Loss of the glutamate transporter splice-variant GLT-1b in inferior colliculus and its prevention by ceftriaxone in thiamine deficiency.

Downregulation of astrocytic glutamate transporters is a feature of thiamine deficiency (TD), the underlying cause of Wernicke's encephalopathy, and plays a major role in its pathophysiology. Recent investigations suggest that ceftriaxone, a β-lactam antibiotic, stimulates GLT-1 expression and confers neuroprotection against ischemic and motor neuron degeneration. Thus, ceftriaxone treatment may be a protective strategy against excitotoxic conditions. In the present study, we examined the effects of ceftriaxone on the glutamate transporter splice-variant GLT-1b in rats with TD and in cultured

Calcifying pseudoneoplasm of the inferior colliculus: an unusual location for a rare tumor: case report.

OBJECTIVE: Quadrigeminal plate lesions are rare and usually present with a silent clinical course. Tumors, vascular lesions, inflammatory and infectious processes have been described in this region. Calcifying pseudoneoplasms, also reported as fibro-osseous lesions, cerebral calculi, and brain stones, are unusual lesions in the central nervous system. They can be revealed by cranial radiography, computed tomography, and magnetic resonance imaging as calcified masses and should be differentiated from neoplastic, inflammatory, and vascular lesions. To the best of our knowledge, the occurrence of

Age-related structural and functional changes in the cochlear nucleus.

Presbycusis - age-related hearing loss - is a key communication disorder and chronic medical condition of our aged population. The cochlear nucleus is the major site of projections from the auditory portion of the inner ear. Relative to other levels of the peripheral and central auditory systems, relatively few studies have been conducted examining age-related changes in the cochlear nucleus. The neurophysiological investigations suggest declines in glycine-mediated inhibition, reflected in increased firing rates in cochlear nucleus neurons from old animals relative to young adults. Responses of young and aged rat inferior colliculus neurons to sinusoidally amplitude modulated stimuli.

The inferior colliculus (IC) is a processing center for monaural and binaural auditory signals. Many units in the central nucleus of the inferior colliculus (CIC) respond to amplitude and frequency modulated tones, features found in communication signals. The present study examined potential effects of age on responses to sinusoidally amplitude modulated (SAM) tones in CIC and external cortex of the inferior colliculus (ECIC) units in young and aged F344 rats. Extracellular recordings from 154 localized single units of aged (24 month) rats were compared to recordings from 135 IC units from you

[Effects of lidocaine on salicylate-induced discharge of auditory neurons in the inferior colliculus of the guinea pig].

The effects of lidocaine (a local anesthetic known to relieve tinnitus) in a guinea pig animal model of tinnitus in which spontaneous discharge of inferior colliculus (IC) neurons was augmented by intravenous application of salicylate (200 mg/kg) were studied by extracellular recording. The salicylate induced discharge was inhibited by intravenous injection of lidocaine at a concentration (lmg/kg) used clinically for treating tinnitus. IC neurons could be divided into two groups according to the difference in sensitivity to lidocaine, weakly sensitive neurons and highly sensitive neurons. In w

Central auditory aging: GABA changes in the inferior colliculus.

Age-related hearing loss (presbycusis) is a complex state that reflects pathologic changes along the entire auditory neuraxis. Loss of speech understanding, decreased ability to localize sounds, and a decreased ability to detect and extract signals in noise are characteristic problems encountered by the elderly. Central (neural) presbycusis frequently results in a dramatic loss in speech understanding without a parallel change in pure-tone thresholds. In spite of evidence that suggests these deficits cannot be fully explained by peripheral changes alone, few studies have examined the neurochem

Salicylate-induced abnormal activity in the inferior colliculus of rats.

The evaluation of the spontaneous activity of 471 units from the external nucleus of the IC revealed that salicylate induces an increase of the spontaneous activity and the emergence of a bursting type of activity longer than 4 spikes. For sharply tuned units, the affected cells were from the frequency range of 10-16 kHz, which corresponds to the behaviorally measured pitch of salicylate-induced tinnitus in rats. An exogenous calcium supplement, provided under the conditions shown to attenuate the behavioral manifestation of salicylate-induced tinnitus, abolished the modification of the sponta

Temporal integration of electrical stimulation of auditory nuclei in normal-hearing and hearing-impaired cat.

Temporal integration functions were measured, before and after a sound-induced hearing loss, in 5 cats using trains of electrical pulses applied to auditory nuclei in the brainstem. The 8 stimuli ranged from 1 pulse (0.25 ms duration) to 16 pulses (0.25 ms pulses spaced over 240 ms). The stimuli were applied to inferior colliculus or cochlear nucleus via permanently implanted electrodes. One electrode was tested extensively in each animal to obtain 10 sets of behaviorally-measured electrical detection thresholds counterbalanced across stimuli. The animal was then exposed to a 110 dB SPL, 2 kHz

Visually evoked activity and variable modulation of auditory responses in the macaque inferior colliculus.

How multisensory cues affect processing in early sensory brain areas is not well understood. The inferior colliculus (IC) is an early auditory structure that is visually responsive (Porter KK, Metzger RR, Groh JM. Proc Natl Acad Sci USA 104: 17855-17860, 2007; Bulkin DA, Groh JM. Front Neural Circuits 6: 61, 2012; Bulkin DA, Groh JM. J Neurophysiol 107: 785-795, 2012), but little is known about how visual signals affect the IC's auditory representation. We explored how visual cues affect both spiking and local field potential (LFP) activity in the IC of two monkeys performing a task involving

Changes in the Proteomic Profile After Audiogenic Kindling in the Inferior Colliculus of the GASH/Sal Model of Epilepsy.

