UVB in solar-simulated light causes formation of BaP-photoproducts capable of generating phosphorylated histone H2AX.

Polycyclic aromatic hydrocarbons (PAHs), wide-spread mutagenic and carcinogenic environmental pollutants, are consistently exposed to sunlight in the environment. Our previous paper showed that benzo[a]pyrene (BaP) exposed to solar-simulated light (SSL) induced phosphorylation of histone H2AX (γ-H2AX) [T. Toyooka, G. Ohnuki, Y. Ibuki, Solar-simulated light-exposed benzo[a]pyrene induces phosphorylation of histone H2AX, Mutat. Res. 650 (2008) 132-139], a marker of DNA double strand breaks. In this study, we found the ultraviolet B (UVB) region of SSL to produce photomodified BaP with high cytot

Imaging and force spectroscopy on desmoglein 1 using atomic force microscopy reveal multivalent Ca(2+)-dependent, low-affinity trans-interaction.

Desmoglein 1 is a desmosomal member of the cadherin family expressed in stratified epithelia. Desmoglein 1 is the target adhesion molecule of severe blistering skin diseases such as pemphigus or bullous impetigo. However, despite this enormous pathological relevance, the molecular binding properties of desmoglein 1 are largely unknown. Using atomic force microscopic imaging, we found that desmoglein 1 molecules displayed Ca(2+)-dependent conformational changes of the extracellular domains. By single-molecule force-distance cycles, we provide evidence that desmoglein 1 undergoes Ca(2+)-dependen

Recovery of Chinese hamster cells following photosensitization by zinc tetrahydroxyphthalocyanine.

The phthalocyanine dyes are attractive sensitizers for photodynamic therapy of cancer. The light fluence response curves for photocytotoxicity of zinc tetrahydroxyphthalocyanine were constructed using the colony-forming ability of Chinese hamster cells as an end-point. The survival curve of cells photosensitized to white light by this dye has a pronounced shoulder followed by an exponential decline. Postillumination hypertonic treatment (0.5 M NaCl for 20 min at 37 degrees C) enhanced log-phase killing, although to a lesser extent than after exposure to ionizing radiation. While such an enhanc

Elevation of tubulin levels by microinjection suppresses new tubulin synthesis.

Most eukaryotic cells rapidly and specifically depress synthesis of alpha- and beta-tubulin polypeptides in response to microtubule inhibitors which cause microtubule depolymerization and presumably increase the intracellular concentration of free subunits. Other drugs which interfere with microtubule function but which lead to a decrease in the subunit pool size have little effect on the rate of new tubulin synthesis. These findings have previously been interpreted to indicate that cultured cells synthesize tubulin constitutively unless the subunit pool rises above a specified level. At this

Cloning of the protective antigen gene of Bacillus anthracis.

The tripartite protein toxin of Bacillus anthracis consists of protective antigen (PA), edema factor (EF), and lethal factor (LF). As a first step in developing a more efficacious anthrax vaccine, recombinant plasmids containing the PA gene have been isolated. A library was constructed in the E. coli vector pBR322 from Bam HI-generated fragments of the anthrax plasmid, pBA1. Two clones producing PA were identified by screening lysates with ELISA (enzyme-linked immunosorbent assay). Western blots revealed a full-size PA protein in the recombinant E. coli, and a cell elongation assay demonstrate

The effect of caffeine on the cytotoxicity of misonidazole and some other nitroheterocyclic compounds.

Caffeine was found to potentiate the cytotoxic effect of misonidazole (1-(2-nitroimidazol-1-yl)-3-methoxy-2-propanol) towards mammalian cells in vitro. This enhancement of toxicity is shown to occur under both aerobic and hypoxic conditions. Split dose experiments indicate that the general shape of the hypoxic survival curve can be restored by exposure of cells of O2 between doses. Exposure of cells to 2mM caffeine during the split dose experiment has no potentiating effect. Further experiments showed that caffeine affects the expression of misonidazole-induced potentially lethal damage. Other

Analysis of glycosaminoglycan from diabetic and normal Chinese hamster cells.

