Exposure to glyphosate and risk of non-Hodgkin lymphoma and multiple myeloma: an updated meta-analysis.

OBJECTIVE: We carried out a systematic review and meta-analysis of epidemiologic studies on the association between occupational exposure to glyphosate and risk of non-Hodgkin lymphoma (NHL) and multiple myeloma (MM). METHODS: We conducted a systematic search of the literature, and identified 18 relevant publications, from which we extracted results from seven non-overlapping studies of NHL and three of MM. We performed random-effects meta-analyses for ever-exposure to glyphosate, dose-response, and risk of specific NHL subtypes. RESULTS: The meta-relative risk (RR) of NHL was 1.03 (95% conf…

A comparison of alum sludge with peat for aqueous glyphosate removal for maximizing their value for practical use.

This study compares and contrasts the glyphosate removal efficiency of alum sludge (waterworks residue) and Irish peat in aqueous solution. Organic phosphonate of glyphosate aqueous solution was removed in pot tests separately filled with peat and alum sludge, while effluent samples were taken from each pot to analyse the concentration of phosphorus (P) and COD (chemical oxygen demand); physical and chemical analysis for both media before and after use was carried out subsequently. The results show that the P removal capacity of alum sludge was significant (>99%), while the removal capacity of…

Glyphosate analysis using sensors and electromigration separation techniques as alternatives to gas or liquid chromatography.

Since its introduction in 1974, the herbicide glyphosate has experienced a tremendous increase in use, with about one million tons used annually today. This review focuses on sensors and electromigration separation techniques as alternatives to chromatographic methods for the analysis of glyphosate and its metabolite aminomethyl phosphonic acid. Even with the large number of studies published, glyphosate analysis remains challenging. With its polar and depending on pH even ionic functional groups lacking a chromophore, it is difficult to analyze with chromatographic techniques. Its analysis is…

In vitro assessment of skin sensitization, photosensitization and phototoxicity potential of commercial glyphosate-containing formulations.

This study evaluated the applicability of a modified Direct Peptide Reactivity Assay (DPRA) (OECD N° 442C, 2015) through the 10-fold reduction of reaction volume (micro-DPRA, mDPRA) for skin sensitization evaluation of six commercial glyphosate-containing formulations. In addition, another modification of DPRA was proposed by adding a UVA (5J/cm2) irradiation step, namely photo-mDPRA, to better characterize (photo)sensitizer materials. The phototoxicity profile of pesticides was also evaluated using the 3T3 Neutral Red Uptake Phototoxicity Test (3T3-NRU-PT) (OECD N° 432, 2004). The mDPRA could…

Context-dependent sublethal hazard of glyphosate to non-target predators revealed by functional response analysis.

Glyphosate, one of the most widely used herbicides globally, is a significant environmental contaminant that may affect non-target organisms crucial to ecosystem services, including biological pest control. While glyphosate targets the shikimate pathway in plants, which is absent in animals, there is increasing evidence that glyphosate induces sublethal effects on arthropod predators. However, the factors modulating these effects remain unclear. Here, we investigated how physiological context, specifically pre-treatment with the broad-spectrum antibiotic tetracycline, commonly used to experime…

Glyphosate-based herbicides induce oxidative stress: Insights from human biomarker and in vitro mutagenic studies.

Glyphosate-based herbicides (GBHs) are widely used and raise concerns about human health risks due to the detection of glyphosate (GLY) in body fluids. Their toxicity remains the subject of ongoing debate, especially regarding their carcinogenic potential, while epidemiological evidence associates GBHs exposure with increased non-Hodgkin lymphoma incidence. This study addresses key gaps by characterizing the kinetic profiles of GLY, its metabolites, and oxidative stress biomarkers in urine from 11 agricultural workers over 14 days, alongside evaluating mutagenicity using the AS52 cell model. 1…

Glyphosate promotes calcium oxalate crystal-induced renal injury by modulating the PI3K/Akt-mediated mechanism.

The increasing global prevalence of nephrolithiasis represents a significant public health concern. However, the role of environmental contaminants such as glyphosate remains unclear. Herein, epidemiological analysis, network toxicology, and experimental validation were integrated to investigate this association. Cross-sectional analysis of NHANES data demonstrated a significant dose-dependent relationship between urinary glyphosate levels and nephrolithiasis prevalence. Each unit increase in log-transformed glyphosate concentration was associated with a 25% increase in nephrolithiasis risk (O…

Multigenerational effects of perinatal glyphosate exposure in rats: Developmental alterations, increased mortality and oxidative imbalance.

Glyphosate (Gly), the active ingredient in glyphosate-based herbicides (GBHs), is the most widely used agrochemical, raising concerns about developmental toxicity. This study evaluated the multigenerational effects of perinatal GBH or Gly exposure on fetal and postnatal development. Pregnant Wistar rats received vehicle, GBH, or Gly (2 mg Gly/kg/day) orally from gestational day 9 until weaning. Adult F1 females were mated to generate F2 offspring. In F1 and F2 generations, fetal growth and intrauterine oxidative stress markers were assessed, and postnatal growth and survival were monitored. Gl…

Interfacial engineering-tailored Mn-clay-based nanozyme for glyphosate laccase-amplified sensing and inhibitor screening.

