Thymol and carvacrol from the class of monoterpene phenols are one of the most potent plant essential oil components possessing antimicrobial effects. Known for their wide bioactive spectrum, these positional isomers of isopropyl cresol deplete ergosterol content, compromise membrane permeability, block efflux pumps and restore antifungal susceptibility to fluconazole in resistant Candida strains. Exposure to these natural compounds induces a cascade of stress responses, which are important to comprehend their microbicidal mechanisms. This study evaluates the antioxidant defense response to lo

To map out the mobile-phase parameter space for the separation of the positional isomers carvacrol and thymol, naturally occurring monoterpenoid phenols of astonishingly wide biological activity, by reversed-phase liquid chromatography, we tested conventional and green candidates for the organic solvent (OS) in the mobile phase. Binary and ternary compositions were evaluated at different solvent ratios regarding resolution, selectivity, and analysis time of the isomer separation on a C18 column. Baseline separation with binary compositions was obtained with ethanol (EtOH), acetonitrile (ACN),

Multilayer systems are an innovative strategy to improve biodegradable polymers and incorporate active compounds. In the current work, taro starch-cellulose nanofiber (CNF) three-layer films, consisting of two outer starch-CNF layers and an inner starch layer containing 0, 5, or 10% carvacrol, were developed as biodegradable active separators for Paipa cheese slices. Carvacrol incorporation promoted pores, cracks, and microstructural heterogeneity. Besides, carvacrol increased surface wettability, reducing the water contact angle from 45.44° to 34.97°, but did not significantly affect water va

A total of 12 new β-halo alcohols and 12 new β-halo thiol derivatives were synthesized. Natural alcohol compounds with known pharmacological properties were selected as starting substrates, aiming to synthesize compounds that have the potential to exhibit biological activity. The synthesis of β-halo alcohol derivatives involved a two-step process, while β-halo thiol derivatives were carried out in three steps. Effective and inexpensive methods were used for all transformations. Yields for β-halo alcohol derivatives ranged from 79% to 82%, and for β-halo thiol derivatives from 66% to 71%. Their

Candida albicans typically thrives in a commensal relationship with humans but is also an opportunistic fungal pathogen. As an opportunistic pathogen, C. albicans relies heavily on its ability to assimilate nutrients, for which it must compete with the host and other microorganisms. Amino acid biosynthesis, sensing, and uptake play pivotal roles in C. albicans growth and pathogenicity. C. albicans biosynthesizes aromatic amino acids and co-enzyme Q de novo through the shikimate pathway, including the Aro1, Aro2, and Aro7 enzymes, but also has amino acid transporters for uptake from the environ

Streptococcus pyogenes is a leading cause of chronic and acute infections, including streptococcus pharyngitis. Biofilm formation by S. pyogenes can cause tolerance against antibiotics. Although penicillin remains the first choice of antibiotic for S. pyogenes, alternative approaches have gained interest due to treatment failures and hypersensitive individuals. Carvacrol is a monoterpenoid from herbal plants with selective biocidal activity on S. pyogenes. Therefore, the present study reveals the efficacy of carvacrol in inhibiting and eradicating S. pyogenes biofilm. The antibiofilm activitie

Australian tea tree (Melaleuca alternifolia) oil (TTO) and its monoterpene constituents such as terpinen-4-ol (T4O), 1,8-cineole, limonene, p-cymene, and α-terpinene have been shown to be effective in controlling a wide range of parasitic infections. The anti-parasitic effects of these compounds are mainly due to their anti-histamine and anti-acetylcholinesterase activities as well as their ability to modulate host inflammatory responses. This review attempts to summarize recent advances in the uses of TTO and its 15 major monoterpene constituents in treating parasitic infections in both human
