Spastin mutations impair coordination between lipid droplet dispersion and reticulum.

Lipid droplets (LD) are affected in multiple human disorders. These highly dynamic organelles are involved in many cellular roles. While their intracellular dispersion is crucial to ensure their function and other organelles-contact, underlying mechanisms are still unclear. Here we show that Spastin, one of the major proteins involved in Hereditary Spastic Paraplegia (HSP), controls LD dispersion. Spastin depletion in zebrafish affects metabolic properties and organelle dynamics. These functions are ensured by a conserved complex set of splice variants. M1 isoforms determine LD dispersion in t…

Microtubule-dependent and independent roles of spastin in lipid droplet dispersion and biogenesis.

Lipid droplets (LDs) are metabolic organelles that store neutral lipids and dynamically respond to changes in energy availability by accumulating or mobilizing triacylglycerols (TAGs). How the plastic behavior of LDs is regulated is poorly understood. Hereditary spastic paraplegia is a central motor axonopathy predominantly caused by mutations in SPAST, encoding the microtubule-severing protein spastin. The spastin-M1 isoform localizes to nascent LDs in mammalian cells; however, the mechanistic significance of this targeting is not fully explained. Here, we show that tightly controlled levels …

The coccidian parasites Toxoplasma and Neospora dysregulate mammalian lipid droplet biogenesis.

Upon infection, the intracellular parasite Toxoplasma gondii co-opts critical functions of its host cell to avoid immune clearance and gain access to nutritional resources. One route by which Toxoplasma co-opts its host cell is through hijacking host organelles, many of which have roles in immunomodulation. Here we demonstrate that Toxoplasma infection results in increased biogenesis of host lipid droplets through rewiring of multiple components of host neutral lipid metabolism. These metabolic changes cause increased responsiveness of host cells to free fatty acid, leading to a radical increa…

Genome-wide screens for gene products regulating lipid droplet dynamics.

Lipid droplets (LDs) are emerging as dynamic cellular organelles that play a key role in lipid and membrane homeostasis. Abnormal lipid droplet dynamics are associated with the pathophysiology of many metabolic diseases, such as obesity, diabetes, atherosclerosis, fatty liver, and even cancer. Understanding the molecular mechanisms governing the dynamics of LDs, namely, their biogenesis, growth, maintenance, and degradation, will not only shed light on the cellular functions of LDs, but also provide additional clues to treatment of metabolic diseases. Genome-wide screen is a powerful approach …

A Polarity-Sensitive FLIM Probe With Large Lifetime Dynamic Range for Visualizing Intracellular Lipid Droplets.

Fluorescence lifetime imaging microscopy (FLIM) is a powerful tool for probing intracellular microenvironments, but successful polarity imaging relies on probes that combine high organelle specificity, pronounced lifetime responses, and sufficient sensitivity to detect subtle polarity variations in lipid droplets (LDs). In this study, we report BTDO-MEA, a fluorescent probe with a donor-acceptor structure for high-resolution imaging of intracellular LD polarity. BTDO-MEA shows excellent lipophilicity, a large Stokes shift, and a large linear fluorescence lifetime response to environmental pola…

Resilient Membranized Coacervates Formed through Spontaneous Wrapping of Heat-Destabilized Lipid Bilayers around Coacervate Droplets.

Membranes and the membraneless biocondensates help organize cells and work synergistically to drive cellular processes. Separately, membrane-bound and membraneless compartments face difficulties as stable protocells or synthetic cell systems. Here, we present a new method to create membranized coacervates (MCs) for coacervates with any surface charge and a wide range of phospholipid membrane compositions. MCs are formed when liposomes, destabilized using heat and divalent ions, are mixed with coacervate dispersions. Unlike previous reports of hybrid coacervates surrounded by membranes, the MC

Astrocytic lipid droplets contain MHCII and may act as cogs in the antigen presentation machinery.

Lipid droplets (LDs) are crucial for energy homeostasis, but are also involved in a wide spectrum of other cellular processes. Accumulating data identifies LDs as an important player in inflammation. However, the underlying mechanisms and the impact of LDs on neuroinflammation remain unclear. Here, we describe a novel function of LDs in human astrocytes, in the context of Alzheimer's disease (AD). Although, the overall lipid profile was unchanged in astrocytes with AD pathology, our data show a clear effect on LD metabolism and specific fatty acids involved in neuroinflammation. Importantly, w…

Lipid droplets-related Perilipin-3: potential immune checkpoint and oncogene in oral squamous cell carcinoma.

BACKGROUND: Lipid droplets (LDs) as major lipid storage organelles are recently reported to be innate immune hubs. Perilipin-3 (PLIN3) is indispensable for the formation and accumulation of LDs. Since cancer patients show dysregulated lipid metabolism, we aimed to elaborate the role of LDs-related PLIN3 in oral squamous cell carcinoma (OSCC). METHODS: PLIN3 expression patterns (n = 87), its immune-related landscape (n = 74) and association with B7-H2 (n = 51) were assessed by immunohistochemistry and flow cytometry. Real-time PCR, Western blot, Oil Red O assay, immunofluorescence, migration a…

sEV-mediated lipid droplets transferred from bone marrow adipocytes promote ferroptosis and impair osteoblast function.

