Paraventricular organ of the lungfish Protopterus dolloi: morphology and projections of CSF-contacting neurons.

The morphology and projections of neurons in the paraventricular organ (PVO) were studied by means of silver impregnation after intraocular … ibular and hypothalamic commissures, the commissure of the posterior tuberculum, the postoptic, the habenular, and the anterior commissures. Projections to the infundibulum and the median eminence are relatively sparse; no fibers are labeled in the pituitary gland. Ascending projections to the forebrain are extensive. Major targets include the dorsal hypot …

Leu-enkephalin immunoreactivity in the pig pineal gland.

Leu-enkephalin-positive structures in the pig pineal gland were demonstrated immunohistochemically using mouse monoclonal antibody. The pineal glands were obtained from the newborn, 21-day and 7-month old female pig. The immunopositive nerve fibers were observed in the pineal gland as well as in the epithalamic areas. The leu-enkephalin-immunoreactive nerve fibers (single or forming small bundles) were localized mainly in the proximal part of the pineal and they were scarce in other parts. The localization of the fibers points to a central source of this innervation. The study did not show any

The Wnt/beta-catenin signaling pathway establishes neuroanatomical asymmetries and their laterality.

The vertebrate brain is an immensely complex structure, which exhibits numerous morphological and functional asymmetries. The best described brain asymmetries are found in the diencephalic epithalamus, where the habenulae and the dorso-laterally adjacent pineal complex are lateralized in many species. Research in the past decade has shed light on the establishment of the laterality of these structures as well as their asymmetry per se. In particular work in zebrafish (Danio rerio) has substantially contributed to our understanding, which genetic pathways are involved in these processes. The Wn…

Modulatory effects of Hizikia fusiformis on the dopaminergic system in a neonatal habenular lesion-induced attention-deficit hyperactivity disorder-like rodent model.

BACKGROUND/OBJECTIVES: The dysregulation of the dopamine (DA) system is a key factor in neurodevelopmental and neurodegenerative disorders, including attention-deficit hyperactivity disorder (ADHD), Parkinson's disease, and schizophrenia. The habenula is a crucial regulator of DA neurotransmission and has been implicated in the pathophysiology of ADHD. Hizikia fusiformis (HF), a brown seaweed rich in fucoidan and fucosterol, has demonstrated neuroprotective properties. However, its effects on DA system modulation and neurodevelopmental disorders such as ADHD remain unclear. This study aimed to…

Prefrontal-habenular microstructural impairments in human cocaine and heroin addiction.

The habenula (Hb) is central to adaptive reward- and aversion-driven behaviors, comprising a hub for higher-order processing networks involving the prefrontal cortex (PFC). Despite an established role in preclinical models of cocaine addiction, the translational significance of the Hb and its connectivity with the PFC in humans is unclear. Using diffusion tractography, we detailed PFC structural connectivity with the Hb and two control regions, quantifying tract-specific microstructural features in healthy and cocaine-addicted individuals. White matter was uniquely impaired in PFC-Hb projectio…

Distribution of growth associated protein (GAP-43) immunoreactivity in nerve fibers supplying the pig pineal gland.

An immunohistochemical study of the pig pineal gland was carried out using monoclonal mouse antiserum against growth-associated protein GAP-43. The pineal glands were obtained from the 3, 5, 8 weeks old piglets. The immunopositive nerve fibers were observed in the pineal gland as well as in the habenular and the posterior comissural areas. They formed a dense network in the habenular area and the proximal part of the pineal gland. In the comissural area and in the apical part of the gland. single positive fibers were observed. The obtained results may suggest a difference in the plasticity of …

Endoscopic fenestration at the splenial-habenular junctional area for symptomatic cavum and tumor at the foramen of Monro: case reports and anatomical review.

The splenial-habenular junctional area is an alternative site for neuroendoscopic fenestration to divert CSF flow into the quadrigeminal cistern in cases in which endoscopic third ventriculostomy is not amenable. In some patients with obstructive hydrocephalus, the splenium of the corpus callosum can be elevated from the habenular complex. This exposes the membranous connection between the splenium and habenula, which can be fenestrated to divert the CSF flow into the quadrigeminal cistern. This technique can be performed in patients in whom the foramen of Monro or the third ventricle is block…

Early development of the retina and pineal complex in the sea lamprey: comparative immunocytochemical study.

Lampreys have a complex life cycle, with largely differentiated larval and adult periods. Despite the considerable interest of lampreys for understanding vertebrate evolution, knowledge of the early development of their eye and pineal complex is very scarce. Here, the early immunocytochemical organization of the pineal complex and retina of the sea lamprey was studied by use of antibodies against proliferating cell nuclear antigen (PCNA), opsin, serotonin, and gamma-aminobutyric acid (GABA). Cell differentiation in the retina, pineal organ, and habenula begins in prolarvae, as shown by the app…

Mediation of mPFC-LHb pathway in acupuncture inhibition of cocaine psychomotor activity.

The medial prefrontal cortex (mPFC) and the lateral habenula (LHb) play roles in drug addiction and cognitive functions. Our previous studies have suggested that acupuncture at Shenmen (HT7) points modulates mesolimbic reward system in order to suppress drug-induced addiction behaviours. To explore whether an mPFC-LHb circuit mediates the inhibitory effects of acupuncture on addictive behaviours, we examined the projection from mPFC to LHb, excitation of mPFC neurons during acupuncture stimulation, the effects of optogenetic modulation of mPFC-LHb on HT7 inhibition of cocaine-induced locomotio…

Identification of the subventricular tegmental nucleus as brainstem reward center.

Rewards are essential for motivation, decision-making, memory, and mental health. We identified the subventricular tegmental nucleus (SVTg) as a brainstem reward center. In mice, reward and its prediction activate the SVTg, and SVTg stimulation leads to place preference, reduced anxiety, and accumbal dopamine release. Mice self-stimulate the SVTg, which can also be activated directly by the neocortex, resulting in effective inhibition of the lateral habenula, a region associated with depression. This mechanism may also explain why SVTg suppression induces aversion and increases fear. The trans…

Three Rostromedial Tegmental Afferents Drive Triply Dissociable Aspects of Punishment Learning and Aversive Valence Encoding.

Persistence of reward seeking despite punishment or other negative consequences is a defining feature of mania and addiction, and numerous brain regions have been implicated in such punishment learning, but in disparate ways that are difficult to reconcile. We now show that the ability of an aversive punisher to inhibit reward seeking depends on coordinated activity of three distinct afferents to the rostromedial tegmental nucleus (RMTg) arising from cortex, brainstem, and habenula that drive triply dissociable RMTg responses to aversive cues, outcomes, and prediction errors, respectively. The…

Effects of calcitonin injected into various brain areas on pain threshold and Ca2+ in rats.

AIM: To study the effects of calcitonin (Cal) injected into different brain areas on pain threshold. METHODS: The analgesic effects of Cal were investigated in rats by the tail-flick test. RESULTS: Cal injected into lateral cerebral ventricle (LCV) or periaqueductal gray (PAG) increased obviously the pain threshold to 49 +/- 22% or to 68 +/- 12% (P < 0.01), respectively. When PAG was blocked with lidocaine, the analgesic effect of Cal injected into LCV was lowered 41 +/- 9%. Cal injected into habenula (Hab) decreased the pain threshold to -30 +/- 5% (P < 0.01). CONCLUSION: Cal in different …
