The permeabilizing effect of P2X(7) agonists was tested in rat submandibular acinar cells using the uptake of ethidium bromide as an index. A palladium-catalyzed tandem Heck cyclization/lactonization reaction has been developed for the efficient construction of functionalized isocoumarins. This tandem reaction forms two carbon-oxygen bonds and two carbon-carbon bonds in a single operation. The mechanism of this reaction involves sequential intramolecular Heck cyclization, alkyne coordination, and nucleophilic attack by oxygen. Mechanistic studies indicate that water or the base serves as oxygen source and that the aryl iodide substrate plays a dual role as both a coupling partner and an oxidant.

Natural lactones represent a promising anti-virulence strategy against Pseudomonas aeruginosa by targeting quorum-sensing (QS) pathways. Overdoses of macrocyclic lactones in dogs and cats can result in such signs as tremors, ataxia, seizures, coma, and blindness. Dogs with the ABCB1-1Δ gene defect are predisposed to macrocyclic lactone toxicosis at lower dosages than dogs without the defect. Intravenous lipid emulsion therapy has been suggested for treatment of macrocyclic lactone toxicosis but evidence of efficacy is limited. Initial decontamination and supportive care remain the mainstays of therapy for macrocyclic lactone toxicosis.

Cu/nitroxyl catalysts have been identified that promote highly efficient and selective aerobic oxidative lactonization of diols under mild reaction conditions using ambient air as the oxidant.
