SPARC (secreted protein acidic and rich in cysteine) of the intestinal nematode Strongyloides ratti is involved in mucosa-associated parasite-host interaction.

The secreted protein acidic and rich in cysteine (SPARC), found in the excretory/secretory products of Strongyloides ratti, is most strongly expressed in parasitic females. Since SPARC proteins are involved in the modulation of cell-matrix interactions, a role of the secreted S. ratti SPARC (Sr-SPARC) in the manifestation of the parasite in the host's intestine is postulated. The full-length cDNA of Sr-SPARC was identified and the protein was recombinantly expressed. The purified protein was biologically active, able to bind calcium, and to attach to mucosa-associated human cells. Addition of

Erythrocyte invasion by Babesia parasites: current advances in the elucidation of the molecular interactions between the protozoan ligands and host receptors in the invasion stage.

During an asexual growth cycle of Babesia parasites in a natural host, the extracellular merozoites invade (i.e., attach to, penetrate, and internalize) the host erythrocytes (RBC) via multiple adhesive interactions of several protozoan ligands with the target receptors on the host cell surface. After internalizing the host RBC, they asexually multiply, egress from the RBC by rupturing the host cells, and then invade the new RBC again. In the invasion stage, several surface-coating molecules of merozoites might be involved in the initial attachment to the RBC, while proteins secreted from apic

A review of filarial nematodes parasitizing tick vectors: unraveling global patterns in species diversity, host associations, and interactions with tick-borne pathogens.

Ticks are well-known vectors of pathogens, including filarial nematodes, which can affect animal and human health. This review synthesizes current knowledge of filarial nematodes that utilize ticks as vectors, focusing on the Onchocercidae. Five genera of onchocercid filarial nematodes (Cercopithifilaria, Cherylia, Cruorifilaria, Monanema, and Yatesia) have demonstrated vector-parasite relationships with ticks, while Acanthocheilonema has only demonstrated molecular detection without confirmed vector competence. Vector competence has been experimentally proven for several species through docum

Evans blue staining reveals vascular leakage associated with focal areas of host-parasite interaction in brains of pigs infected with Taenia solium.

Cysticidal drug treatment of viable Taenia solium brain parenchymal cysts leads to an acute pericystic host inflammatory response and blood brain barrier breakdown (BBB), commonly resulting in seizures. Naturally infected pigs, untreated or treated one time with praziquantel were sacrificed at 48 hr and 120 hr following the injection of Evans blue (EB) to assess the effect of treatment on larval parasites and surrounding tissue. Examination of harvested non encapsulated muscle cysts unexpectedly revealed one or more small, focal round region(s) of Evans blue dye infiltration (REBI) on the surf

Co-expression analysis of lncRNA and mRNA suggests a role for ncRNA-mediated regulation of host-parasite interactions in primary skin lesions of patients with American tegumentary leishmaniasis.

Leishmaniasis, caused by different Leishmania species, manifests as cutaneous or visceral forms. In the American continent, the cutaneous form is called American tegumentary leishmaniasis (ATL) and is primarily caused by Leishmania (Viannia) braziliensis. Mucosal leishmaniasis (ML), the most severe form of ATL, arises in approximately 20% of patients from a primary cutaneous lesion. Evidence indicates changes in overall expression patterns of mRNAs and lncRNAs of the host in response to Leishmania infection, with the parasite capable of modulating host immune response, which may contribute to

Fasciola hepatica: comparative effects of host resistance and parasite intra-specific interactions on size and reproductive histology in flukes from rats infected with isolates differing in triclabendazole sensitivity.

The efficacies of putative fasciolicides and vaccines against Fasciola hepatica are frequently monitored in clinical and field trials by determination of fluke egg output in host faeces and by worm counts in the host liver at autopsy. Less often used are parameters based on fluke size and histology, yet these can provide important indications of specific effects on the development of particular germ-line or somatic tissues, especially in relation to the timing and profligacy of egg production. In this study, F. hepatica metacercariae of two distinct isolates, the triclabendazole (TCBZ)-sensiti

Hymenolepis diminuta infections in tenebrionid beetles as a model system for ecological interactions between helminth parasites and terrestrial intermediate hosts: a review and meta-analysis.

The cestode Hymenolepis diminuta (Cyclophyllidea) uses a variety of insects as its intermediate host, where ingestion of eggs results in development in the hemocoel of a cysticercoid that is infective to a rat definitive host. Species in 2 genera, Tenebrio and Tribolium (Coleoptera: Tenebrionidae) have been used extensively as laboratory intermediate hosts. This review examines experimental studies on ecological aspects of the relationship between H. diminuta and tenebrionid beetles, including the acquisition and establishment of the parasite, host effects on the parasite, and parasite effects

Proteomic analysis of Sarcoptes scabiei reveals that proteins differentially expressed between eggs and female adult stages are involved predominantly in genetic information processing, metabolism and/or host-parasite interactions.

Presently, there is a dearth of proteomic data for parasitic mites and their relationship with the host animals. Here, using a high throughp … s (myosin and troponin), are proposed to be involved in key biological processes within S. scabiei, host-parasite interactions and/or the pathogenesis of scabies. These proteomic data will enable future molecular, biochemical and physiological investigations of early developmental stages of S. scabiei and the discovery of novel interventions, targeting the egg stage, given …

Research note: The critical role of the interaction between Eimeria tenella invasion protein RON2 and host receptor annexin A2 in mediating parasite invasion.