Osteoporosis is linked to increased bone marrow adipocyte (BMAd) proliferation, which displaces bone-forming cells and alters the local environment. The impact of BMAd lipid droplets on bone health and osteoblast function remains unclear. This study investigates the interplay between BMAd-derived lipid droplets and osteoblast functionality, focusing on ferroptosis pathways. Osteoblast cultures were treated with conditioned media from adipocytes to simulate in vivo conditions. High-throughput mRNA sequencing and Western blot analysis were used to profile changes in gene expression and protein l…

Expression of hepatic lipid droplets is decreased in the nitrofen model of congenital diaphragmatic hernia.

BACKGROUND: Prenatal mortality in newborn infants with congenital diaphragmatic hernia (CDH) has been attributed to increased amounts of liver hernia ion through the diaphragmatic defect. Antenatal studies in human and rodent fetus with CDH further demonstrated a contribution of the developing liver in the pathogenesis of CDH. The abnormal hepatic growth in experimental animal models, therefore, indicates a disruption of normal liver development in CDH. However, the underlying structural, histological and functional changes in the liver of animals with CDH remain unclear. We design this study

Effects of intracranial surgery on pineal lipid droplets, on other structures, and on melatonin secretion.

Unique effects of sham-pinealectomy [intracranial surgery (IS)] which include reduced functional activity of the adrenal gland and suppressed circadian rhythms of the adrenal medulla, and which are reversed by pinealectomy, have been reported in rodents. To clarify the mechanisms, we investigated whether or what changes occur in pineal functional activity after IS. Sixty-six male rats of normal and IS groups were used at 50 days of age. The pineal gland was first examined by quantitative electron microscopy. The Sudan III-stained lipid droplet content of the pinealocytes and plasma melatonin l

Cathodoluminescence-emitting lipid droplets in rat testis: a study by analytical color fluorescence electron microscopy.

Cathodoluminescence (CL) from lipid droplets (LDs) in the rat testis was examined by analytical color fluorescence electron microscopy. The results show that: (1) the CL at wavelengths of 320 nm (CL320) and 450 nm (CL450) is derived from cholesterol esters and a mixture of lipids including vitamin A esters, respectively; (2) CL320 in the LDs of Leydig cells sharply decreases on postnatal day 21, while CL320 and CL450 in the LDs of Sertoli cells begin to be detectable; (3) the CL450-emitting LDs in seminiferous tubules, whose distributional patterns display cyclic changes during the spermatogen…

Modulation of lipid droplets by Mycobacterium leprae in Schwann cells: a putative mechanism for host lipid acquisition and bacterial survival in phagosomes.

The predilection of Mycobacterium leprae (ML) for Schwann cells (SCs) leads to peripheral neuropathy, a major concern in leprosy. Highly infected SCs in lepromatous leprosy nerves show a foamy, lipid-laden appearance; but the origin and nature of these lipids, as well as their role in leprosy, have remained unclear. The data presented show that ML has a pronounced effect on host-cell lipid homeostasis through regulation of lipid droplet (lipid bodies, LD) biogenesis and intracellular distribution. Electron microscopy and immunohistochemical analysis of lepromatous leprosy nerves for adipose di

Acetylated PLIN2 coordinates the innate immune system and oxylipin synthesis on lipid droplets to propagate inflammation.

Research has implicated lipid droplets (LDs) and their associated proteins in inflammation and the innate immune response. The key LD protein Perilipin 2 (PLIN2) is tightly correlated with lipid storage and inflammation, but little is known about the mechanisms underlying its proinflammatory effects. Here, we show that PLIN2 is required for the inflammatory response to mitogens and the recruitment of pro-inflammatory proteins to the surface of LDs. PLIN2 directly binds arachidonic acid and interacts with key oxylipin synthetic enzymes such as PLA2 and COX2 to promote LD-localized oxylipin synt…

Protocol for isolation of lipid droplets and mitochondria from acute myeloid leukemia cells for lipidomic analysis.

Lipid droplets (LDs) are dynamic organelles critical for lipid and energy homeostasis. LD dysregulation is implicated in various diseases, including acute myeloid leukemia (AML), which exhibits aberrant lipid metabolism and chemoresistance. Here, we describe an effective protocol for the co-isolation of LDs and mitochondria from lipid-deficient cell types, e.g., AML cells. We detail procedures for the isolation, quality verification, and results from mass spectrometry-based lipidomic analysis. This protocol enables the functional study of organellar lipidomic regulation in AML.

The immunity-related GTPase IRGC mediates interaction between lipid droplets and mitochondria to facilitate sperm motility.

The immunity-related GTPases (IRGs) belong to the interferon-inducible GTPase protein family, which mediates cell autonomous and innate immunity response to intracellular pathogens. Yet, the cellular and physiological function of IRGC, a member of the IRG subfamily, has not been elucidated. Here, we show that testis-specific IRGC is specifically and highly expressed in mature spermatozoa and is required for sperm motility. IRGC induction results in the clustering of lipid droplets and initiation of their physical contact with mitochondria. When examining clinical semen samples, IRGC expression…
