Potential Role of Imaging in the Evaluation of Adiposity and Approval of Anti-Obesity Drugs.

The global increase in obesity highlights the need for accurate tools to assess body composition and monitor treatment efficacy. Traditional metrics, including body mass index and waist circumference, offer limited precision for fat quantification. Imaging-based techniques capable of visualizing internal structures are increasingly being recognized for their ability to provide comprehensive fat assessment. This review outlines the principles, strengths, and limitations of key modalities, including dual-energy X-ray absorptiometry (DXA), CT, MRI, and bioelectrical impedance analysis (BIA). DXA

FDA regulatory experience with drug manufacturing process models in approved applications.

Drug manufacturing process models have been successfully applied to process development and control in regulatory submissions in at least 15 FDA-approved applications from 8 companies between 2012-2025. These submissions span drug substance and drug product manufacturing in approved NDAs, BLAs (including biosimilars), originals, and supplements. Eleven model-supported applications include continuous manufacturing, which requires an understanding of material residence time distribution (RTD) through a process. Models in approved submissions have been applied to design space establishment and re

New drugs and their performance 10 years after approval: a systematic analysis.

More than 10,000 drugs are available on today's market in Germany, with an increasing number receiving institutional approval every year. However, their general efficacy and utility for patients are often not thoroughly analyzed and, in some cases, remain questionable. The lack of systematic analysis for these parameters hinders learning from failures and successes. As a result, there is a risk of wasting resources by the pharmaceutical industry and approving new drugs that offer no additional benefit to patients in need of innovative treatments. Therefore, we set out to analyze the evolution

Do GPCRs constitute the target of 30% of newly approved drugs in Germany?

G protein-coupled receptors (GPCRs) are often stated to constitute the target of 30% (or even more) of all approved drugs. This number, probably first mentioned in ( https://pubmed.ncbi.nlm.nih.gov/12209152/ ) and later has been often cited by many other studies and in some studies even taken for granted, but is this true? In this study, the number of approved drugs acting on GPCRs was analyzed in relation to the total number of approved drugs in Germany. Newly approved drugs in the years 1987-2023 were analyzed. Of the newly approved drugs in Germany in this period, 209 out of 1110 drugs (19%

Global first-in-class drugs approved in 2023-2024: Breakthroughs and insights.

First-in-class (FIC) drugs are considered the main drivers of new drug discovery and have novel targets and mechanisms. The proportion of FIC drugs among new drugs provides fundamental guidance for both academic and pharmaceutical research. From a global perspective, 81 FIC drugs were approved in 2023 and 2024. Among them, small-molecule drugs account for a greater percentage (51.9%), illustrating the discovery of new chemical entities. Macromolecule drugs (48.1%), which mainly consist of antibody analogs, represent a growing trend as new biotechnology techniques have emerged. In terms of FIC

Unveiling FDA-approved Drugs and Formulations in the Management of Bladder Cancer: A Review.

Urological cancers are one of the most prevalent malignancies around the globe. Specifically, bladder cancer severely threatens the health of humans because of its heterogeneous and aggressive nature. Extensive studies have been conducted for many years in order to address the limitations associated with the treatment of solid tumors with selective substances. This article aims to provide a summary of the therapeutic drugs that have received FDA approval or are presently in the testing phase for use in the prevention or treatment of bladder cancer. In this review, FDA-approved drugs for bladde

Repurposing of propafenone, an FDA approved anti-arrhythmic drug for antileishmanial therapy.

Elimination of the Neglected Tropical Disease Visceral Leishmaniasis (VL) is a Sustainable Development Goal. It is caused by Leishmania donovani. The therapeutic options for the treatment of VL are limited due to problems such as drug resistance and toxicities. Drug repurposing can be a promising alternative in this case. In this work, the antileishmanial potential of FDA approved anti-arrhythmic drug propafenone has been evaluated for repurposing. It reduced the viability of L. donovani promastigotes and intracellular amastigotes with an IC50 of 8.25 ± 2.48 μM and 11.19 ± 0.01 μM respectively

Safety considerations for drugs newly approved for treating acute myeloid leukemia.

