cytotoxicity,

GDNF blended chitosan nerve guides: an in vivo study.

Chitosan nerve guides are currently being utilized to repair damaged or injured peripheral nerves. To enhance the nerve regeneration process, instead of using the material alone, researchers are focusing on blending different proteins or molecules with chitosan to facilitate nerve repair and regeneration. In our study, we have blended chitosan with glial cell-line derived neurotrophic factor and laminin within our nerve guides (GLC). The rat sciatic nerve injury model was used to test these nerve guides and histologically evaluated at 6, 9, and 12 weeks. Histologically at 6 weeks, the axon are

Physicochemical properties and sensing ability of metallophthalocyanines/chitosan nanocomposites.

Electroactive nanostructured films of chitosan (Ch) and tetrasulfonated metallophthalocyanines containing nickel (NiTsPc), copper (CuTsPc), and iron (FeTsPc) were produced via the electrostatic layer-by-layer (LbL) technique. The multilayer formation was monitored with UV-vis spectroscopy by measuring the increase of the Q-band absorption from metallophthalocyanines. Results from transmission and reflection infrared spectroscopy suggested specific interactions between SO(3)(-) groups from metallophthalocyanines and NH(3)(+) from chitosan. The electroactive multilayered films assembled onto an

Heparin immobilized chitosan--poly ethylene glycol interpenetrating network: antithrombogenicity.

This work deals with the synthesis and blood compatibility studies of Heparin immobilized chitosan--polyethyleneglycol (Chit-PEG) hydrogels for various biomedical applications. Chit-PEG interpenetrating net work (IPN) had been synthesised by crosslinking different ratios of chitosan with glutaraldehyde using schiffs base reaction mechanism and interpenetrating polyethyleneglycol (PEG) to form hydrogen bonding between the amino hydrogen in chitosan and polyether oxygen. An optimum gel combination was selected from the IPN of Chit-PEG and used for bonding heparin. This modified gel had dramatica

Chitosan in regenerative medicine: a translational perspective on biomaterial design.

Chitosan is an aminopolysaccharide platform with broad relevance to regenerative medicine because its charge density, solubility, degradation, processability, and biological interactions can be adjusted through source selection, deacetylation chemistry, molecular-weight control, derivatization, and formulation. However, this tunability is accompanied by substantial source- and process-dependent variability, which represents a central translational challenge: materials labelled as chitosan can differ markedly in degree of deacetylation, molecular weight, pattern of acetylation, polydispersity,

Chitosan-Lignin Cryogel Beads for the Adsorption of Diclofenac in Water.

The production, use, and disposal of pharmaceuticals contribute to widespread pollution of the aquatic environment. Conventional wastewater treatment plants are not designed to completely remove bioactive pharmaceuticals such as diclofenac, posing risks to aquatic ecosystems and drinking water sources. To address this challenge, we report the production of biogenic adsorbent beads for enhanced removal of anionic contaminants from water. The beads consist of glutaraldehyde-cross-linked chitosan (or quaternary amine-functionalized chitosan) combined with lignin, and a macroporous structure was i

Chitosan Hydrogels for Antibiotic Remediation and Dye Removal: A Review.

The co-contamination of aquatic environments by antibiotic residues and organic dyes poses a serious threat to ecological security and human health, underscoring the urgent need for high-efficiency, recyclable, and environmentally benign adsorbents. Chitosan, a naturally occurring alkaline polysaccharide rich in reactive functional groups, has attracted considerable attention in water treatment applications. Nevertheless, its practical use is often constrained by intrinsic limitations, including poor stability in acidic media, inadequate mechanical strength, and difficulties in solid-liquid se

Effect of chitosan and chlorhexidine on demineraslised dentin to enhance adhesion.

The bond strength (BS) and failure status of demineralized dentin following the application of chitosan solution and chlorhexidine (CHX) is of interest to dentists. A total of 30 non-pathological extracted premolar teeth were subjected to caries induction through the pH cycle procedure. The teeth were divided equally into 3 groups as such as group I: control (distilled water), Group II-2.5% chitosan solution and Group III - chlorhexidine (CHX). Later teeth were subjected to a microtensile bond strength test (µTBS). The failure mode was assessed using stereo microscope. The bond strength of the

Stabilization of chitosan-based nanomedicines in cancer therapy: a review.

