Salivary protein profile of PLHIV on long-term antiretroviral therapy suggests latent reservoir and immune exhaustion.

OBJECTIVE: To compare salivary protein profiles in people living with HIV (PLHIV) undergoing antiretroviral therapy (ART) for different durations. This cross-sectional study examined how long-term ART affects the salivary proteome in PLHIV. DESIGN: Unstimulated saliva was collected from 23 adult men: those on ART for more than 7 years (group HIV.UP7, n = 11) and for up to 7 years (group HIV.DOWN7, n = 12). Samples were analyzed by nano-LC-ESI-MS/MS, followed by label-free quantification and bioinformatic analysis using the UNIPROT database. Differential protein expression was assessed using S

Dysregulation of cytoskeletal organization, energy metabolism, complement cascade, and coagulation associated with suicidal behavior: Insights from proteomic analyses.

Suicidal behavior is highly variable regarding severity of ideation and lethality, which may be reflected in central and systemic proteomic modifications. This study aimed to identify the proteomic profile by mass spectrometry analysis, pathways, and biological processes underlying suicidal behavior. We systematically reviewed three databases, including studies of postmortem brain tissue of suicide completers, and analyzed serum from individuals hospitalized after recent suicide attempts. The Biological General Repository for Interaction Datasets (BioGRID) was used to verify interactions betwe

Proteomic profile of the acquired enamel pellicle of children with early childhood caries and caries-free children.

Acquired enamel pellicle plays an important role in the pathogenesis of early childhood caries (ECC), working as a protective interface between the tooth and the oral cavity. The aim of this cross-sectional in vivo proteomic study was to compare the acquired enamel pellicle protein profile of 3-5-year-old children with ECC (n = 10) and caries-free children (n = 10). Acquired enamel pellicle samples were collected and processed for proteomic analysis (nLC-ESI-MS/MS). In total, 241 proteins were identified. Basic salivary proline-rich protein 1 and 2, Cystatin-B, and SA were found only in the ca

Дробовик и протеомический анализ оварианских протеинов вектора насекомых, Aedes aegypti (Diptera: Culicidae).

Cell physiology and molecular mechanism of anion transport by erythrocyte band 3/AE1.

The major transmembrane protein of the red blood cell, known as band 3, AE1, and SLC4A1, has two main functions: 1) catalysis of Cl-/[Formula: see text] exchange, one of the steps in CO2 excretion, and 2) anchoring the membrane skeleton. This review summarizes the 150-year history of research on red cell anion transport and band 3 as an experimental system for studying membrane protein structure and ion transport mechanisms. Important early findings were that red cell Cl- transport is a tightly coupled 1:1 exchange and band 3 is labeled by stilbenesulfonate derivatives that inhibit anion trans

Detectable Unmetabolized Folic Acid and Elevated Folate Concentrations in Folic Acid-Supplemented Canadian Children With Sickle Cell Disease.

Sickle cell disease (SCD) is an inherited hemoglobinopathy caused by a variant (rs344) in the HBB gene encoding the β-globin subunit of hemoglobin. Chronic hemolytic anemia and increased erythropoiesis and RBC turnover in individuals with SCD can result in increased needs for folate and other B-vitamins. We assessed B-vitamin status, and the distribution of folate forms, including unmetabolized folic acid (UMFA), in Canadian children with SCD supplemented with 1 mg/d folic acid (current routine practice). Non-fasted serum and plasma samples were analyzed for concentrations of folate, and vitam

Interactions of an Anti-Sickling Drug with Hemoglobin in Red Blood Cells from a Patient with Sickle Cell Anemia.

The pathophysiology associated with sickle cell disease (SCD) includes hemolytic anemia, vaso-occlusive events, and ultimately end organ damage set off by the polymerization of deoxygenated hemoglobin S (HbS) into long fibers and sickling of red blood cells (RBCs). One approach toward mitigating HbS polymerization is to pharmacologically stabilize the oxygenated (R) conformation of HbS and thereby reduce sickling frequency and SCD pathology. GBT440 is an α-subunit-specific modifying agent that has recently been reported to increase HbS oxygen binding affinity and consequently delay in vitro po

Anti-diabetic effects of CTB-APSL fusion protein in type 2 diabetic mice.

