Nanoparticle-mediated inhibition of Yes-associated protein prevents corneal scarring after traumatic injury.

Corneal scars develop during the regenerative process following traumatic injuries. Verteporfin (VP), a Yes-associated protein (YAP) inhibitor, has shown promise as an antifibrotic agent, but its application to the ocular surface is limited by unfavorable physicochemical properties. Here, we developed the hyaluronic acid-verteporfin nanoparticles (HA-VP NPs) to increase the apparent solubility of VP and prolong residence on the eye, aiming to promote corneal regeneration and prevent scar formation following injuries. The HA-VP NPs effectively prevented not only the transformation of corneal st

Terahertz Photons Promote Corneal Injury Repair via Epithelial Proliferation, Migration, and Inflammation Reduction.

PURPOSE: Alkali burn of the cornea is a severe ophthalmic emergency that often results in permanent vision loss due to its destructive impact on corneal tissue. Current treatments face challenges in promoting rapid healing and restoring tissue integrity. METHODS: This study demonstrates a non-contact therapeutic approach using terahertz (THz) photons transmitted via optical fibers for alkali-burned corneas. The effects of THz proton treatment on human corneal epithelial cells (HCECs) were evaluated through proliferation and migration assays. RNA sequencing was used to analyze gene expression

Single-Cell Profiling Identifies Glial Transcriptional Remodeling in the Trigeminal Ganglion After Corneal Injury.

The cornea depends on trigeminal sensory innervation for sensation and epithelial homeostasis, yet cell type-specific transcriptional responses in the trigeminal ganglion following corneal injury remain incompletely defined. To address this gap, we combined bulk and single-cell RNA sequencing to profile the mouse trigeminal ganglion after a moderate corneal alkali injury associated with transient denervation and an early phase of nerve regeneration. Corneal injury elicited a robust early transcriptional response dominated by glial populations. In myelinating Schwann cells, injury responses wer

Sterile peripheral corneal infiltration mimicking infectious keratitis following chemical injury: a case report.

BACKGROUND: Sterile corneal infiltration is an inflammatory condition typically associated with contact lens wear, staphylococcal hypersensitivity, or corneal procedures. Chemical injuries usually present with diffuse epithelial defects or stromal haze, whereas the development of a discrete sterile corneal infiltrate following chemical injury appears to be uncommon. We describe an unusual case of unilateral sterile peripheral corneal infiltration following accidental chemical injury caused by contact lenses soaked in a makeup remover. CASE PRESENTATION: A 35-year-old woman presented with seve

Corneal injury associated with scrub typhus following mite exposure in a child in Taiwan: a case report.

BACKGROUND: Scrub typhus is a mite-borne zoonosis endemic to the Asia-Pacific region. Although ocular complications are occasionally reported during later stages of infection, direct mechanical ocular injury related to mite exposure has rarely been reported. Recognition of early ocular manifestations may facilitate timely diagnosis and treatment of this potentially serious infection. CASE PRESENTATION: We describe a rare case of a 9-year-old Taiwanese boy who developed acute ocular and systemic symptoms after exposure to mites during a visit to Alishan Forest, Taiwan. Live mites were identifi

Topical Hepatocyte Growth Factor Accelerates Wound Healing and Inhibits Scarring in Experimental Corneal Injury.

PURPOSE: The aim of this study was to compare the rates of healing, scar formation, and regression of preexisting scars using topical murine hepatocyte growth factor (mHGF), recombinant human deleted HGF (dHGF), murine nerve growth factor (mNGF), and phosphate-buffered saline (PBS). METHODS: Mechanical corneal epithelial and stromal injury was induced in C57BL/6 mice. Five groups of mice were studied in each of phase I (wound healing and scar formation) and phase II (regression of preexisting scars). Each group received topical 0.1% mHGF, 0.1% dHGF, 0.2% dHGF, 0.1% mNGF, or PBS. In phase I, a

Treatment of Sulfur Mustard Corneal Injury by Augmenting the DNA Damage Response (DDR): A Novel Approach.

Sulfur mustard (SM) is a highly reactive organic chemical has been used as a chemical warfare agent and terrorist threat since World War I. The cornea is highly sensitive to SM toxicity and exposure to low vapor doses can cause incapacitating acute injuries. Exposure to higher doses can elicit persistent secondary keratopathies that cause reduced quality of life and impaired or lost vision. Despite a century of research, there are no specific treatments for acute or persistent ocular SM injuries. SM cytotoxicity emerges, in part, through DNA alkylation and double-strand breaks (DSBs). Because

Topical application of calcitonin gene-related peptide as a regenerative, antifibrotic, and immunomodulatory therapy for corneal injury.

Calcitonin gene-related peptide (CGRP) is a multifunctional neuropeptide abundantly expressed by corneal nerves. Using a murine model of corneal mechanical injury, we found CGRP levels in the cornea significantly reduced after injury. Topical application of CGRP as an eye drop accelerates corneal epithelial wound closure, reduces corneal opacification, and prevents corneal edema after injury in vivo. CGRP promotes corneal epithelial cell migration, proliferation, and the secretion of laminin. It reduces TGF-β1 signaling and prevents TGF-β1-mediated stromal fibroblast activation and tissue fibr

Cell spray printing combined with Lycium barbarum glycopeptide promotes repair of corneal epithelial injury.