INTRODUCTION: Acute myeloid leukemia (AML) is typically characterized by a poor prognosis, mainly due to the median age at diagnosis. Until recently, treatment options were limited to intensive chemotherapy (IC) for young patients or hypomethylating agents for those ineligible for IC. Since 2017, nine molecules were registered for newly-diagnosed AML: midostaurin, gilteritinib, quizartinib, enasidenib, ivosidenib, gemtuzumab ozogamicin, CPX-351, glasdegib, and venetoclax. AREAS COVERED: The review examines the safety profile of these drugs and their interactions with other agents used in supp

Clinical Benefit and Regulatory Outcomes of Cancer Drugs Receiving Accelerated Approval.

ImportanceThe US Food and Drug Administration's (FDA) accelerated approval pathway allows approval of investigational drugs treating unmet medical needs based on changes to surrogate measures considered "reasonably likely" to predict clinical benefit. Postapproval clinical trials are then required to confirm whether these drugs offer clinical benefit.ObjectiveTo determine whether cancer drugs granted accelerated approval ultimately demonstrate clinical benefit and to evaluate the basis of conversion to regular approval.Design, setting, and participantsIn this cohort study, publicly available F

Characteristics and outcomes of new molecular oncology drug approvals, in combination or monotherapy.

IMPORTANCE: Understanding the factors that are associated with new molecular entity (NME) cancer drug approvals as a single agent and in combination, and European Society for Medical Oncology (ESMO) scores, can aid in identifying suitable factors to consider in trial designs for future drugs. In addition, the association between the various outcomes can aid in determining benefit when surrogate outcomes are used in approval consideration. OBJECTIVE: This study aims to (1) use the measures used in evaluating clinical trials by ESMO scores to determine the differences in the characteristics of

Ebanga™: The most recent FDA-approved drug for treating Ebola.

Ebolavirus (EBOV) is a virulent pathogen that causes Ebola virus disease (EVD), which is a life-threatening human condition with a fatality rate of up to 90%. Since the first outbreak in Africa in 1976, several outbreaks and epidemics of EBOV have occurred across the globe. While EVD is recognized as a serious threat to human health and outbreaks occur almost every year, the treatment options for the disease are limited. In designing therapeutic strategies against EBOV infection, viral structural proteins, such as glycoprotein (GP), could be an excellent target for neutralizing the virus. Acco

Lethal Mutagenesis of RNA Viruses and Approved Drugs with Antiviral Mutagenic Activity.

In RNA viruses, a small increase in their mutation rates can be sufficient to exceed their threshold of viability. Lethal mutagenesis is a therapeutic strategy based on the use of mutagens, driving viral populations to extinction. Extinction catastrophe can be experimentally induced by promutagenic nucleosides in cell culture models. The loss of HIV infectivity has been observed after passage in 5-hydroxydeoxycytidine or 5,6-dihydro-5-aza-2'-deoxycytidine while producing a two-fold increase in the viral mutation frequency. Among approved nucleoside analogs, experiments with polioviruses and ot

Screening of an FDA-Approved Library for Novel Drugs against Y. pestis.

Yersinia pestis is a Gram-negative pathogen that causes plague, a devastating disease that kills millions worldwide. Although plague is efficiently treatable by recommended antibiotics, the time of antibiotic therapy initiation is critical, as high mortality rates have been observed if treatment is delayed for longer than 24 h after symptom onset. To overcome the emergence of antibiotic resistant strains, we attempted a systematic screening of Food and Drug Administration (FDA)-approved drugs to identify alternative compounds which may possess antibacterial activity against Y. pestis. Here, we

In vivo High-Content Screening in Zebrafish for Developmental Nephrotoxicity of Approved Drugs.

Despite widespread drug exposure, for example during gestation or in prematurely born children, organ-specific developmental toxicity of most drugs is poorly understood. Developmental and functional abnormalities are a major cause of kidney diseases during childhood; however, the potential causal relationship to exposure with nephrotoxic drugs during nephrogenesis is widely unknown. To identify developmental nephrotoxic drugs in a large scale, we established and performed an automated high-content screen to score for phenotypic renal alterations in the Tg(wt1b:EGFP) zebrafish line. During earl
