BACKGROUND: Massive fluid shift after severe burn injury leads to edema and intravascular fluid loss that may result in burn-induced compartment syndrome (BICS) when corrected by aggressive fluid resuscitation. Factors causing this fluid shift remain unclear. Because glycocalyx regulates endothelial permeability, we hypothesized that glycocalyx shedding would increase fluid requirements in burn patients. This prospective cohort study aimed to identify relationships between shedding of the glycocalyx and fluid requirements after burn injury.
METHODS: Patients aged more than 18 years with burn …

UNLABELLED: During collateral artery growth, monocytes adhere to the endothelium and secrete cytokines from the perivascular space promoting arteriogenesis. Recently, the endothelial glycocalyx has been shown to modulate leucocyte infiltration in atherogenic regions. The role of this endothelial surface coating in arteriogenesis, however, has not been investigated so far. We now report that local plasma levels of hyaluronic acid are specifically increased in collateral arterial blood of coronary artery disease patients and hypothesized that components of the endothelial glycocalyx are shed dur…

Based on the amino-acid sequence of the macaque epididymal secretory protein, ESP 13.2 (Q9BEE3/AJ236909), it has now been classified as beta … molgus macaque sperm as they move through the corpus/caudal region of the epididymis. It is a major glycocalyx barrier to the external environment and is retained until the completion of capacitation. Sperm exposed to fluorescein-conjugated poly-L-lysine or Alexa488-histone showed a very uniform fluorescent labeling pattern over the entire sperm surface, alm …

Effect of dibutyryl cyclic AMP on glycocalyx formation in intestinal epithelial cells of salivectomised mice was studied. After salivectomy, a marked reduction of glycocalyx layer on the surface of microvilli was noted. Twenty four hours after the first injection of dibutyrylo cyclic AMP, restoration of the thick layer of glycocalyx was confirmed. The results obtained suggest that dibutyrylo cyclic AMP regulates the synthesis of glycocalyx in Golgi complex.

BACKGROUND: Pulmonary microvascular endothelial cells (MVECs) are not only targets of the highly pathogenic porcine reproductive and respiratory syndrome virus (HP-PRRSV) but also the pivotal hub of inflammation, interacting with alveolar macrophages to amplify the inflammatory response and determine the extent of tissue damage. Glycocalyx disruption in MVECs is often the initiating factor of numerous functional disorders, and astragalus polysaccharide (APS) has been shown to improve glycocalyx integrity in MVECs.
AIM: This study aimed to investigate the mediating role of glycocalyx disruptio…

OBJECTIVE AND DESIGN: Sepsis is a life-threatening condition and a leading cause of in-hospital mortality, characterized by dysregulated inflammatory responses. Using murine models, this study investigated whether Shenfu Injection (SFI), a clinically approved botanical formulation, protects against sepsis by preserving glycocalyx integrity and modulating noncanonical inflammasome signaling mediated by murine caspase-11.
MATERIAL AND SUBJECTS: C57BL/6 mice were subjected to cecal ligation and puncture (CLP) or endotoxemia and treated with SFI or saline. Seven-day survival, organ injury, plasma…

INTRODUCTION: Damage to the endothelial glycocalyx plays a key role in the pathogenesis of sepsis. Syndecan-1 is a widely used biomarker of glycocalyx degradation. The objectives of this study were (1) to determine whether the pathogen type is correlated with differences in glycocalyx disruption in critically ill patients with bacterial bloodstream infection (BSI) and (2) to assess the association of plasma syndecan-1 levels with host response changes implicated in the pathogenesis of sepsis.
METHODS: We measured 24 plasma biomarkers reflective of key pathophysiological domains and whole-bloo…

Sepsis-associated encephalopathy (SAE) is a severe complication of sepsis, frequently accompanied by disruption of the blood-brain barrier (BBB), neuroinflammation, and cerebral edema. The endothelial glycocalyx, an essential component of the BBB, is vital for preserving vascular homeostasis and integrity. However, the specific contribution of glycocalyx degradation in cerebral endothelial cells to BBB permeability and subsequent brain injury in SAE remains inadequately defined. In this study, we hypothesized that glycocalyx degradation is a pivotal early event in SAE pathogenesis and investig…

INTRODUCTION: The administration of intravenous thrombolysis (IVT) before mechanical thrombectomy (MT) in the treatment of acute ischemic stroke (AIS) has been a subject of debate, and its potential benefits remain uncertain. This retrospective study aimed to investigate the effect of preoperative IVT on glycocalyx damage in patients with cerebral ischemia-reperfusion injury (IRI).
METHODS: A cohort of 106 patients with acute large vessel occlusion in the anterior circulation treated with mechanical thrombectomy was enrolled. The levels of the glycocalyx damage marker, syndecan-1, were measur…

Dysfunction of vascular endothelial cells (ECs) facilitates imbalanced immune responses and tissue hyperinflammation. However, the heterogeneous functions of skin ECs and their underlying mechanism in dermatoses remain to be determined. Here, focusing on the pathogenic role of skin ECs in psoriasis, we characterized the molecular and functional heterogeneity of skin ECs from healthy individuals and psoriasis patients at the single-cell level. We found that endothelial glycocalyx destruction, a major feature of EC dysfunction in psoriasis, was a driving force during the process of T cell extrav…