Epilepsy is a multifaceted neurological disorder characterized by recurrent seizures and associated with molecular and immune alterations in key brain regions. The GASH/Sal (Genetic Audiogenic Seizure Hamster, Salamanca), a genetic model for audiogenic epilepsy, provides a powerful tool to study seizure mechanisms and resistance in predisposed individuals. This study investigates the proteomic and immune responses triggered by audiogenic kindling in the inferior colliculus, comparing non-responder animals exhibiting reduced seizure severity following repeated stimulation versus GASH/Sal naïve

Ultrastructural Synaptic Differences in the Central Inferior Colliculus in the 3xTG Mouse Across Three Disease Stages.

Numerous studies support a mechanistic link between hearing loss and increased risks of cognitive decline and dementia. Hearing loss is widely viewed as a modifiable risk factor for Alzheimer's disease (AD). During normal aging the inferior colliculus (IC), a large auditory midbrain nucleus, undergoes numerous changes to neurotransmission, and these changes contribute to the development of central gain and presbycusis. Recent reports also implicate the IC as a nucleus that undergoes processing changes during AD. We used transmission electron microscopy (EM) to examine the synaptic ultrastructu

Crossed Brainstem Syndrome of the Tectal Plate's Inferior Colliculus Revealing Quadrigeminal Midbrain Cavernous Malformation.

Cavernous malformations are low-flow fragile vascular lesions prone to extralesional bleeding that can occur in the cerebral hemispheres, the brainstem, or the spinal cord. This paper reports the case of a 32-year-old right-handed man with acute-onset headaches associated with right-sided tinnitus, right-sided hemianesthesia, and binocular diplopia related to cranial nerve IV palsy. Neuroimaging displayed left-sided isolated cavernous malformation of the inferior tectal plate, with evidence of extralesional bleeding. Clinical presentation and neuroimaging features were compatible with inferior

The endogenous opioid system modulates defensive behavior evoked by Crotalus durissus terrificus: Panicolytic-like effect of intracollicular non-selective opioid receptors blockade.

BACKGROUND: There is a controversy regarding the key role played by opioid peptide neurotransmission in the modulation of panic-attack-related responses. AIMS: Using a prey versus rattlesnakes paradigm, the present work investigated the involvement of the endogenous opioid peptide-mediated system of the inferior colliculus in the modulation of panic attack-related responses. METHODS: Wistar rats were pretreated with intracollicular administration of either physiological saline or naloxone at different concentrations and confronted with rattlesnakes ( Crotalus durissus terrificus). The prey v

Behavioral consequences of unilateral inferior colliculus lesions in the rat.

This study was carried out to determine the behavioral sensitivity to sound of rats with unilateral lesions of inferior colliculus (IC) located ipsilateral or contralateral to the projection pathway from one ear. Absolute thresholds for the detection of a broad-band noise burst were compared for rats with a profound conductive hearing loss in one ear and a lesion placed either ipsilateral or contralateral to the normally functioning ear. The rats were trained to make withdrawal responses to avoid a shock when they detected the presence of a noise burst. Sound pressure level was systematically

Reversible conductive hearing loss: restored activity in the central auditory system.

The effect of a reversible, unilateral hearing loss on 2-deoxyglucose (2-DG) uptake in the central auditory system was studied using young gerbils. All animals had a unilateral conductive hearing loss (CHL), induced by atresia, on postnatal day 21 (P21). One week later, on P28, animals had their atresia repaired (CHL/R), or not repaired (CHL/NR), and CHL/NR animals entered the 2-DG experiments. CHL/R animals were allowed a 1-week period of restored binaural hearing experience prior to entering 2-DG experiments on P35. Animals in each group were injected with 2-DG and exposed to ambient sounds

The auditory system in methyl mercurial intoxication: a neuropathological investigation on 14 autopsy cases in Niigata, Japan.

The auditory system in 14 autopsy cases of methyl mercurial intoxication in Niigata was examined neuropathologically. In addition to degeneration of the transverse temporal gyrus, there was also either significant loss of small myelinated fibers or decrease of large neurons in the cochlear nerve, ventral cochlear nucleus and inferior colliculus. The degree of decrease was higher in acute patients than in the chronic, and was not correlative to the severity of cerebrovascular sclerosis. The hearing impairment in methyl mercurial intoxication may be induced by the combined degeneration of the ne

Transient CD15 expression reflects stages of differentiation and maturation in the human subcortical central auditory pathway.

The expression of the terminal saccharide determinant CD15 (3[a1-3]-fucosyl-N-acetyl-lactosamine) was evaluated in the central auditory system of the human developing brain by using monoclonal antibodies against this epitope. CD15 immunoreactivity was first observed in the ventral cochlear nucleus at 10 weeks of gestation, whereas the dorsal cochlear nucleus became positive from 13 weeks of gestation. In both nuclei, the intensity of immunoreactivity increased until 16 weeks of gestation and lasted until 25 weeks of gestation. In the inferior colliculi, CD15 was poorly expressed in the central…