Diabetic and normal cell lines from Chinese hamster kidneys were cultured in media containing 35SO4 and 3H-glucosamine. Glycosaminoglycans (GAG) were extracted and analyzed from the media, trypsin, and cell pellet by enzymatic and electrophoretic procedures. Significant increases in the hyaluronic acid content were noted in all three fractions of diabetic GAGs when compared with normals. In addition, an increased heparan sulfate content and decreased chondroitin sulfate amounts were noted in diabetic cell lines. These data suggest that in vivo changes in GAG types and amounts in diabetic kidne

Endogenous eukaryotic CHO-K1 cell promoters as tools to improve the production of a difficult-to-express recombinant protein.

Recombinant proteins with applications in human and veterinary medicine are mainly produced in mammalian expression systems, particularly Chinese hamster ovary (CHO) cells. Although these cells enable proper folding and post-translational modifications, they often exhibit low specific productivity, which represents a critical limitation in large-scale biopharmaceutical manufacturing. Strong viral promoters, including CMV and its enhancer-enriched variant CMV+E, are commonly used to drive transgene expression. However, they are susceptible to epigenetic silencing and progressive loss of activit

Harnessing design of experiments and automation significantly improves transient protein production in Chinese hamster ovary cells and accelerates drug discovery.

Chinese Hamster Ovary (CHO) cells produce the majority of approved therapeutic proteins and are the de facto platform for rapid, milligram-scale production via transient expression during early biologics discovery. However, performance is highly sensitive to transfection composition, feed strategy, and execution details; and vendor protocols often underperform without platform-specific optimization. We integrated Design of Experiments (DOE) with liquid-handling automation to develop a rapid, data-driven optimization workflow for CHO transient expression. Key factors and ranges were defined for

Gene overexpression reduces inhibitory metabolites to enhance CHO cell growth and IgG1 production.

Controlling the generation of toxic by-products in mammalian bioprocess to maximize therapeutic protein production and glycosylation patterns is a challenge. Intracellular metabolism is often not well-regulated and known to secrete toxic intermediate by-products which hampers cellular performance and negatively impacts critical quality attributes (CQA) of cells. Previous studies have identified trigonelline (TRI), n-acetyl putrescine (NAP), aconitic acid (AA), and cytidine monophosphate (CMP) generated through CHO cell metabolism and verified their negative impacts on growth and antibody produ

Constitutive activation of MC1R in the large-billed crow (Corvus macrorhynchos) and its potential role in black plumage.

Melanin-based plumage coloration in birds is largely regulated by the melanocortin 1 receptor (MC1R), a G protein-coupled receptor that promotes eumelanin synthesis via cAMP signaling. In domestic chickens, constitutively activating mutations such as the MC1R^E (E92K) allele cause melanistic phenotypes, demonstrating that persistent MC1R activation can drive generalized darkening. However, to our knowledge, no experimental study has directly demonstrated constitutive MC1R activation in wild birds exhibiting uniformly black plumage. We investigated the sequence and signaling properties of MC1R

13C metabolic flux analysis and gene expression analysis of perfusion culture reveal metabolic bottlenecks for stable glycosylation profiles of biologics.

Perfusion Chinese hamster ovary (CHO) cell culture offers potential benefits over fed-batch cultures, including higher productivity and a reduced production footprint. However, maintaining a stable profile of product quality attributes (PQAs) over the entirety of a perfusion culture has proven to be a challenge across the biopharmaceutical industry, which is critical for ensuring lot-to-lot consistency. A deeper understanding of the underlying molecular mechanisms of PQA variability and productivity can allow for more targeted interventions. In this study, 13C metabolic flux analysis (13C MFA)

Recent Advancements in Proteomic Sample Preparation from Recombinant Chinese Hamster Ovary Cells.

Chinese hamster ovary (CHO) cells are the most commonly used mammalian host cell line for biopharmaceutical production because of their ability to correctly fold and post-translationally modify recombinant proteins that are compatible with human use. Proteomics, along with other "omic platforms," are being used to understand the biology of CHO cells with the ultimate aim to enhance CHO cell factories for more efficient production of biopharmaceuticals. Liquid chromatography-mass spectrometry (LC-MS) for proteomic analysis has become the standard technique for profiling proteomes. There are thr

Barriers Composed of tRNA Genes Can Complement the Benefits of a Ubiquitous Chromatin Opening Element to Enhance Transgene Expression.