Monitoring pesticide residues and lanthanide metals in water system is crucial for safeguarding human health and environmental integrity. Herein, we precisely designed a type of Mn-clay-based composite nanozyme via interfacial engineering and, for the first time, evaluated the effect of the interfacial configuration of glyphosate (Glyp) on laccase-like activity using in-situ ATR-FTIR spectroscopy and mass spectrometry. DFT calculations revealed that the phosphonate group interacts with Mn - Mn to form inner-sphere complexes with multiple configurations tailoring electron transfer and redox abi…

Immobilization of 4-MBA & Cu2+ on Au nanoparticles modified screen-printed electrode for glyphosate detection.

This study introduces an innovative electrochemical biosensor, engineered through the functionalization screen-printed electrode (SPE) with a coordination complex comprised of 4-mercaptobenzoic acid (4-MBA) and copper ions (Cu2+), achieving precise quantitative determination of glyphosate. Electrodepositing gold nanoparticles (AuNPs) onto the electrode surface, forming a self-assembled monolayer (SAM) of 4-MBA via thiol-gold interactions, and immobilizing Cu2+ via coordination bonding with the monolayer, finalizing the electrochemical biosensor construction as Cu2+/4-MBA/AuNPs/SPE. The success…

Melamine-Based Molecularly Imprinted Monoliths Targeting Glyphosate in Aqueous Media: Synthesis and Binding Mechanism Elucidation.

Cross-linked melamine imprinted monoliths targeting glyphosate were synthesized using 4-phosphonobutanoic acid (PBA) and N-(phosphonomethyl)-iminodiacetic acid (PMIDA) as templates. The binding capacities, evaluated in an aqueous medium, showed that both PMIDA and PBA promoted selective binding sites with imprinting factors of 2.5 and 1.7, respectively. Despite a relatively low imprinting factor, the polymer imprinted with PMIDA showed a noticeably higher binding efficiency in the presence of sodium chloride compared to the nonimprinted reference, demonstrating an ability to selectively target…

Position-specific carbon stable isotope analysis of glyphosate: isotope fingerprinting of molecules within a mixture.

Glyphosate [N-(phosphonomethyl) glycine] is a widely used herbicide and a molecule of interest in the environmental sciences, due to its global use in agriculture and its potential impact on ecosystems. This study presents the first position-specific carbon isotope (13C/12C) analyses of glyphosates from multiple sources. In contrast to traditional isotope ratio mass spectrometry (IRMS), position-specific analysis provides 13C/12C ratios at individual carbon atom positions within a molecule, rather than an average carbon isotope ratio across a mixture or a specific compound. In this work, glyph…

Glyphosate without Co-formulants affects embryonic development of the south african clawed frog Xenopus laevis.

BACKGROUND: Glyphosate (GLY) is the most widely used herbicide in the world. Due to its mode of action as an inhibitor of the 5-enolpyruvylshikimate-3-phosphate synthase, an important step in the shikimate pathway, specifically in plants, GLY is considered to be of low toxicity to non-target organisms. However, various studies have shown the negative effects of GLY on the mortality and development of different non-target organisms, including insects, rodents, fish and amphibians. To better understand the various effects of GLY in more detail, we studied the effects of GLY without co-formulants…

Intergenerational thyroid hormone homeostasis imbalance in cerebellum of rats perinatally exposed to glyphosate-based herbicide.

Agrochemicals became a public health concern due to increased human exposure and possible endocrine disruption effects in several organs, including the brain. Thyroid hormones controls neurodevelopment, which turn them sensitive to endocrine disruptors (EDs). In this work, we evaluated the effect of glyphosate-based herbicides (GBH) as an intergenerational endocrine disrupter on thyroid homeostasis in cerebellar cells. Female pregnant Wistar rats were exposed to Roundup Transorb® solution at 5 and 50 mg/kg/day, from gestation day 18 to post-natal day 5 (P5). Cerebellum of male offspring was us…

Short exposure to glyphosate induces locomotor, craniofacial, and bone disorders in zebrafish (Danio rerio) embryos.

Glyphosate [N-(phosphonomethyl)glycine] is the active ingredient in widely used broad-spectrum herbicides. Even though the toxicity mechanism of this herbicide in vertebrates is poorly understood, evidence suggests that glyphosate is an endocrine disruptor capable of producing morphological anomalies as well as cardiotoxic and neurotoxic effects. We used the zebrafish model to assess the effects of early life glyphosate exposure on the development of cartilage and bone tissues and organismal responses. We found functional alterations, including a reduction in the cardiac rate, significant chang…

A novel inhibition based biosensor using urease nanoconjugate entrapped biocomposite membrane for potentiometric glyphosate detection.

A potentiometric biosensor based on agarose-guar gum (A-G) entrapped bio-nanoconjugate of urease with gold nanoparticles (AUNps), has been reported for the first time for glyphosate detection. The biosensor is based on inhibition of urease activity by glyphosate, which was measured by direct potentiometry using ammonium ion selective electrode covered with A-G-urease nanoconjugate membrane. TEM and FTIR analysis revealed nanoconjugate formation and its immobilization in A-G matrix respectively. The composite biopolymer employed for immobilization yields thin, transparent, flexible membrane hav…
