Enhanced biosynthetic gene expressions and production of ganoderic acids in static liquid culture of Ganoderma lucidum under phenobarbital induction.

Static liquid culture of Ganoderma lucidum, a traditional Chinese medicinal mushroom, is a proven technology for producing ganoderic acids, which are secondary metabolites that possess antitumor properties. In this work, the addition of phenobarbital, a P450 inducer, was used to enhance the production of total and individual ganoderic acids in a two-stage cultivation involving a period of initial shake flask culture followed by static liquid culture of G. lucidum. The dosage and time of phenobarbital induction were critical for the enhanced production of ganoderic acids. The addition of 100 mu

DHN melanin biosynthesis in the plant pathogenic fungus Botrytis cinerea is based on two developmentally regulated key enzyme (PKS)-encoding genes.

Botrytis cinerea is the causal agent of gray mold disease in various plant species and produces grayish macroconidia and/or black sclerotia at the end of the infection cycle. It has been suggested that the pigmentation is due to the accumulation of 1,8-dihydroxynaphthalene (DHN) melanin. To unravel its basis and regulation, the putative melanogenic and regulatory genes were identified and functionally characterized. Unlike other DHN melanin-producing fungi, B. cinerea and other Leotiomycetes contain two key enzyme (PKS)-encoding enzymes. Bcpks12 and bcpks13 are developmentally regulated and ar

The molecular pathogenicity of Fusarium keratitis: a fungal transcriptional regulator promotes hyphal penetration of the cornea.

PURPOSE: The pathogenic mechanisms of fungal infection during human keratomycosis were investigated in an ex vivo corneal model that used strains of Fusarium oxysporum differing in the production of a fungal transcription factor. METHODS: A pacC loss-of-function mutant and a pacC dominant-activating mutant were constructed from a wild-type isolate of F. oxysporum, and the 3 strains were characterized by in vitro growth kinetics. Twenty-seven human donor corneas maintained in tissue culture were superficially scarified and topically inoculated with the wild-type, the pacC loss-of-function muta

Overexpression of an LaeA-like Methyltransferase Upregulates Secondary Metabolite Production in Aspergillus nidulans.

Fungal secondary metabolites (SMs) include medically valuable compounds as well as compounds that are toxic, carcinogenic, and/or contributors to fungal pathogenesis. It is consequently important to understand the regulation of fungal secondary metabolism. McrA is a recently discovered transcription factor that negatively regulates fungal secondary metabolism. Deletion of mcrA (mcrAΔ), the gene encoding McrA, results in upregulation of many SMs and alters the expression of more than 1000 genes. One gene strongly upregulated by the deletion of mcrA is llmG, a putative methyl transferase related

Evolution of beta-lactam biosynthesis genes and recruitment of trans-acting factors.

Penicillins and cephalosporins belong chemically to the group of beta-lactam antibiotics. The formation of hydrophobic penicillins has been reported in fungi only, notably Penicillium chrysogenum and Emericella nidulans, whereas the hydrophilic cephalosporins are produced by both fungi, e.g., Acremonium chrysogenum (cephalosporin C), and bacteria. The producing bacteria include Gram-negatives and Gram-positives, e.g. Lysobacter lactamdurans (cephabacins) and Streptomyces clavuligerus (cephamycin C), respectively. For a long time the evolutionary origin of beta-lactam biosynthesis genes in fung

Bioinformatics of the Paracoccidioides brasiliensis EST Project.

Paracoccidioides brasiliensis is the etiological agent of paracoccidioidomycosis, an endemic mycosis of Latin America. This fungus presents a dimorphic character; it grows as a mycelium at room temperature, but it is isolated as yeast from infected individuals. It is believed that the transition from mycelium to yeast is important for the infective process. The Functional and Differential Genome of Paracoccidioides brasiliensis Project--PbGenome Project was developed to study the infection process by analyzing expressed sequence tags--ESTs, isolated from both mycelial and yeast forms. The PbGe

Global histone acetylation and deacetylation in yeast.

