A Dipteran's Novel Sucker Punch: Evolution of Arthropod Atypical Venom with a Neurotoxic Component in Robber Flies (Asilidae, Diptera).

Predatory robber flies (Diptera, Asilidae) have been suspected to be venomous due to their ability to overpower well-defended prey. However, details of their venom composition and toxin arsenal remained unknown. Here, we provide a detailed characterization of the venom system of robber flies through the application of comparative transcriptomics, proteomics and functional morphology. Our results reveal asilid venoms to be dominated by peptides and non-enzymatic proteins, and that the majority of components in the crude venom is represented by just ten toxin families, which we have named Asilid

High-throughput multiplexed xMAP Luminex array panel for detection of twenty two medically important mosquito-borne arboviruses based on innovations in synthetic biology.

Mosquito-borne arboviruses are emerging world-wide as important human and animal pathogens. This makes assays for their accurate and rapid identification essential for public health, epidemiological, ecological studies. Over the past decade, many mono- and multiplexed assays targeting arboviruses nucleic acids have been reported. None has become established for the routine identification of multiple viruses in a "single tube" setting. With increasing multiplexing, the detection of viral RNAs is complicated by noise, false positives and negatives. In this study, an assay was developed that avoi

Spatial and temporal variation of malaria entomological parameters at the onset of a hydro-agricultural development in central Côte d'Ivoire.

BACKGROUND: A deeper understanding of the ecology and small-scale heterogeneity of malaria transmission is essential for the design of effective prevention, control and elimination interventions. The spatial and temporal distribution of malaria vectors was investigated in five villages in close proximity to a hydro-agricultural system in Côte d'Ivoire over the course of construction and the early phase of irrigated rice farming. METHODS: The study was carried out in five villages (Raffierkro, N'Douakro, Ahougui, Kpokahankro, Koffikro) near Bouaké, central Côte d'Ivoire, between early 2007 and

Characterising subsea gas pipeline contaminants to inform decommissioning practices, an ecosystem perspective.

Operation of offshore oil and gas infrastructure may result in contaminant accumulation on the internal surfaces of infrastructure such as subsea gas pipelines. Contamination varies due to factors including the type of substance being transported, internal surface coating, and the type of maintenance cleaning. Successful decommissioning of subsea pipelines requires an ecological risk assessment of contaminant residues, especially when in situ decommissioning is considered. This study characterised the inorganic and radiological constituents of pigging residues collected from a subsea gas pipel

Effects of Long-Term Ocean Acidification Exposure on the Structural, Mineralogical, and Mechanical Properties of Sea Urchin (Echinometra spp.) Skeletons at a Natural Volcanic CO2 Seep.

Two decades of mesocosm studies document generally negative effects of ocean acidification (OA) on adult sea urchin growth, feeding performance, skeletal structure, and strength. Whether experimental observations hold true in natural systems will determine whether they can be extrapolated to predict responses under ecologically relevant contexts. Here, we employ a suite of imaging, chemical, and mechanical techniques to examine the skeletal properties of two closely related sea urchin species (genus Echinometra) living at a natural carbon dioxide (CO2) seep in Japan. Test plates and spines fro

Metacollinia emscheri n. sp., a novel sanguicolous apostome ciliate of freshwater amphipods (Gammarus spp.).

We describe a novel sanguicolous parasitic ciliate, Metacollinia emscheri n. sp., found in the freshwater amphipods Gammarus pulex and G. fossarum. This ciliate infected 8.05 % of the amphipods collected in a German stream catchment, the Boye, a tributary of the river Emscher. The ciliate showed morphological characteristics fitting the genus Metacollinia. Different life stages of variable size occurred simultaneously in the hemocoel throughout the hosts' body. The tomont had 40-47 slightly spiraled kineties, a non-ciliated cortical band, a large macronucleus, and contractile vacuoles arranged

Sensory specializations drive octopus and squid behaviour.

The evolution of new traits enables expansion into new ecological and behavioural niches. Nonetheless, demonstrated connections between divergence in protein structure, function and lineage-specific behaviours remain rare. Here we show that both octopus and squid use cephalopod-specific chemotactile receptors (CRs) to sense their respective marine environments, but structural adaptations in these receptors support the sensation of specific molecules suited to distinct physiological roles. We find that squid express ancient CRs that more closely resemble related nicotinic acetylcholine receptor

When Small Meets Smaller: Immune Modulation and Virulence Strategies in Insect-Bacteria Interactions.

Insects represent powerful experimental systems for investigating host-microorganism interactions, providing valuable insights into bacterial pathogenicity, immune regulation, symbiosis, and antimicrobial discovery. This review examines the complex relationships between insects and bacteria, focusing on the mechanisms that control infection, immune activation, and microbial adaptation. Particular attention is given to the routes of pathogen entry and to the conserved innate immune pathways that coordinate host defenses, including the Toll, Imd, Duox, and Jak/Stat signaling cascades. The review

Faecalibacterium harmsenii sp. nov., an abundant but previously overlooked Faecalibacterium in the human gut.

Faecalibacterium is one of the most abundant anaerobes in the human colon. At the genus level, this bacterium shows a strong positive association with human health. Expanding collections of isolates and metagenome-assembled genomes have revealed its species diversity, yet species-level functions remain so far underexplored. Here, we describe a novel species, Faecalibacterium harmsenii. In addition, we reclassify another isolate as a member of the recently reported Faecalibacterium langellae species. Despite close genomic relatedness, these isolates exhibit distinct physiological and biochemica

Revisiting the Status of Yellow Fever Epizootics and Its Surveillance in South America: New Non-Human Primates, Spillover and Ecological Drivers.

