Mechanisms and individuality in chromium toxicity in humans.

With regards to health, chromium (Cr) is an ambiguous chemical element. Although it is considered to be an important micronutrient, it also is connected with several pathologies, including carcinogenicity. The mechanism of action of Cr and its compounds in humans is not yet fully understood. Currently, three possible mechanisms have been proposed for carcinogenesis: Cr(VI)-induced multistage carcinogenesis, genomic instability, and epigenetic modification. Therefore, in addition to the toxicity of this metal and its ions, human susceptibility to Cr-induced pathologies depends on external facto…

Structure and Reactivity of Chromium(VI) Alkylidenes.

Bis(arylimido)Cr(VI) dialkyls lacking β-hydrogen decompose by α-hydrogen abstraction and, upon trapping with triphenylphosphine, yield isolable alkylidene complexes. Two such complexes, namely (ArN)2Cr═CHR(PPh3) (R = tBu, SiMe3), have been structurally characterized. The coordinatively unsaturated alkylidene intermediates are highly reactive; they effect C-H activation of saturated hydrocarbons and they react with olefins to produce metallacyclobutanes.

Cytokine detection for the diagnosis of chromium allergy.

BACKGROUND: Patch testing remains the gold standard method for the identification of the etiologic agent of allergic contact dermatitis. However, it is a subjective, time-consuming exam whose technique demands special care and which presents some contraindications, which hamper its use. In a recent study, we showed that the proliferation assay can suitably replace patch testing for the diagnosis of chromium allergy, which had been previously demonstrated only for nickel allergy. In this study, we try to refine the method by reducing the incubation period of cultures for lymphocyte proliferatio…

Strategies for chromium bioremediation of tannery effluent.

Bioremediation offers the possibility of using living organisms (bacteria, fungi, algae,or plants), but primarily microorganisms, to degrade or remove environmental contaminants, and transform them into nontoxic or less-toxic forms. The major advantages of bioremediation over conventional physicochemical and biological treatment methods include low cost, good efficiency, minimization of chemicals, reduced quantity of secondary sludge, regeneration of cell biomass, and the possibility of recover-ing pollutant metals. Leather industries, which extensively employ chromium compounds in the tanning…

Mechanisms of bacterial resistance to chromium compounds.

Chromium is a non-essential and well-known toxic metal for microorganisms and plants. The widespread industrial use of this heavy metal has caused it to be considered as a serious environmental pollutant. Chromium exists in nature as two main species, the trivalent form, Cr(III), which is relatively innocuous, and the hexavalent form, Cr(VI), considered a more toxic species. At the intracellular level, however, Cr(III) seems to be responsible for most toxic effects of chromium. Cr(VI) is usually present as the oxyanion chromate. Inhibition of sulfate membrane transport and oxidative damage to …

The safety and efficacy of high-dose chromium.

The data on the standards for chromium requirements and the safety of various chromium compounds and doses are reviewed. The 350-fold difference between the acceptable daily intake and the calculated reference dose for humans of 70 mg per day seems without precedent with respect to other nutritional minerals. Previous claims of mutagenic effects of chromium are of questionable relevance. While studies have found DNA fragmentation (clastogenic effects) by chromium picolinate, anecdotal reports of high-dose chromium picolinate toxicity are few and ambiguous. The beneficial effects of chromium on…

Overproduction of insulin in the chromium-deficient rat.

The hypothesis that the insulin secretory hyperresponsiveness observed in rats with diet-induced insulin resistance may be a basic characteristic of dietary chromium (Cr) deficiency was evaluated. Two groups of weanling rats were fed ad libitum a purified diet containing 64% sucrose, 20% casein, 5% corn oil, and the recommended levels of vitamins and minerals without added Cr. Cr-deficient (-Cr) rats were provided with distilled drinking water only, while Cr-supplemented (+Cr) rats received water containing 5 ppm Cr as CrCl3. A third group of rats fed a commercial chow diet served as sucrose c…

Analytical methodology for biological monitoring of chromium.

Analytical aspects associated with biological and ambient monitoring are outlined with an emphasis on processing samples at very low concentrations and optimization of analytical methods. Verification of chromium determination in body fluids by means of "Round Robin" tests is discussed. In addition, an approach for indirect valency discrimination of chromium species relevant in biological monitoring is presented. Application of voltammetry for model tests is described. This is aimed at studying the reaction of hexavalent chromium in human plasma. As a result, a natural reducing capacity of pla…

Bioaccumulation and toxicology of chromium: implications for wildlife.

The major source of exposure to Cr for wild birds and mammals is through ingestion with food. Chromium(VI) compounds are absorbed significantly more efficiently (2-10% of dose) from the GI tract than inorganic Cr(III) compounds (0.5-3%), due to the increased membrane permeability of the former. Transfer of Cr(VI) into mammalian fetuses has been documented at oral doses of 500 mg Cr/L in drinking water, and injected single doses of 5 mg Cr(VI)/kg BW in dams were teratogenic. Cr concentration data for mammalian and avian wildlife species and their potential food organisms are scarce. Worldwide, …

Inhibition of DNA synthesis by chromium compounds.

Cr(VI) irreversibly inhibited DNA synthesis in cultured mouse L cells to 50% of controls at 10 microM; 3.3 mM Cr(III) did not. At 0.3 mM, Cr(III) and Cr(VI) inhibited DNA synthesis in permeabilized L cells to 50% of control values. Cr(III) was a stronger inhibitor of DNA synthesis in the DNA-Escherichia coli DNA polymerase I system than was Cr(VI). The inhibitory effect of Cr(VI) depended on the ratio of Cr/DNA and Cr/enzyme; on the other hand, the increase in the concentration of DNA polymerase did not affect the inhibition of Cr(III), Cr(III), below the inhibitory concentration, produced an

[The change of valence of chromium by acid treatment].

There are many kinds of chromium compounds in nature, and most of them exist as Cr6+- and Cr3+-compounds. Generally, these two kinds of compounds are stable in normal circumstances. When the chromium in biomaterial or in atmospheric substances is analyzed, the samples are usually ashed with strong acids at high temperatures to destroy the organic components, and chromium compounds are changed to water soluble salts in this ashing process. And then chromium is determined in various methods. The relation between the used acids and the behavior of chromium were investigated, and the following fac…

Chromium depletion in the pathogenesis of diabetes and atherosclerosis.

Dietary Cr deficiency may cause the aging-related Cr depletion observed in humans. Such depletion, when it occurs with excessive consumption of sugar and other carbohydrates, may result in glucose intolerance, glycosuria, hyperinsulinemia, and hyperlipidemia. These metabolic disorders could explain why athero-sclerotic diseases are endemic to most Western industrial societies. This review detailed significant current research and findings on the manifestations of Cr deficiency in humans and animal models.

The measurement of volatile chromium in biological materials.

Chromium is an essential trace element in mammals since dietary chromium deficiency results in glucose intolerance due to decreased sensitivity to insulin. In humans, both adults and children with glucose intolerance have been improved by treatment with chromium. Furthermore, chromium deficiency has been implicated as a causative factor in hypercholesterolemia and atherosclerosis. However, little is known of the metabolism of chromium in humans, primarily because of analytical difficulties. The biologically active form of chromium is the "glucose tolerance factor" (GTF) which is a co-ordinatio…
