Three novel antimicrobial peptides (AMPs), named panurgines (PNGs), were isolated from the venom of the wild bee Panurgus calcaratus. The dodecapeptide of the sequence LNWGAILKHIIK-NH₂ (PNG-1) belongs to the category of α-helical amphipathic AMPs. The other two cyclic peptides containing 25 amino acid residues and two intramolecular disulfide bridges of the pattern Cys8-Cys23 and Cys11-Cys19 have almost identical sequence established as LDVKKIICVACKIXPNPACKKICPK-OH (X=K, PNG-K and X=R, PNG-R). All three peptides exhibited antimicrobial activity against Gram-positive bacteria and Gram-negative

We report the infestation of stingless bee nests by the mite Pyemotes tritici, which killed four colonies of Tetragonisca angustula and one colony of Frieseomelitta varia in Brazil. The first infected colony, a colony of T. angustula, came from an area between Uberlândia and Araguari, Minas Gerais. The transfer of the mites to the other colonies occurred through the transfer of infected combs and subsequent manipulations. Other colonies in the same meliponary, which had not been manipulated, were not infected. The infestation was terminated by isolating the dead colonies from the meliponary.

BACKGROUND: Hymenoptera venom allergy represents a potentially life-threatening condition. Venom immunotherapy (VIT) is the treatment of choice in patients with systemic reactions.
CASE PRESENTATION: We report the case of a 65-year-old male, who experienced systemic reactions following hymenoptera stings. In vitro and in vivo allergy tests revealed sensitization predominantly to Polistes dominula and specific VIT was initiated. During VIT, the patient experienced two subsequent stings with only local reactions. After 5 years of VIT, repeated serological evaluation and skin tests turned negati…

We present a review about histological sub-lethal effects due to anthropogenic contaminants on the midgut of bees and other aculeate hymenopterans. Contaminant types, damage types, and methodology were extracted and summarized from 74 published articles, and then quantitatively analyzed. We found that the Western honeybee (Apis mellifera) is by far the most widely used model. Contaminants have largely been tested under laboratory conditions, particularly insecticides and fungicides. Tissue-level damage (e.g., degradation of epithelium and of peritrophic membrane) were often detected together w…

Hymenoptera venom allergy (HVA) is an IgE-mediated hypersensitivity reaction caused by Hymenoptera species stings (honeybee, vespid, or ant). The only effective treatment is Hymenoptera venom immunotherapy (VIT). Our study aimed to evaluate whether humoral and cellular biomarkers measured before, during, and after honeybee VIT are associated with the success of VIT, which was assessed by the response to a sting challenge one year after finishing VIT. In this prospective study, blood biomarkers of 25 patients undergoing honeybee VIT at the referral center in Slovenia were evaluated. A controlle…

The use of insecticides based on metabolites found in live organisms, such as the insecticide spinosad, has been an option for the control of agricultural pests because of the allegedly low toxicological risk for nontarget arthropods, such as stingless bees. In the current study, we evaluate the effects of chronic oral exposure to spinosad during the larval phase on survival, developmental time, body mass, midgut epithelial remodeling, and the peritrophic matrix (PM) of Partamona helleri stingless bee workers. Worker larvae that were raised in the laboratory were orally exposed to different co

BACKGROUND: Trichogramma pretiosum Riley is an important egg parasitoid and biological control agent of caterpillar pests. We studied the acute toxicity of 20 pesticides (14 insecticides/miticides, three fungicides and three herbicides) exposed to recommended field rates. Egg, larval, and pupal stages of the parasitoid in their hosts were dipped in formulated solutions of the pesticides and evaluated 10 days later for percentage of host eggs with holes, number of parasitoids emerged per egg with holes, and stage-specific mortality of immature as well as adult wasps within the host eggs.

New molecular studies suggested that the family Melittidae is either a paraphyletic group from which all the other bees are derived, or the sister clade to all other existing bees. Studying the historical biogeography and evolution of each major lineage within this group is a key step to understand the origin and early radiation of bees. Melitta is the largest genus of melittid bees, for which a robust molecular phylogeny and a biogeographic analysis are still lacking. Here, we derive a phylogenetic hypothesis from the sequences of seven independent DNA fragments of mitochondrial and nuclear o

BACKGROUND: According to current guidelines, skin testing for hymenoptera venom allergy should be performed in a stepwise manner, maintaining 15- to 20-min intervals between the injections of venom. Given the long-winded procedure of sequential skin testing, we retrospectively explored the safety of simultaneous intradermal testing.
METHODS: Four hundred and seventy-eight consecutive patients with a convincing history of an anaphylactic reaction after a hymenoptera sting were tested. All venom concentrations (0.02 ml of 0.001, 0.01, 0.1, and 1.0 μg/ml of honey bee and wasp venom) were adminis…

