Molecular detection of Gram-positive bacteria in the human lung through an optical fiber-based endoscope.

PURPOSE: The relentless rise in antimicrobial resistance is a major societal challenge and requires, as part of its solution, a better understanding of bacterial colonization and infection. To facilitate this, we developed a highly efficient no-wash red optical molecular imaging agent that enables the rapid, selective, and specific visualization of Gram-positive bacteria through a bespoke optical fiber-based delivery/imaging endoscopic device. METHODS: We rationally designed a no-wash, red, Gram-positive-specific molecular imaging agent (Merocy-Van) based on vancomycin and an environmental me…

A novel combination approach of human polyclonal IVIG and antibiotics against multidrug-resistant Gram-positive bacteria.

BACKGROUND: Gram-positive bacteria, especially methicillin-resistant Staphylococcus aureus (MRSA) and enterococci, have shown a remarkable ability to develop resistance to antimicrobial agents. OBJECTIVE: We aimed to assess possible enhancement of the antimicrobial activity of vancomycin, amoxicillin, clarithromycin, and azithromycin by human polyclonal intravenous immunoglobulin G (IVIG) against 34 multidrug-resistant (MDR) bacterial isolates, including MRSA, Enterococcus faecium, and Enterococcus faecalis. MATERIALS AND METHODS: Double combinations of the antibiotics with the IVIG were ass…

Treatments for skin and soft-tissue and surgical site infections due to MDR Gram-positive bacteria.

Gram-positive aerobes are the most common organisms in hospitalized patients with skin and soft-tissue infections (SSTIs). Staphylococcus aureus is the most common Gram-positive aerobe among these infections with methicillin-resistant S. aureus (MRSA) the most common pathogen. The increased prevalence of MRSA has been noted in the hospital as well in the community setting. In choosing antimicrobial therapy, assessment of the infection and patient characteristics, such as animal exposure, travel history, underlying diseases, recent trauma, bites, burns, and water exposure, must be considered. Antimicrobial-resistant gram-positive bacteria in PD peritonitis and the newer antibiotics used to treat them.

The incidence of resistant gram-positive bacteria in nosocomial and, more recently, community-acquired infections is increasing. Staphylococci, because of their natural habitat on the skin, have always been the leading cause of peritonitis in patients receiving peritoneal dialysis (PD). These organisms have demonstrated a remarkable ability to develop resistance to antibiotics, first with penicillin, then antistaphylococcal penicillins (methicillin-resistant Staphylococcus aureus), and more recently, strains expressing resistance to vancomycin (vancomycin-intermediate and vancomycin-resistant …

[Post-marketing surveillance of antibacterial activities of cefozopran against various clinical isolates--I. Gram-positive bacteria].

As a post-marketing surveillance, the in vitro antibacterial activities of cefozopran (CZOP), an agent of cephems, against various clinical isolates were yearly evaluated and compared with those of other cephems, oxacephems, penicillins, and carbapenems. Changes in the bacterial susceptibility for CZOP were also evaluated with the resistance ratio calculated with breakpoint MIC. Sixteen species (2,363 strains) of Gram-positive bacteria were isolated from the clinical materials annually collected from 1996 to 2001, and consisted of methicillin-susceptible Staphylococcus aureus (MSSA), methicill…

Antibacterial and antibiofilm activity of a triterpenoid-rich Eucalyptus globulus leaf extract against gram-positive bacteria.

Eucalyptus globulus leaf extract (EGL) is a phytocomplex associated with antimicrobial and anti-inflammatory effects, but a comprehensive characterization of its chemical composition and its relationship with biological activity remains limited. In this study, we combined untargeted metabolomic profiling with functional antibacterial and antibiofilm assays to characterize EGL and to investigate the relationship between its chemical composition and biological activity. Untargeted UHPLC-TWIMS-QTOF analysis revealed a complex profile dominated by polyphenols and pentacyclic triterpenoids. Targete

Zinc-enhanced activity of an antimicrobial halogenated phenazine against Streptococcus mutans and other gram-positive bacteria.

We showed that HP-29 inhibits growth of the oral and systemic pathogen Streptococcus mutans and that its antimicrobial effect is greatly potentiated by the addition of zinc. The zinc-media …

Discovery of guanidinium salt incorporating cinnamic acid skeleton as new antibacterial agents against Gram-positive bacteria.

The global infection rates caused by multidrug-resistant bacteria continue to rise, while the pipeline for novel antibiotics is increasingly drying up, highlighting the urgent need to develop new antimicrobial agents. Drawing inspiration from the amphiphilic structure of cationic antimicrobial peptides (AMPs), a collection of amphiphilic guanidinium salts incorporating cinnamic acid skeleton was designed and prepared. Bioactivity screening revealed that compound 10c exhibited excellent inhibitory effects against Gram-positive bacteria, with MIC values ranging from 1 to 2 μg/mL, comparable to t…

The avermectin, emamectin benzoate, kills gram-positive bacteria and targets the cell envelope of Bacillus subtilis.

The avermectin family of anthelmintics is largely considered to lack antibacterial activity against gram-positive and gram-negative bacteria. Here, we screened six avermectins (ivermectin, eprinomectin, doramectin, abamectin, selamectin, and emamectin benzoate) and a structurally related milbemycin (moxidectin) for antibacterial activity against a panel of representative gram-positive and gram-negative bacteria. We report that emamectin benzoate exhibited activity against several species of gram-positive bacteria, whereas selamectin was active against Staphylococcus aureus and Staphylococcus e…

Novel valproate half-sandwich rhodium and iridium conjugates to fight against multidrug-resistant Gram-positive bacteria.

New valproate Ir(III) and Rh(III) half-sandwich conjugates containing a C,N-phenylbenzimidazole chelated ligand have been synthesized and characterized. The valproic acid conjugation to organometallic fragments seems to switch on the antibacterial activity of the complexes towards Enterococcus faecium and Staphylococcus aureus Gram-positive bacteria.

A toxin-deformation dependent inhibition mechanism in the T7SS toxin-antitoxin system of Gram-positive bacteria.

Toxin EsaD secreted by some S. aureus strains through the type VII secretion system (T7SS) specifically kills those strains lacking the antitoxin EsaG. Here we report the structures of EsaG, the nuclease domain of EsaD and their complex, which together reveal an inhibition mechanism that relies on significant conformational change of the toxin. To inhibit EsaD, EsaG breaks the nuclease domain of EsaD protein into two independent fragments that, in turn, sandwich EsaG. The originally well-folded ββα-metal finger connecting the two fragments is stretched to become a disordered loop, leading to d

The antimicrobial activity and possible synergistic effects of extracts and compounds isolated from Usnea steineri were evaluated against four resistant bacterial species.
