Differential effects of homocytotropic antibodies on the mast cell and anaphylactic responses in actively and passively sensitized mice.

The relationship between mast cell degranulation, anaphylaxis, and the production of homocytotropic antibodies was examined in ICR mice after trichinella infection. It was found that the active cutaneous response to trichinella antigen had a rapid onset (within 2 weeks) and the sensitivity increased over a 9-week period after infection. Two types of mast cell degranulation were observed: (i) moderate, with few extracellular, deep-blue-staining granules (May-Grunwald-Giemsa), and (ii) "explosive," with many extracellular reddish-staining, swollen granules, indicating an apparent breakdown of ce

Placental Transfer of Dengue Virus (DENV)-Specific Antibodies and Kinetics of DENV Infection-Enhancing Activity in Brazilian Infants.

BACKGROUND: Maternal-fetal transferred dengue virus (DENV)-specific antibodies have been implicated in the immunopathogenesis of dengue during infancy. METHODS: A prospective birth cohort was established in a dengue-endemic area in the Northeast Region of Brazil. DENV-specific immunoglobulin G (IgG) and DENV-1-4 serotype-specific neutralizing antibody (NAb) levels were assessed in 376 paired maternal and umbilical cord blood samples. The kinetics of enhancing activity by maternally acquired DENV antibodies was determined in serum samples from children enrolled in the cohort. RESULTS: Mothers

Protection against scrub typhus infection engendered by the passive transfer of immune sera.

The passive transfer of convalescent sera did not protect the majority of mice against challenge with the homologous strain and was completely ineffective against challenge with strains unrelated by fluorescent antibody techniques. When the immune sera was incubated with the rickettsia in vitro and then inoculated into the mice a dramatic increase occurred in the number of surviving mice. The importance of these data in relation to published results with other species of rickettsia is discussed.

Timing of Maternal Immunization Affects Immunological and Behavioral Outcomes of Adult Offspring in Siberian Hamsters (Phodopus sungorus).

Maternal influences are an important contributing factor to offspring survival, development, and behavior. Common environmental pathogens can induce maternal immune responses and affect subsequent development of offspring. There are likely sensitive periods during pregnancy when animals are particularly vulnerable to environmental disruption. Here we characterize the effects of maternal immunization across pregnancy and postpartum on offspring physiology and behavior in Siberian hamsters (Phodopus sungorus). Hamsters were injected with the antigen keyhole limpet hemocyanin (KLH) (1) prior to p

Vaccine immune response and interference of colostral antibodies in calves vaccinated against rabies at 2, 4 and 6 months of age born from antirabies revaccinated females.

Considering the high prevalence of rabies in cattle, we aimed to evaluate the interference of colostral antibodies transferred to calves after birth and the benefit of administering an antirabies vaccination in two-month-old calves compared to vaccinating at 4 and 6 months of age. Calves born from females revaccinated against rabies during the third trimester of pregnancy were studied. Forty-eight hours after parturition, blood samples from dams and offspring were collected, and antirabies neutralizing antibody titers were analyzed using the Rapid Focus Fluorescent Inhibition Test. We found th

Kinetics of decline of maternal measles virus-neutralizing antibodies in sera of infants in France in 2006.

The optimal age for measles vaccination is an important health issue, since maternal antibodies may neutralize the vaccine antigen before a specific immune response develops, while delaying vaccination may increase the risk of complicated diseases in infants. However, measles vaccination impacts the duration of protection afforded by transplacental transfer of maternal antibodies: vaccination-induced maternal antibodies disappear faster than disease-induced antibodies. In order to maintain protection against measles in infants, it is important to monitor the dynamics of this phenomenon in vacc

Genetic control of immunity to parasites: adoptive transfer of immunity between inbred strains of mice characterized by rapid and slow immune expulsion of Trichinella spiralis.

Adoptive transfer of immunity with immune mesenteric lymph node cells (IMLNC) was used to analyse the roles of immune and inflammatory events in determining the strain-characteristic time of expulsion of Trichinella spiralis from mice. Transfer of IMLNC within and between three rapidly responding strains (NIH, SWR, DBA1-all H-2q) resulted in accelerated worm expulsion, worm loss commencing before day 8 in each case. When NIH cells were transferred to slow-responder B10 congenic mice (B10G-H-2q) mice, immunity was evident at 8 days as a reduction in worm fecundity and only by 12 days as a reduc

The long term effect of age and maternally derived antibodies against foot and mouth disease on the serological response following vaccination in young dairy calves.

In Israel, occurrence of foot and mouth disease (FMD) in dairy farms is rare. However, when FMD outbreaks occur, dairy calves are the most affected, despite routine vaccination. Contradictory findings exist regarding the effect of age and maternally derived antibodies (MDA) on the serological response following vaccinations against FMD in dairy calves. Furthermore, the long term effect of FMD vaccination regimen during early life was rarely assessed. This study was conducted in order to assess both the short and long term effects. In total 44 non-vaccinated calves were divided into four groups

Immunological priming of one dose of inactivated hepatitis A vaccine given during the first year of life in presence of maternal antibodies.