Histone acetyltransferases and deacetylases can be targeted to promoters to activate or repress genes. For example, the histone acetyltransferase GCN5 is part of a yeast multiprotein complex that is recruited by the DNA-binding activator protein GCN4 (refs 1-3). The histone deacetylase RPD3 complex is recruited to DNA by the repressor UME6 (refs 4, 5); similar mechanisms exist in other eukaryotes. However, deletion of RPD3 also increases expression of the PHO5 gene that is repressed by nucleosomes, and regulated by GCN5 (ref. 10) but not by UME6. We have determined whether acetylation and deac

A Dual-Role Transcription Factor FpCys3 Couples Glucose Sensing to Methionine Biosynthesis and Virulence in Fusarium pseudograminearum.

Fusarium pseudograminearum is a soil-borne phytopathogen that causes Fusarium crown rot (FCR) in wheat and barley, posing a severe threat to global grain yield and quality. A deeper understanding of its pathogenicity is essential for developing effective control strategies. Methionine (Met) has been reported that involved in plant-pathogen interactions at multiple levels, regulating both plant immunity and fungal pathogenicity. Here, we identified a conserved bZIP transcription factor, FpCys3, which is a key mediator of methionine biosynthesis in F. pseudograminearum. The FpCys3 mutant exhibit

The GATA factor AreB regulates nitrogen metabolism, fungal development, and aflatoxin production in Aspergillus flavus.

Nitrogen is important for fungal growth and development, and the GATA transcription factor AreA has been widely studied as a key regulator of nitrogen catabolite repression (NCR) in many fungi. However, AreB, another GATA transcription factor in the NCR pathway, remains less studied, and its role in Aspergillus flavus is still unclear. In this study, we characterized areB in A. flavus and investigated its role in regulating nitrogen utilization, fungal growth, and aflatoxin production. The areB gene produces three transcripts, with areB-α being the most abundantly expressed, particularly under

High-Resolution Genome-Wide Occupancy in Candida spp. Using ChEC-seq.

To persist in their dynamic human host environments, fungal pathogens must sense and adapt by modulating their gene expression to fulfill their cellular needs. Understanding transcriptional regulation on a global scale would uncover cellular processes linked to persistence and virulence mechanisms that could be targeted for antifungal therapeutics. Infections associated with the yeast Candida albicans, a highly prevalent fungal pathogen, and the multiresistant related species Candida auris are becoming a serious public health threat. To define the set of a gene regulated by a transcriptional r

Field-derived insights into rice bakanae management: A novel transcription factor RARF1 regulates resistance to respiration-inhibiting fungicides in Fusarium fujikuroi.

A comprehensive survey of field-isolated Fusarium fujikuroi populations was conducted to evaluate their sensitivity to the quinone outside inhibitor (QoI) orysastrobin. The assessment revealed widespread insensitivity to this fungicide, whereas canonical resistance-conferring mutations in the target cytochrome b (cytb) gene were absent in the representative isolates examined. To clarify the non-target site resistance (NTSR) mechanisms driving this phenomenon, we identified a novel Zn2Cys6 transcription factor, FFUJ_00276 (designated as RARF1, Respiration Associated Regulatory Factor 1), which

MrDnaJ, a heat shock protein, regulates stress tolerance and virulence in the entomopathogenic fungus Metarhizium robertsii.

Heat shock proteins are essential molecular chaperones synthesized in response to environmental stresses, yet their roles in entomopathogenic genus Metarhizium remain poorly understood. Here, we identified and characterized MrDnaJ, a heat shock protein DnaJ family member that physically interacts with MrWetA, a key transcription factor in Metarhizium robertsii. MrdnaJ is highly expressed in appressoria and strongly induced by heat stress, with nuclear localization. MrdnaJ knockout mutants (ΔMrdnaJ) exhibited significantly impaired growth and reduced conidial yield under heat stress. Additional

Evaluation of Ogataea polymorpha DUR31 TPP riboswitch as a tool to downregulate gene expression in the yeast Komagataella phaffii.