Beekeepers apply various dusts to honey bee, Apis mellifera L., colonies to dislodge parasitic mites and control bacterial brood diseases. Anecdotal reports by beekeepers indicate that the antibiotic oxytetracycline (OTC) can be toxic when applied in powdered sugar to cells containing immature bee brood, but it was not known whether the purported toxicity is caused by the antibiotic or the sugar carrier. Additionally, the toxicity of various dusts, proposed for parasitic mite control, is poorly known. In the current studies, we tested OTC and two other antibiotics (tylosin and lincomycin, cand

Honey bee (Apis mellifera L.) colonies bred for hygienic behavior were tested in a large field trial to determine if they were able to resist the parasitic mite Varroa destructor better than unselected colonies of"Starline" stock. Colonies bred for hygienic behavior are able to detect, uncap, and remove experimentally infested brood from the nest, although the extent to which the behavior actually reduces the overall mite-load in untreated, naturally infested colonies needed further verification. The results indicate that hygienic colonies with queens mated naturally to unselected drones had s

We studied Hymenoptera stings in 72 pest-control operators without any previous systemic reactions to Hymenoptera stings, and investigated their venom-specific IgE levels in serial specimens collected over one year. At the initial evaluation, venom-specific IgE was present in 25 (34.7%) of 72 pest-control operators, and venom-specific IgE titer significantly decreased as the time interval from the last sting increased (p < 0.001). In most cases, venom-specific IgE disappeared less than 3 years after the last sting. On the other hand, the ratio of subjects with positive CAP for venom-specific I…

Data are summarized in this Hymenoptera venom study (HVS) article on the safety of skin testing with venom extracts. Of the 3236 subjects studied, 89% had experienced an historical sting systemic reaction (SSR). Seventy-four percent of all subjects and 76% of subjects who had experienced an historical SSR had a positive skin test to at least one venom. More subjects tested positive to yellow jacket venom (51.8%) than to any other venom. There were no significant differences of the wheal and erythema sizes associated with different venoms or different historical sting reactions. Forty-five perc…

Pure venom from Solenopsis invicta was collected by having the insects sting into a capillary tube. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) demonstrated that three major protein bands were present in the venom. A prototype commercial extract was compared and found to contain the three major venom proteins and additional components as well as high potency and specificity by RAST. Immunoblot studies were performed with sera from allergic individuals with blots prepared from denatured SDS-PAGE gels and from two types of nondenaturing gels. The nondenaturing systems de

Two female Thraxan sp. (Diptera: Bombyliidae: Anthracinae) emerged from parasitised Pison simillimum Smith (Hymenoptera: Crabronidae) larvae found inside a Sceliphron formosum Smith (Hymenoptera: Sphecidae) mud wasps nest. Thraxan sp. larvae are endoparasites of P. simillimum larvae. The endoparasite pupates inside the host larva, kills it, and emerges free of the host body. We describe and illustrate the pupal exuviae of the newly found species and two known species, T. luteus Yeates Lambkin and T. misatulus Yeates Lambkin, and we compare these pupal exuviae to those of Anthrax Scopoli.

Bumblebees (Bombus spp.) are important pollinators that are widely used for greenhouse crop pollination and are produced annually by commercial breeding facilities. In this study, we used the commercially important bumblebee species Bombus terrestris Linnaeus (Hymenoptera: Apidae: Bombini) to evaluate the effects of artificial box stocking density on the reproductive performance of newly eclosed females under commercial rearing conditions. Newly eclosed gynes and workers were artificially held in separate wooden boxes at different stocking densities (1 to 150 individuals per box for gynes and …

The nest of leaf-cutting ants is founded by a newly mated queen, which lays the first eggs, giving rise to workers responsible for nest construction and colony maintenance. Bioecological and physiological aspects of queens may help explain variations in the size of newly founded nests of species in the genus Atta. This study aimed to evaluate the relationship between ovary and spermatheca morphology and the oviposition rate of Atta sexdens Linnaeus (Hymenoptera: Formicidae) queens. Forty-one newly founded nests of A. sexdens were collected, and the volume of each fungus garden was measured. Th…

The male genitalia of insects are among the most variable, complex, and informative character systems for evolutionary analysis and taxonomic purposes. Because of these general properties, many generations of systematists have struggled to develop a theory of homology and alignment of parts. This struggle continues to the present day, where fundamentally different models and nomenclatures for the male genitalia of Hymenoptera, for example, are applied. Here, we take a multimodal approach to digitalize and comprehensively document the genital skeletomuscular anatomy of the bullet ant (Paraponer…