Random integration of transgenes into host cell genomes often occurs in epigenetically unstable regions, leading to variable and unreliable transgene expression. To address this, biomanufacturing organizations frequently employ barrier elements, such as the widely-used ubiquitous chromatin opening element (UCOE). We have compared UCOE barrier activity against a barrier provided by tRNA genes. We demonstrate that the tRNA genes provide a more effective barrier than a UCOE in preventing transgene silencing in Chinese hamster ovary (CHO) cells. Nevertheless, the UCOE offers other benefits, increa

Next-Generation Sequencing-Based In Silico Transgene Integration Profiling Tool Accelerates Cell Line Genetic Characterizations.

Genetic characterization including Southern blot analysis of recombinant Chinese hamster ovary (CHO) cell lines expressing biologics is an essential prerequisite for regulatory submissions. The transposon-based expression system for CHO cell lines enables the insertion of DNA sequences into the genome and has been widely used by the biopharmaceutical industry. The system yields clones that are both highly productive and genetically stable. However, it often incorporates target genes at multiple locations in the CHO genome, presenting challenges to profile the integration sites using Southern b

Elevated Mammalian Cell Cytotoxicity of Chlorinated Drinking Waters in the Presence of CuO.

It has been demonstrated previously that cupric oxide (CuO) can enhance the formation of one- to two-carbon-atom disinfection byproducts (DBPs) during water chlorination. Effects of CuO on the Chinese hamster ovary cell cytotoxicity of chlorinated water and the underlying mechanisms were investigated in this study. Results showed that 0.1 g L-1 CuO elevated the cytotoxicity of chlorinated waters by 20% and 120% at initial bromide concentrations of 15 and 415 μg L-1, respectively. Increasing the CuO dose increased the cytotoxicity induced by both known and unknown DBP fractions, and a more pron

Production of multi-subunit proteins in CHO cells by transposase-mediated integration of subunit-splitting vectors.

The Chinese hamster ovary (CHO) cell line is a widely employed system for producing therapeutic proteins. In our previous study, we established an efficient method for generating stable cell lines by utilizing the Tol2 transposon system, combined with cycloheximide (CHX) resistance as a selection marker. This DNA-based transposon allows the integration of the gene of interest into various genomic loci within the host cell genome. To further develop this system, we performed gene transfer using single vectors carrying each subunit separately, rather than using dual vector linking subunits in ta

Glycoprotein Preparation by Heterologous Expression.

The eukaryotic expression, typically using cultured mammalian cells, is used for stable isotope labeling of glycoproteins. More recently, innovations have been made to reduce the cost of cell culture, control glycoforms, and employ more convenient expression systems other than mammalian cells. Here we describe the procedures involved in expressing isotope-labeled immunoglobulin G by Chinese hamster ovary cells and silkworms.

Evaluation of "Difficult-to-Express" Monoclonal Antibodies in a CHO-Based Hybrid Site-Specific Integration System Under Industrially Relevant Conditions.

Variation in the primary sequence of monoclonal antibodies (mAbs) can negatively affect their behavior in biopharmaceutical manufacturing platforms, and efforts to identify mAbs with poor "developability" characteristics lack robust methods for assessing mAb expression from an industrially relevant platform. Recent advancements in site-specific integration-based (SSI) platforms in Chinese hamster ovary (CHO) cells can mitigate the high transcriptional variation observed with random integration and the low industrial relevance of transient expression by providing a flexible platform for mAb exp

Utilizing Cole-Cole parameters for in-line feedback: Cell culture process adjustments based on cell health.

In the biopharmaceutical industry, effective process control strategies are essential for enhancing drug substance quality and yield. This study presents dielectric spectroscopy as a novel approach for in-line monitoring of cell apoptosis, enabling earlier detection of apoptotic events and related cellular changes. Utilizing permittivity measurements, we assessed Cole-Cole parameters, specifically critical frequency (fc) and delta epsilon (Δε), as key performance indicators (KPIs) for real-time monitoring of cell health in CHO cell cultures during batch and perfusion processes.