Chitosan (CS), a versatile and alkaline polysaccharide, has gained significant attention in nanomedicine due to its biocompatibility and biodegradability. In recent years, its applications in cancer therapy, particularly for the delivery of chemotherapeutic drugs, diagnostic agents, and genes, have advanced considerably. However, many CS-based nanomedicines suffer from poor stability in biological fluids, especially under physiological conditions. The neutral pH and the presence of electrolytes in physiological environments reduce the charge density of CS, which can account for this applicatio

The Application of Mucoadhesive Chitosan Nanoparticles in Nasal Drug Delivery.

Mucosal delivery of antigens can induce both humoral and cellular immune responses. Particularly, the nasal cavity is a strongly inductive site for mucosal immunity among several administration routes, as it is generally the first point of contact for inhaled antigens. However, the delivery of antigens to the nasal cavity has some disadvantages such as rapid clearance and disposition of inhaled materials. For these reasons, remarkable efforts have been made to develop antigen delivery systems which suit the nasal route. The use of nanoparticles as delivery vehicles enables protection of the an

Chitosan functionalized nanocochleates for enhanced oral absorption of cyclosporine A.

It remains a significant challenge to overcome the poor permeability of cyclosporine A and enhance its oral absorption. In this study, we have identified a positively charged chitosan that is able to induce coiling up of anionic lipids to form nanocochleates with an average size of 114.2 ± 0.8 nm, without the need for calcium ions. These functional chitosan-induced nanocochleates enhanced gastrointestinal absorption of cyclosporine A, up to a 3-fold increase in oral bioavailability. A fluorescence-labeling study confirmed that absorption mainly occurred in the duodenum and jejunum. Transport s

The effect of pH on the bonding of Cu2+ and chitosan-montmorillonite composite.

Based on the contradict statements on the mechanisms of the bonding between metal ions and chitosan based adsorbent, this paper present studies on the Cu2+ adsorption by the chitosan-montmorillonite composite. The in situ attenuated total reflectance Fourier transform infrared spectroscopy and two dimensional correlation analysis were used to record the spectra during the adsorption and the pH perturbation, and analyze the interaction sequence of the groups related to the adsorption, respectively. The results showed that the bonding with -NH3+ might attribute the adsorption capacity greater th

Application of Chitosan, Chitooligosaccharide, and Their Derivatives in the Treatment of Alzheimer's Disease.

Classic hypotheses of Alzheimer's disease (AD) include cholinergic neuron death, acetylcholine (ACh) deficiency, metal ion dynamic equilibrium disorder, and deposition of amyloid and tau. Increased evidence suggests neuroinflammation and oxidative stress may cause AD. However, none of these factors induces AD independently, but they are all associated with the formation of Aβ and tau proteins. Current clinical treatments based on ACh deficiency can only temporarily relieve symptoms, accompanied with many side-effects. Hence, searching for natural neuroprotective agents, which can significantly

Carboxymethyl chitosan represses tumor angiogenesis in vitro and in vivo.

Carboxymethyl chitosan (CMCS), with potent water solubility, biocompatibility, and non-toxicity, has emerged as a promising candidate for biomedical applications. In this study, the anti-tumor angiogenesis effects of CMCS were evaluated in vitro and in vivo. Our results showed that CMCS could inhibit the 2-dimensional and 3-dimensional migration of human umbilical vein endothelial cells (HUVECs) in vitro. CMCS significantly inhibited the growth of mouse hepatocarcinoma 22 tissues and could promote tumor cell necrosis as suggested by pathological observations. The CD34 expression in H22 tumor t

Protective effect of dietary chitosan on cadmium accumulation in rats.

BACKGROUND/OBJECTIVES: Cadmium is a toxic metal that is an occupational and environmental concern especially because of its human carcinogenicity; it induces serious adverse effects in various organs and tissues. Even low levels of exposure to cadmium could be harmful owing to its extremely long half-life in the body. Cadmium intoxication may be prevented by the consumption of dietary components that potentially reduce its accumulation in the body. Dietary chitosan is a polysaccharide derived from animal sources; it has been known for its ability to bind to divalent cations including cadmium,