To determine whether cholera toxin B subunit and active peptide from shark liver (CTB-APSL) fusion protein plays a role in treatment of type 2 diabetic mice, the CTB-APSL gene was cloned and expressed in silkworm (Bombyx mori) baculovirus expression vector system (BEVS), then the fusion protein was orally administrated at a dose of 100 mg/kg for five weeks in diabetic mice. The results demonstrated that the oral administration of CTB-APSL fusion protein can effectively reduce the levels of both fasting blood glucose (FBG) and glycosylated hemoglobin (GHb), promote insulin secretion and improve

An immunogenic, surface-exposed domain of Haemophilus ducreyi outer membrane protein HgbA is involved in hemoglobin binding.

HgbA is the sole TonB-dependent receptor for hemoglobin (Hb) acquisition of Haemophilus ducreyi. Binding of Hb to HgbA is the initial step in heme acquisition from Hb. To better understand this step, we mutagenized hgbA by deletion of each of the 11 putative surface-exposed loops and expressed each of the mutant proteins in trans in host strain H. ducreyi FX547 hgbA. All mutant proteins were expressed, exported, and detected on the surface by anti-HgbA immunoglobulin G (IgG). Deletion of sequences in loops 5 and 7 of HgbA abolished Hb binding in two different formats. In contrast, HgbA protein

Porcine haemoglobin peptides attenuate hypobaric hypoxia-induced fatigue via modulating the gut-muscle axis.

Porcine haemoglobin peptides (PP) exhibit remarkable antioxidant properties and anti-fatigue effects. However, their potential to alleviate fatigue by modulating gut microbiota in high-altitude environments merits further investigation. This study explored the antifatigue effects of PP in hypobaric hypoxia rats. After 10 days of oral administration of PP, rich in branched-chain amino acids, rats exhibited improved exercise endurance and reduced serum fatigue biomarkers. PP treatment ameliorated the activation of Hif-1α and the elevated expression of Atrogin-1 induced by hypoxia, mitigated the

Sickle Cell Disease: Can genetic variability influence pregnancy outcomes?

UNLABELLED: Pregnancy in Sickle Cell Disease (SCD), a severe hereditary genetic condition, highly prevalent in Sub-Saharan African countries, is associated with increased risk of complications and severe outcomes in pregnancy, like intrauterine growth restriction, low birth weight, premature birth, miscarriage, stillbirth, pre-eclampsia, and maternal mortality. Several factors have been identified as associated with the heterogeneity of SCD phenotypes, namely the hemoglobin subunit beta (HBB) haplotype and -3.7 kb α-thalassemia deletion. OBJECTIVE: This study aimed to identify pregnancy compl

Functionality of the V-type ATPase during asexual growth and development of Plasmodium falciparum.

Vacuolar type ATPases (V-type ATPases) are highly conserved hetero-multisubunit proton pumping machineries found in all eukaryotes. They utilize ATP hydrolysis to pump protons, acidifying intracellular or extracellular compartments, and are thus crucial for various biological processes. Despite their evolutionary conservation in malaria parasites, this proton pump remains understudied. To understand the localization and biological functions of Plasmodium falciparum V-type ATPase, we employed CRISPR/Cas9 to endogenously tag the subunit A of the V1 domain. V1A (PF3D7_1311900) was tagged with a t

Herbal Drug use in Sickle Cell Disease Management; Trends and Perspectives in Sub-Saharan Africa - A Systematic Review.

INTRODUCTION: Nigeria has the largest burden of Sickle Cell Disease (SCD) with estimated 100,000 new born affected annually. SCD is a Hemoglobin (Hb) disorder with the major form resulting from the substitution of a polar glutamate (Glu) by non-polar Valine (Val) in an invariant region of Hbβ chain-subunit. Species of Hb found in the sickle cell trait are HbA and HbS in a 60:40 proportion, in SCD only HbS, in the HbC disease only HbC, and in the SC disease it's HbS and HbC in a 50:50 equal proportion. OBJECTIVE: This paper reviews herbal medicines usage in sub-Saharan Africa (sSA) to ameliora

Paired proteomic analysis reveals protein alterations in sun-exposed skin of professional drivers : 1Laboratory for structural and Computational Proteomics, Carlos Chagas Institute, Fiocruz, Paraná, Brazil.

Professional drivers represent an ideal cohort for investigating the effects of solar radiation on skin due to their unique, asymmetric exposure to sun, a consequence of vehicle window orientations. Consequently, one side of the face is naturally subjected to more solar radiation, resulting in uneven sunlight exposure. This scenario supports a paired experimental design for precise within-individual comparisons, crucial for assessing sun exposure's impact on skin health, including signs of aging. Leveraging this approach, our study reveals sun-induced overexpression of proteins linked to photo
