SuFEx-Enabled Reprogramming of Flavonoids for Selective α-Glucosidase Covalent Inhibition.

Selective inhibition of intestinal α-glucosidase is an established strategy for controlling postprandial hyperglycemia. However, currently available inhibitors are predominantly reversible and nonselective, leading to incomplete target engagement, concomitant α-amylase inhibition, and gastrointestinal side effects associated with gut microbiota perturbation. Here, we report a SuFEx-enabled reprogramming strategy that converts natural flavonoids into highly selective α-glucosidase covalent inhibitors. Guided by the structure of human intestinal maltase-glucoamylase, fluorosulfate warheads were

Natural flavonoids in multiple sclerosis: molecular insights and emerging therapeutic strategies.

Multiple sclerosis is a chronic immune mediated disease in which current disease modifying therapies reduce inflammatory relapses but incompletely address neurodegeneration and remyelination. Natural flavonoids are pleiotropic polyphenols that can modulate immune and glial signaling, oxidative stress, and mitochondrial function. This review synthesizes evidence from experimental models and human studies on flavonoids relevant to multiple sclerosis, emphasizing mechanisms involving NF-κB, Nrf2, inflammasome signaling, and microglia and macrophage polarization that shape oligodendrocyte precurso

Flavonoids as new regulators of mitochondrial potassium channels: contribution to cardioprotection.

OBJECTIVES: Acute myocardial ischemia is one of the major causes of illness in western society. Reduced coronary blood supply leads to cell death and loss of cardiomyocyte population, resulting in serious and often irreversible consequences on myocardial function. Mitochondrial potassium (mitoK) channels have been identified as fine regulators of mitochondrial function and, consequently, in the metabolism of the whole cell, and in the mechanisms underlying the cardioprotection. Interestingly, mitoK channels represent a novel putative target for treating cardiovascular diseases, particularly my