The corneal epithelium, located as the outermost layer of the cornea, is inherently susceptible to injuries that may lead to corneal opacities and compromise visual acuity. Rapid restoration of corneal epithelial injury is crucial for maintaining the transparency and integrity of the cornea. Cell spray treatment emerges as an innovative and effective approach in the field of regenerative medicine. In our study, a cell spray printing platform was established, and the optimal printing parameters were determined to be a printing air pressure of 5 PSI (34.47 kPa) and a liquid flow rate of 30 ml/h.

PI3K signaling and lysyl oxidase is critical to corneal stroma fibrosis following mustard gas injury.

Sulfur mustard gas (SM), an alkylating and vesicating agent, has been used frequently in many wars and conflicts. SM exposure to the eye results in several corneal abnormalities including scar/fibrosis formation. However, molecular mechanism for SM induced corneal fibrosis development is poorly understood. After SM insult to the eye, excessive synthesis/secretion of extracellular matrix components (ECM) including collagen (COL) I, COL III, and lysyl oxidase (LOX) by corneal myofibroblasts causes corneal fibrosis, however, precise mechanism remains elusive. This study tested the hypothesis that

Treatment of Non-Infectious Corneal Injury: Review of Diagnostic Agents, Therapeutic Medications, and Future Targets.

Corneal injuries can occur secondary to traumatic, chemical, inflammatory, metabolic, autoimmune, and iatrogenic causes. Ocular infection may frequently occur concurrent to corneal injury; however, antimicrobial agents are excluded from this present review. While practitioners may primarily rely on clinical examination techniques to assess these injuries, several pharmacological agents, such as fluorescein, lissamine green, and rose bengal, can be used to formulate a diagnosis and develop effective treatment strategies. Practitioners may choose from several analgesic medications to help with p…

Corneal keloid presenting forty years after penetrating injury: Case report and literature review.

A 64-year-old Caucasian man had been diagnosed with presumed ocular surface squamous neoplasia in the left eye and started on topical interferon alpha 2b drops. When we saw him, he provided a history of penetrating corneal injury more than forty years before. Slit lamp examination revealed a large, elevated, opaque lesion involving the left cornea. High-definition anterior segment optical coherence tomography revealed a hyperreflective lesion involving the anterior stroma with a relatively normal overlying epithelium. Based on the clinical history, slit lamp, and anterior segment optical coher

Case Report: Penetrating Corneal Injury Under an Intact Laser-assisted in Situ Keratomileusis Flap.

SIGNIFICANCE: As the prevalence of post-laser-assisted in situ keratomileusis (LASIK) patients rises, recognition of possible traumatic complications and their management in this population becomes increasingly relevant. Penetrating injuries and intraocular foreign bodies are possible in post-LASIK corneas despite an intact anterior corneal surface. However, with prompt recognition and treatment, excellent visual recovery can be achieved. PURPOSE: The purpose of this study was to report an interesting case of a penetrating foreign body through an intact LASIK flap. CASE REPORT: A 48-year-old

Injury and defective regeneration of the epithelial basement membrane in corneal fibrosis: A paradigm for fibrosis in other organs?

Myofibroblast-mediated fibrosis is important in the pathophysiology of diseases in most organs. The cornea, the transparent anterior wall of the eye that functions to focus light on the retina, is commonly affected by fibrosis and provides an optimal model due to its simplicity and accessibility. Severe injuries to the cornea, including infection, surgery, and trauma, may trigger the development of myofibroblasts and fibrosis in the normally transparent connective tissue stroma. Ultrastructural studies have demonstrated that defective epithelial basement membrane (EBM) regeneration after injur

Therapeutic potential of Jatimukuladi Varti Anjana in alkali burn-induced corneal injury and neovascularization in albino rats.

INTRODUCTION: Corneal transparency is maintained by its avascular nature, and disruption following injury can lead to corneal neovascularization (CNV), resulting in visual impairment. Although current treatment such as corticosteroids, NSAIDs, anti-VEGF agents, and surgical interventions are available, their use is often limited by adverse effects, recurrence, and cost. Jatimukuladdiv arti Anjana (JMV), a classical ayurvedic ophthalmic formulation with reported anti-inflammatory and tissue-healing properties, was evaluated for its therapeutic efficacy in an alkali burn-induced CNV model in alb

Toll-like receptor 2 deficiency exacerbates corneal angiogenesis in injury by impairing regulatory T cells.

Background: Toll-like receptor (TLR) 2 is a primary sensor of injury, and regulatory T cells (Tregs) are crucial mediators of tissue homeostasis. In this study, we aimed to investigate whether TLR2 is necessary for Treg-mediated restoration of corneal homeostasis following injury. Methods: We evaluated inflammatory corneal neovascularization and the proportions of Tregs, along with pro-angiogenic, pro-inflammatory monocytes, using a suture-induced corneal angiogenesis model in mice that either lacked TLR2 or were subjected to temporary TLR2 inhibition. The roles of injury-induced Tregs in corn
