Leukocyte motility in acne conglobata.

Leukocytes derived from the peripheral blood of 10 patients with acne conglobata demonstrated enhanced chemotactic and random migratory activities in comparison with leukocytes from 10 healthy subjects. This enhancement is most probably secondary to the state of chronic infection and inflammation. These findings probably rule out a defect in leukocyte motility as a cause of the severity of the lesions in acne conglobata.

Controlling leukocyte trafficking in IBD.

Inflammatory bowel disease (IBD) is characterized by the accumulation of immune cells, myeloid cells and lymphocytes in the inflamed intestine. The presence and persistence of these cells, together with the production of pro-inflammatory mediators, perpetuate intestinal inflammation in both ulcerative colitis and Crohn's disease. Thus, blockade of leukocyte migration to the intestine is a main strategy used to control the disease and alleviate symptoms. Vedolizumab is the only anti-integrin drug approved for the treatment of IBD but several other drugs also targeting integrins, chemokines or r

Leukocyte Imaging of the Diabetic Foot.

BACKGROUND: Diagnosing diabetic foot infection is often difficult, despite several available diagnostic methods. Amongst these, several imaging modalities exist to evaluate the diabetic foot in case of a suspected osteomyelitis. Nuclear Medicine, in particular, offers a variety of radiopharmaceuticals and techniques. Nowadays the gold standard radionuclide procedure, when an osteomyelitis is suspected, is represented by the use of radiolabelled leukocytes with either 99mTc-HMPAO or 111In-oxine. METHODS: In this review, we describe the correct acquisition and interpretation of white blood cell…

Platelet-leukocyte interactions in thrombosis.

Activated platelets may adhere to leukocytes and form circulating mixed aggregates. The latter are considered a reliable marker of a prothrombotic state and are associated with several cardiovascular conditions. The molecular mechanisms responsible of this cellular interaction include a central role of platelet P-selectin and of P-selectin glycoprotein ligand-1 (PSGL-1), its counter receptor on leukocytes in a signaling cascade, resulting in the activation of the beta-2 integrin Mac-1 and in the firm adhesion between the two cell types. The interaction of P-selectin with PSGL-1 also induces up…

Leukocyte trafficking in primary immunodeficiencies.

Leukocyte recirculation through central and peripheral lymphoid organs and peripheral tissues is essential to maintain immune homeostasis. Some of the genetically determined, primary immunodeficiencies compromise leukocyte trafficking. Here, we review the mechanisms and consequences of impaired leukocyte trafficking in leukocyte adhesion-deficiency syndromes, Warts-Hypo-gamma-globulinemia-Infections-Myelokathexis syndrome and Wiskott-Aldrich syndrome.

[Human leukocyte antigen and otosclerosis].

Fifty patients (32 women, 18 men) who underwent stapedectomy [FISCH, V. (1), 1982] for otosclerosis were studied. Histo-compatibility antigens were typed in all patients using the microlymphocytic test to determine antigens of the HLA system loci A and B. Statistical comparison of results with those obtained in a control group of 339 persons without otosclerosis showed that otosclerosis has a genetic HLA-related component.

[Leukocyte adhesion molecules and their abnormality].

A brief commentary on leukocyte adhesion molecules, including integrin family, immunoglobulin superfamily and selectin or LEC-CAM family is made. A recent concept on the molecular mechanisms of the leukocyte adhesion to cell surface is also described. An immunodeficiency disease called "leukocyte adhesion deficiency (LAD)" that is defective in expression of leukocyte adhesion molecules of integrin family, LFA-1, Mac-1 and p150, 95, is discussed with special reference to its clinical features, laboratory findings, neutrophil functions, lymphocyte functions and molecular defects. Some abnormal e

Automated differential leukocyte counts.

Automated differential counts have the advantage of precision, efficiency, safety, and economy. They could potentially serve effectively in 90 percent of patients with normal counts or in 75 percent of patients with anemia only (64 percent of the total in this study). Even patients with increased white blood cell counts and major population shifts (toward granulocytes or lymphocytes) could be followed with automated differential counts. Such a tactic would decrease turnaround time for results, be less expensive, and reduce exposure of technologists to direct contact with patients' blood. Howev

Leukocyte survival in cerebrospinal fluid.

Delays in the laboratory examination of cerebrospinal fluid are commonly encountered in clinical medicine. The present studies were designed to evaluate changes in cerebrospinal fluid leukocyte counts relative to time elapsed before analysis. Neutrophil counts decreased most rapidly, being 68 +/- 10% (standard error of the mean) and 50 +/- 12% of initial values at 1 and 2 h, respectively. Lymphocyte and monocyte numbers were not significantly altered until 3 h.

Automated leukocyte differential counters.

The leukocyte or white blood cell (WBC) differential count has been an established laboratory examination for almost a century. In recent years, the number of clinicians' requests for the test has grown steadily, and presently over 1,000,000 WBC differentials are performed daily in the United States. The manual WBC differential count requires tedious and repetitious cell classification, which is prone to subjective bias. Quite understandably, the WBC differential count has been a prime candidate for automation. Manufacturers have developed automated WBC differential counters that are accurate