We recently reported that loss of hyaluronan (HA) from the endothelial glycocalyx leads to loss of vessel stability in specific microcirculatory vascular beds. Here we hypothesized that such derangements in the glycocalyx may also impair the adaptive response to vascular ischemia. Endothelial specific conditional hyaluronan synthase 2-KO (Has2-cKO) mice revealed reduced endothelial HA expression and lower hindlimb perfusion at baseline compared to control mice. After a single ligation of the common femoral artery in these mice, we observed dysregulated angiogenesis in the gastrocnemius muscle …

To determine (a) whether chronic heart failure with reduced ejection fraction (HFrEF) is associated with increased glycocalyx shedding; (b) whether glycocalyx shedding in HFrEF with left ventricular dyssynchrony is related to inflammation, endothelial dysfunction and/or redox stress and is ameliorated by cardiac resynchronisation therapy. Glycocalyx shedding has been reported to be increased in heart failure and is a marker of increased mortality. Its role in dyssynchronous systolic heart failure and the effects of cardiac resynchronisation therapy (CRT) are largely unknown. Twenty-six patient…

Vascular leakage is one of the salient characteristics of severe dengue. Nonstructural protein 1 (NS1) of dengue virus (DENV) can stimulate endothelial cells to secrete endothelial hyperpermeability factor, macrophage migration inhibitory factor (MIF), and the glycocalyx degradation factor heparanase 1 (HPA-1). However, it is unclear whether MIF is directly involved in NS1-induced glycocalyx degradation. In this study, we observed that among NS1, MIF and glycocalyx degradation-related molecules, the HPA-1, metalloproteinase 9 (MMP-9) and syndecan 1 (CD138) serum levels were all increased in de…

BACKGROUND: Shear stress induces coronary dilatation via production of nitric oxide (NO). This should involve the endothelial glycocalyx (EG). A greater effect was expected of albumin versus hydroxyethyl starch (HES) perfusion, because albumin seals coronary leaks more effectively than HES in an EG-dependent way.
METHODS: Isolated hearts (guinea pigs) were perfused at constant pressure with Krebs-Henseleit buffer augmented with 1/3 volume 5% human albumin or 6% HES (200/0.5 or 450/0.7). Coronary flow was also determined after EG digestion (heparinase) and with nitro-L-arginine (NO-L-Ag).

Previous work demonstrated that ectodermal cells exhibit greater susceptibility to SARS-CoV-2 than endodermal and mesodermal cells, raising concerns about potential vulnerability of the developing nervous system. We hypothesized that neural stem cells (NSCs) derived from human ectoderm are susceptible to SARS-CoV-2. Using pseudotyped viral particles representing both wild-type and Omicron spike variants, we confirmed efficient transduction in NSCs, with Omicron variants preferentially utilizing endocytosis-mediated entry. Inhibition of endocytosis with filipin and OcTMAB significantly reduced

The endothelial glycocalyx (eGC) is a protein-rich, hairlike structure covering the luminal surface of endothelial cells, vital for vascular health. Aging and cardiovascular disease (CVD) can compromise eGC integrity, potentially preceding conventional indicators of vascular dysfunction such as reduced endothelium-dependent vasodilation and increased arterial stiffness. SDC-1 and HS are key glycosaminoglycans and validated plasma biomarkers reflecting glycocalyx integrity. During menopause the decline in estrogen, a vascular-protective hormone, has been linked to increased glycocalyx degradati…

The endothelial glycocalyx (eGCX), a delicate carbohydrate-rich layer coating the vascular endothelium, critically regulates vascular homeostasis, controlling permeability, thrombosis, and inflammation. Despite its fundamental importance, assessing the morphology of the eGCX remains technically challenging because of its fragile structure, which collapses during conventional fixation. Existing visualization methods require complex preparation, expensive equipment, and fresh tissues, severely limiting accessibility and clinical applicability. Here, we present a practical approach for visualizin…

Phagocytosis is a cornerstone of the immune defence, allowing cells to eliminate pathogens through both opsonic and non-opsonic pathways. In protozoa such as Entamoeba histolytica, non-opsonic phagocytosis is not only central to survival but also to its ability to cause disease, with its life cycle closely intertwined with the intestinal microbiota, which supplies bacteria and their metabolic byproducts to support trophozoite growth. Yet, despite its significance, the molecular basis of how E. histolytica recognises and engulfs bacteria remains largely unresolved. This review examines the cell

Dysfunctional high-density lipoprotein (d-HDL) in chronic kidney disease is known to have a change in composition towards an endothelial-damaging phenotype, amongst others, via the accumulation of symmetric dimethylarginine. The endothelial glycocalyx, a carbohydrate-rich layer lining the endothelial luminal surface, is a first line defense against vascular diseases including atherosclerosis. Here we conducted a translational, cross-sectional study to determine the role of symmetric dimethylarginine in d-HDL as a mediator of glycocalyx damage. Using confocal and atomic force microscopy, intact…

Acute respiratory distress syndrome (ARDS) is a syndrome of acute respiratory failure characterized by major pathologic mechanisms of increased microvascular permeability and inflammation. The glycocalyx lines on the endothelial surface, which determines the vascular permeability, and heparanase play pivotal roles in the degradation of heparan sulfate (HS). HS is the major component of the glycocalyx. The aim of this study is to examine the effects of Ulinastatin (UTI) on vascular permeability and pulmonary endothelial glycocalyx dysfunction induced by lipopolysaccharide (LPS). In our study, C…