Komagataella phaffii (syn. Pichia pastoris) is a methylotrophic yeast widely established as host for recombinant protein production and increasingly used as cell factory for C1-based metabolites. Nevertheless, synthetic biology tools for regulating protein expression in this host remain limited, relying largely on promoters of genes from carbon metabolism. Riboswitches, i.e. mRNA elements that regulate translation or splicing via ligand-dependent folding, offer a complementary layer of regulation, yet remain underused in K. phaffii synthetic biology. In the methylotrophic yeast Ogataea polymor

The natural diyne-furan fatty acid EV-086 is an inhibitor of fungal delta-9 fatty acid desaturation with efficacy in a model of skin dermatophytosis.

Human fungal infections represent a therapeutic challenge. Although effective strategies for treatment are available, resistance is spreading, and many therapies have unacceptable side effects. A clear need for novel antifungal targets and molecules is thus emerging. Here, we present the identification and characterization of the plant-derived diyne-furan fatty acid EV-086 as a novel antifungal compound. EV-086 has potent and broad-spectrum activity in vitro against Candida, Aspergillus, and Trichophyton spp., whereas activities against bacteria and human cell lines are very low. Chemical-gene

Differential gene expression to heat stress drives thermotolerance, pathogenicity, and reproduction in Fusarium head blight pathogens: from transcriptomic profiling to functional validation.

BACKGROUND: Fusarium head blight (FHB) is a destructive wheat disease caused primarily by Fusarium graminearum and F. asiaticum. Climate warming increases the frequency of high-temperature episodes during anthesis, yet the molecular mechanisms underlying thermal adaptation in FHB pathogens remain unclear. Here, we integrated ecological, transcriptomic, and functional approaches to investigate how differential gene expression contributes to heat adaptation and its interplay with virulence and reproduction. RESULTS: Field surveys in Sichuan Province identified naturally occurring strains capabl

Manganese homeostasis modulates glucan and chitin unmasking in the opportunistic yeast Candida albicans.

Candida albicans is a commensal fungus and also the most prevalent human fungal pathogen. The ability of this opportunistic yeast to acquire and maintain homeostatic levels of manganese (Mn), particularly in the metal-limited host environment, is an important determinant of its fitness. Recent studies have underscored the importance of Mn acquisition through members of Smf transporters, in C. albicans virulence and its ability to withstand various stresses. In the present study, we undertook transcriptional profiling in the mutant of the Mn transporter Smf12 under restricted Mn availability to

Genome-wide transcriptional profiling identifies HNV1 as a hydroxyurea-responsive negative regulator of virulence in Candida albicans.

Precise DNA replication is essential for maintaining genomic stability, ensuring cellular homeostasis and faithful genetic transmission. Nevertheless, intrinsic and extrinsic factors frequently compromise replication fidelity, leading to replication stress-induced genomic instability, a potential contributor to pathogen virulence. In the fungal pathogen Candida albicans, the global transcriptomic adaptations underpinning the replication stress response and their potential contributions to fungal pathogenicity remain poorly understood. This study employed genome-wide RNA sequencing to map trans

Regulation of gliotoxin biosynthesis and protection in Aspergillus species.

Aspergillus fumigatus causes a range of human and animal diseases collectively known as aspergillosis. A. fumigatus possesses and expresses a range of genetic determinants of virulence, which facilitate colonisation and disease progression, including the secretion of mycotoxins. Gliotoxin (GT) is the best studied A. fumigatus mycotoxin with a wide range of known toxic effects that impair human immune cell function. GT is also highly toxic to A. fumigatus and this fungus has evolved self-protection mechanisms that include (i) the GT efflux pump GliA, (ii) the GT neutralising enzyme GliT, and (i

Intraspecies Transcriptional Profiling Reveals Key Regulators of Candida albicans Pathogenic Traits.

The human commensal and opportunistic fungal pathogen Candida albicans displays extensive genetic and phenotypic variation across clinical isolates. Here, we performed RNA sequencing on 21 well-characterized isolates to examine how genetic variation contributes to gene expression differences and to link these differences to phenotypic traits. C. albicans adapts primarily through clonal evolution, and yet hierarchical clustering of gene expression profiles in this set of isolates did not reproduce their phylogenetic relationship. Strikingly, strain-specific gene expression was prevalent in some