Avian coccidiosis, caused by protozoan parasites of the genus Eimeria, is a globally prevalent and highly pathogenic disease that poses a serious threat to the poultry industry, resulting in significant economic losses. However, the mechanism by which Eimeria species invade host cells remains unclear. Previous studies have identified rhoptry neck protein 2 (RON2) from Eimeria tenella as a critical factor in host cell invasion, but a comprehensive understanding of the role of EtRON2 in host cell invasion and its relationship with E. tenella invasion is lacking. To address this gap, this study f

How parasite exposure and time interact to determine Australapatemon burti (Trematoda: Digenea) infections in second intermediate hosts (Erpobdella microstoma) (Hirudinea: Erpodellidae).

Australapatemon spp. are cosmopolitan trematodes that infect freshwater snails, aquatic leeches, and birds. Despite their broad geographic distribution, relatively little is known about interactions between Australapatemon spp. and their leech hosts, particularly under experimental conditions and in natural settings. We used experimental exposures to determine how Australapatemon burti cercariae dosage (number administered to leech hosts, Erpobdella microstoma) affected infection success (fraction to encyst as metacercariae), infection abundance, host survival, and host size over the 100 days

Macrophage Migration Inhibitory Factor in Psoroptes ovis: Molecular Characterization and Potential Role in Eosinophil Accumulation of Skin in Rabbit and Its Implication in the Host-Parasite Interaction.

Psoroptes ovis, a common surface-living mite of domestic and wild animals worldwide, results in huge economic losses and serious welfare issues in the animal industry. P. ovis infestation rapidly causes massive eosinophil infiltration in skin lesions, and increasing research revealed that eosinophils might play an important role in the pathogenesis of P. ovis infestation. Intradermal injection of P. ovis antigen invoked massive eosinophil infiltration, suggesting that this mite should contain some relative molecules involved in eosinophil accumulation in the skin. However, these active molecul

Zone of Interaction Between the Parasite and the Host: Protein Profile of the Body Cavity Fluid of Gasterosteus aculeatus L. Infected with the Cestode Schistocephalus solidus (Muller, 1776).

PURPOSE: During infection, the host and the parasite "communicate" with each other through various molecules, including proteins. The aim of this study was to describe the excretory-secretory proteins from the helminth Schistocephalus solidus and its intermediate host, the three-spined stickleback Gasterosteus aculeatus L., which are likely to be involved in interactions between them. METHODS: Combined samples of washes from the G. aculeatus sticklebacks cavity infected with the S. solidus, and washes from the parasite surface were used as experimental samples, while washes from the uninfecte

[Structural organization and function of parasitic systems].

The structural organization and functioning of parasitic systems are considered in the context of the general theory of systems. By using the parasitic system of an epizootic process of plague as an example, the essence of the concepts of the structural organization and functioning of the systems is determined, and a scheme of a relationship of elements in the trinomial parasitic systems is presented. The mechanism of parasite-host interaction at the molecular, cellular, body's, and populational levels is considered on the basis of the theory of immune-specific wave information in an living or

Current and future strategies against cutaneous parasites.

Cutaneous parasites are identified by their specific cutaneous symptoms which are elicited based on the parasite's interactions with the host. Standard anti-parasitic treatments primarily focus on the use of specific drugs to disrupt the regular function of the target parasite. In cases where secondary infections are induced by the parasite itself, antibiotics may also be used in tandem with the primary treatment to deal with the infection. Whilst drug-based treatments are highly effective, the development of resistance by bacteria and parasites, is increasingly prevalent in the modern day, th

Long-term trends in parasite diversity and infection levels: approaches and patterns.

Parasites exist in every ecosystem, affecting nearly all organisms and playing a complex role in human societies. On the one hand, they contribute substantially to biodiversity and support ecosystem stability by performing essential ecological functions. On the other, they can impose health burdens on their hosts, causing diseases in both animals and humans. Despite their significance, our understanding of how parasitic organisms are affected by human-driven environmental change remains poor. In other well-studied groups such as free-living birds, mammals and insects, long-term ecological data

Drivers of parasite sharing among Neotropical freshwater fishes.

Because host-parasite interactions are so ubiquitous, it is of primary interest for ecologists to understand the factors that generate, maintain and constrain these associations. Phylogenetic comparative studies have found abundant evidence for host-switching to relatively unrelated hosts, sometimes related to diversification events, in a variety of host-parasite systems. For Monogenoidea (Platyhelminthes) parasites, it has been suggested that the co-speciation model alone cannot explain host occurrences, hence host-switching and/or non-vicariant modes of speciation should be associated with t…

Ensemble modeling highlights importance of understanding parasite-host behavior in preclinical antimalarial drug development.

Emerging drug resistance and high-attrition rates in early and late stage drug development necessitate accelerated development of antimalarial compounds. However, systematic and meaningful translation of drug efficacy and host-parasite dynamics between preclinical testing stages is missing. We developed an ensemble of mathematical within-host parasite growth and antimalarial action models, fitted to extensive data from four antimalarials with different modes of action, to assess host-parasite interactions in two preclinical drug testing systems of murine parasite P. berghei in mice, and human …
