Identification of Mutations in Cell-Free Circulating Tumor DNA in Adrenocortical Carcinoma: A Case Series.

CONTEXT: The disease course of adrenocortical carcinoma (ACC) patients is heterogeneous. A marker for prognosis and treatment response would facilitate choices for diagnosis and therapy. In other cancer types, circulating cell-free tumor DNA predicted tumor dynamics. CASE DESCRIPTIONS: The present pilot study included six patients. Next-generation sequencing (NGS) showed mutations in three ACC cases. From these patients, blood was drawn before (1 to 2 weeks) and after surgery and cell-free circulating DNA (cfDNA) was isolated. Tumor-specific mutations were found in the cfDNA of one of the thr

Deoxyribonuclease activity and circulating DNA concentration in blood plasma of patients with prostate tumors.

The DNase activity and circulating DNA (cirDNA) concentration in blood plasma of healthy donors, patients with chronic prostatitis, and patients with prostate tumors were analyzed. The concentration of the cirDNA from plasma was determined by PicoGreen fluorescent assay. DNase activity in blood was measured using the immunoassay based on the cleavage of a hapten-labeled 974-bp DNA substrate. The mean cirDNA concentration in the plasma of healthy donors was low (21 +/- 4 ng/mL total blood) and was accompanied by high DNase activity (0.17 +/- 0.04 U/mL blood). The mean cirDNA concentration was 9

Expert panel consensus recommendations on the use of circulating tumor DNA assays for patients with advanced solid tumors.

Comprehensive genomic profiling is increasingly used to facilitate precision oncology based on molecular stratification. In addition to conventional tissue comprehensive genomic profiling, comprehensive genomic profiling of circulating tumor DNA has become widely utilized in cancer care owing on its advantages, including less invasiveness, rapid turnaround time, and capturing heterogeneity. However, circulating tumor DNA comprehensive genomic profiling has some limitations, mainly false negatives due to low levels of plasma circulating tumor deoxyribonucleic acid and false positives caused by …

Case Report: Concordance of cerebrospinal fluid-derived circulating tumor DNA with tumor tissue in spinal glioblastoma associated with Lynch syndrome.

BACKGROUND: Spinal glioblastoma (GBM) is an exceptionally rare entity and poses significant diagnostic challenges due to a substantial risk of neurological morbidity with surgical biopsy. Tissue-based analyses are frequently limited by sampling constraints and tumor heterogeneity. Conventional diagnostic modalities, including MRI and cerebrospinal fluid (CSF) cytology, often lack specificity and sensitivity. Commercially available CSF liquid biopsy platforms offer a minimally invasive approach for detecting tumor-derived cell-free DNA (cfDNA) to enable genomic profiling. METHODS: We report a

Comparative analysis of EGFR mutations in circulating tumor DNA and primary tumor tissues from lung cancer patients using BEAMing PCR.

In this study, we measured human epidermal growth factor receptor (EGFR) mutations in both tissue and circulating tumor DNA (ctDNA) by using beads, emulsions, amplifications and magnetic polymerase chain reaction (BEAMing PCR). Noninvasive mutation detection by assessing circulating tumor DNA (ctDNA) offers many advantages over tumor biopsy. One hundred non-small cell lung cancer (NSCLC) patients were enrolled, and both preoperative plasma samples and formalin-fixed and paraffin-embedded (FFPE) samples were collected for the study. The EGFR mutation status was determined by BEAMing PCR in ctDN…

Utility of Tumor-Informed Circulating Tumor DNA for Monitoring Treatment Response in Nonmetastatic, Locally Advanced Penile and Primary Urethral Cancers.

PURPOSE: Tumor-informed circulating tumor DNA (ctDNA) testing has not been evaluated in monitoring patients with primary urethral cancer (PUC) or penile squamous cell carcinoma (pSCC). We investigated whether serial ctDNA monitoring correlated with clinical response in these rare genitourinary malignancies in the nonmetastatic, locally advanced setting. MATERIALS AND METHODS: Sixty-six ctDNA tests using a tumor-informed molecular residual disease assay from 13 patients with PUC or pSCC were reviewed. All patients were treated with curative intent and followed with standard imaging and ctDNA t…

Early tumor-informed circulating tumor DNA (ctDNA) dynamics correlate with response to first-line advanced NSCLC treated with pembrolizumab ± chemotherapy.

BACKGROUND: Immune checkpoint inhibitors (ICIs), alone or combined with chemotherapy, constitute the standard first-line treatment for advanced non-small cell lung cancer (NSCLC) without actionable oncogenic drivers. However, radiologic response assessment has limitations in capturing early biological treatment effects. Tumor-informed circulating tumor DNA (ctDNA) monitoring has emerged as a promising biomarker for early evaluation of treatment efficacy during immunotherapy. METHODS: We conducted a prospective, single-center, longitudinal observational study including patients with stage IV N…

Tetrahedral DNA nanostructures-assisted electrochemical assay for detecting circulating tumor DNA by combining a masking tactic with 3D-hybridization chain reactions.

Circulating tumor DNA (ctDNA) is a remarkable noninvasive tumor marker that plays a crucial role in tumor diagnosis, prognosis and treatment. However, detecting low-abundance ctDNA from a substantial amount of nucleic acids originating from healthy cells is challenging. Herein, we proposed a tetrahedral DNA nanostructures (TDNs)-assisted electrochemical biosensor for ctDNA detection. This biosensor combines a masking tactic with 3D-hybridization chain reactions. Masking hairpins (MHs) were initially introduced to prevent interference from wild-type (WT) DNA. Then, the initiator sequence was tr…

MRD-2 in the GHSG HD21 trial assessed by a validated circulating tumor DNA sequencing assay.

Beyond cure, major goals in patients with Hodgkin lymphoma (HL) are tailoring treatment to a patient's individual risk for relapse to reduce acute and late toxicities, identifying candidates for early incorporation of novel agents, and making treatment affordable on a global level. Minimal residual disease (MRD) assessment by circulating tumor DNA (ctDNA) sequencing emerged as a promising strategy to achieve these goals; however, previous studies differed in sampling time points, assay validation, and definitions for MRD negativity. Here, we applied LymphoVista, a validated ctDNA sequencing as…

In Vivo Spatiotemporal Protection and Recognition of Circulating Tumor DNA for Early Cancer Diagnosis and Monitoring.

Circulating tumor DNA (ctDNA) offers a promising avenue for noninvasive cancer diagnosis and prognosis, yet its effective utilization is fundamentally limited by rapid in vivo clearance and nuclease-mediated degradation, resulting in extremely low bioavailability. In addition, conventional in vitro recognition-based ctDNA detection means may potentially compromise the diagnostic accuracy. Here, we report an in vivo spatiotemporal protection and recognition strategy that actively enhances ctDNA bioavailability, enabling accurate early cancer diagnosis and quantitative monitoring of disease prog…

Structural DNA Nanotechnology-Based Branch-Sprouted Biomimetic Nanobamboo for Efficient Isolation of Rare Circulating Tumor Cells.

Because of very few sequence-identified viral scaffolds suitable for origami assembly and the unavailability of tile-based, nondeformable, superlarge DNA structures with architecturally defined collective properties, the biomedical application of structural DNA nanotechnology is hampered. In the current contribution, we demonstrate a backbone-inserted, center-rigidified, stringing-based assembly (CRSA) technology for constructing DNA biomimetic nanobamboo (Apt-BNB) of high aspect ratio, rich with membrane receptor-binding branches. The Apt-BNB structure is additionally rigidified by installing