Yellow fever (YF) remains a re-emerging vector-borne zoonotic disease in tropical regions of the Americas despite the availability of an effective vaccine. In South America, the virus is maintained through a jungle transmission cycle involving Haemagogus and Sabethes mosquitoes and non-human primates (NHPs), which act as amplifying hosts and key epidemiological sentinels. This narrative review examines the current status of YF epizootics in South America, with a focus on the role of NHPs in viral circulation, early detection, and spillover risk to human populations. We synthesize recent eviden

The Impact of Lateral Gene Transfer in Chlamydia.

Lateral gene transfer (LGT) facilitates many processes in bacterial ecology and pathogenesis, especially regarding pathogen evolution and the spread of antibiotic resistance across species. The obligate intracellular chlamydiae, which cause a range of diseases in humans and animals, were historically thought to be highly deficient in this process. However, research over the past few decades has demonstrated that this was not the case. The first reports of homologous recombination in the Chlamydiaceae family were published in the early 1990s. Later, the advent of whole-genome sequencing uncover

Spatiotemporal distribution of insecticide resistance in Anopheles culicifacies and Anopheles subpictus in Sri Lanka.

The malaria situation in Sri Lanka worsened during the 1990s with the emergence and spread of resistance to the drugs and insecticides used for control. Chloroquine resistance has increased rapidly over this period, but adverse changes in malaria transmission are more closely associated with insecticide use rather than drug resistance. Insecticide susceptibility tests were routinely carried out in key anopheline vectors across the country for more than a decade. These sentinel data were combined with data collected by other research programmes and used to map the spatial and temporal trends of

Combined effect of copper, cadmium, and lead upon Cucumis sativus growth and bioaccumulation.

Cucumis sativus (cucumber) was tested to assess an ecotoxicity in soils contaminated by the heavy metals copper (Cu), cadmium (Cd), and lead (Pb) separately and in combinations. The toxicity endpoint was plant growth, which was measured as shoot and root lengths after 5 day exposure. Sum of toxic unit (TU) at 50% inhibition for the mixture (EC50mix) was calculated from the dose (TU-based)-response relationships by the Trimmed Spearman-Karber method. Binary metal combinations of Cu+Cd, Cu+Pb, and Cd+Pb produced all three types of interactions; concentration additive (EC50mix=1TU), synergistic (

To the problem of secondary mycotic infections and their etiological agents.

Modern therapeutic procedures that are based on advanced chemotherapy and modern medical technique have led to a prolongation of chronic patient's survival, but have also created conditions for the development of till now little known infectious complications. Among these are, no doubt, mycotic diseases caused by opportunistic microorganisms. Primarily apathogenic fungi, living as a rule in the external environment as saprophytes or colonizing as harmless epiphytes the integumental localizations of healthy humans, start to behave in the subjects with reduced resistance as invasive pathogens. Molecular Detection and Genetic Characterization of Colpodella spp. DNA in Human Fecal and Tick Samples from Hebei Province, China.

INTRODUCTION: Colpodella spp., a group of predatory protists closely related to the phylum Apicomplexa, are primarily known as free-living organisms associated with aquatic and terrestrial ecosystems. Although Colpodella-related DNA has increasingly been detected in vertebrate-associated samples, its distribution and ecological significance remain poorly understood. This study aimed to investigate the occurrence, molecular characteristics, and genetic diversity of Colpodella spp. DNA detected in human fecal samples and Haemaphysalis longicornis ticks from Hebei Province, China. Managed-bee supplementation may drive parasite-mediated exclusion of wild bees.

Sustaining pollination while limiting parasite spillover from managed bees requires a quantitative understanding of how supplementation of managed colonies, floral resource competition between managed and wild bees, and cross-host transmission jointly shape community dynamics. We develop and analyse a three-dimensional ODE model for wild bees, managed bees and a brood-targeting parasite, incorporating managed-colony supplementation, asymmetric floral competition acting on wild bees, and brood-mediated transmission with host-specific efficiencies. Using two subsystems, we derive explicit ecolog

Revealing the complexity of vampire bat rabies "spillover transmission".

BACKGROUND: The term virus 'spillover' embodies a highly complex phenomenon and is often used to refer to viral transmission from a primary reservoir host to a new, naïve yet susceptible and permissive host species. Spillover transmission can result in a virus becoming pathogenic, causing disease and death to the new host if successful infection and transmission takes place. MAIN TEXT: The scientific literature across diverse disciplines has used the terms virus spillover, spillover transmission, cross-species transmission, and host shift almost indistinctly to imply the complex process of es

Nitrogen-phosphorus stoichiometry and Cladophora growth affected by grass-sourced dissolved organic matter in the littoral zone of the Qinghai Lake, China.

Climate-driven lake expansion on the Qinghai-Tibet Plateau has triggered the extensive release of organic compounds through inundation-induced vegetation decomposition. This process generates substantial dissolved organic matter (DOM), potentially threatening alpine aquatic systems. In this work, the effects of grass-sourced DOM inputs on the nitrogen-to-phosphorus (N/P) ratio and Cladophora growth were studied. Field observation conducted in the lakeshore zone of Qinghai Lake showed that water column N/P ratios were closely related to Cladophora chlorophyll a (Chl.a) content. Microcosm experi

Multisystem Detection and Molecular Evidence Linking Dog-Associated Ticks to Two Fatal Human SFTSV Infections in Thailand.

Severe fever with thrombocytopenia syndrome virus (SFTSV) is an emerging tick-borne pathogen responsible for severe and often fatal disease in East Asia; however, information from Southeast Asia remains limited. We report two fatal human SFTS cases in Thailand confirmed by quantitative reverse transcription polymerase chain reaction, in which viral RNA was detected in multiple specimen types from both patients, including plasma, cerebrospinal fluid, respiratory secretions, urine, and stool, indicating systemic infection.