BACKGROUND: In hepatitis A virus (HAV)-seronegative infants, inactivated hepatitis A vaccines are highly immunogenic. On the contrary, in infants who are HAV-seropositive before vaccination, the interfering effect of passively-transferred maternal anti-HAV antibodies leads to lower post-primary immunization anti-HAV levels, as compared to those achieved by seronegative infants. One possible way to overcome this drawback is to delay hepatitis A vaccination later during the first year of life. The objective of the study was to document the immunogenicity of an inactivated hepatitis A vaccine in

Rapid expulsion of Trichinella spiralis in suckling rats.

Orally administered Trichinella spiralis muscle larvae were rapidly expelled by rat pups suckling an immune dam. The immunity was delivered in the milk; substantial resistance was conferred on normal rat pups suckled for only 24 hours by a Trichinella-immune foster mother. The pups were protected by oral or systemic administration of specific serum antibodies. When infused into a normal lactating dam, these antibodies accumulated in the serum of her suckling pups.

Suppressive effect of bacterial endotoxin on the expression of cell-mediated anti-Listeria immunity.

Intravenous injection of bacterial endotoxin into mice at any time during ongoing infection with Listeria monocytogenes resulted in a markedly increased multiplication of this organism in the liver and spleen. Experiments designed to investigate the basis of this infection-enhancing effect revealed that endotoxin was also capable of inhibiting the expression of adoptive T-cell-mediated anti-Listeria immunity if given to normal recipient mice up to 48 h before they were infused with protective T-cells. On the other hand, endotoxin had only a marginal effect on the expression of adoptive immunit

Human antiserum for prevention of the local shwartzman reaction and death from bacterial lipopolysaccharides.

Bacterial lipopolysaccharides from dead bacteria have been blamed for the continuing high mortality from gram-negative infections despite antibiotic treatment. Because animal antiserum against these lipopolysaccharides has been shown to protect against several of the effects of endotoxin, we undertook the development of antiserum in human subjects. 21 men were immunized with a single injection of Salmonclla typhimurium or Escherichia coli 0:111 heat-killed cells and immune serum was collected at 2 wk. Preimmune serum was obtained as a control in all animal experiments. 1 ml antiserum given int

Advancing protective effects of maternal antibodies in neonates through animal models.

Maternal immunity is critical for neonatal defense during early life, when infants are highly susceptible to infection. Antibodies transferred across the placenta or through breast milk not only provide immediate protection but also shape immune development. Defining the mechanisms and outcomes of this passive protection is essential for guiding maternal vaccination strategies and reducing the global burden of early-life infections. Animal models are indispensable for studying maternal antibody effects on neonatal immunity because they allow controlled investigation of transfer and protective

Resistance against Trichinella spiralis infection in pups delivered by T. spiralis-infected dam.

Maternal antibody transmission via placenta and breastmilk are known to confer protection in infants. In this study, we investigated the maternal immunity transmission in pups delivered by rats infected with Trichinella spiralis and assessed the resulting resistance against subsequent parasitic infection. Our results revealed that parasite-specific IgG, IgG1 and IgG2a antibodies were present in pups prior to breastmilk ingestion (pre-milk), in which IgG and IgG1 antibodies persisted until week 8 after birth while parasite-specific IgG2a antibodies only lasted until week 4. After weaning on wee

Analysis of rubella antibody distribution from newborn dried blood spots using finite mixture models.

Eluted dried blood spot specimens from newborn screening, collected in 2004 in North Thames and anonymously linked to birth registration data, were tested for maternally acquired rubella IgG antibody as a proxy for maternal antibody concentration using an enzyme-linked immunosorbent assay. Finite mixture regression models were fitted to the antibody concentrations from 1964 specimens. The Bayesian Information Criterion (BIC) was used as a model selection criterion to avoid over-fitting the number of mixture model components. This allowed investigation of the independent effect of maternal age

Placental transfer of tetanus toxoid antibodies in Nigerian mothers.

The aim of this study was to assess the efficacy of the tetanus toxoid immunization programme in Nigeria, specifically the placental transfer of antibody to newborn Nigerian babies. Tetanus toxoid antibody levels were measured in 39 mother-baby pairs in Ibadan, Nigeria and compared with 78 British mother-baby pairs. Geometric means of the ratios of cord/mother (sequestration index SI) were 0.776 for Nigerian pairs and 1.306 for British pairs, indicating a limitation in the placental transfer of tetanus toxoid in the Nigerian population. These findings confirm that there is a block in the place
