The effect of irsogladine [2,4-diamino-6-(2,5-dichlorophenyl)-s-triazine maleate], an antiulcer drug, on contents of cyclic nucleotides including cAMP and cGMP was investigated in rat stomachs. Irsogladine concentration-dependently increased cAMP content in rat glandula stomach. However, irsogladine at higher concentration (10(-5) M) was unable to further increase cAMP level in the presence of non-selective phosphodiesterase (PDE) inhibitor 3-isobutyl-1-methylxanthine, although 3-isobutyl-1-methylxanthine by itself increased cAMP level. Kinetics of hydrolysis of N,N- and N,S-dimethyl-2-thiophenobarbital and products of this reaction were investigated. The UV spectroscopy served as a tool for kinetic investigations and chromatography was used to separate and isolate the main products of hydrolysis. These products were identified by spectroscopic methods and the course of hydrolysis of both isomers was compared.

This study was conducted to explore the mechanism of the pharmacokinetic interaction between aspirin (ASP) and indomethacin (IND) using rat erythrocytes (RBCs) and hepatocytes. Two aspartic carboxyl groups act as key catalytic groups in the active site of an aspartic protease. We synthesized an aspartic protease analogue by positioning three salicylate residues in close proximity on a cross-linked polystyrene. The immobile artificial protease effectively hydrolyzed albumin into many small fragments by the catalytic action of carboxyl groups contained in the active site. The artificial protease manifested optimum activity at pH 3 just as aspartic proteases.

Optimum conditions for enzymatic hydrolysis of bovine blood in the presence of brewer's yeast Saccharomyces carlsbergensis were studied. In addition to the amount of blood, yeast, sodium citrate, and ethanol in the mixture, the optimum process duration and temperature were determined. The yield of the major end product (blood hydrolysate) increased from 11 to 55%, and the amino acid composition of the end product was brought to a balanced level as a result of hydrolysis optimization and addition of pancreatin to the reaction mixture.
