Effects of 13-cis-retinoic acid on hindbrain and craniofacial morphogenesis in long-tailed macaques (Macaca fascicularis).

Hindbrain and craniofacial development during early organogenesis was studied in normal and retinoic acid-exposed Macaca fascicularis embryos. 13-cis-retinoic acid impaired hindbrain segmentation as evidenced by compression of rhombomeres 1 to 5. Immunolocalization with the Hoxb-1 gene product along with quantitative measurements demonstrated that rhombomere 4 was particularly vulnerable to size reduction. Accompanying malformations of cranial neural crest cell migration patterns involved reduction and/or delay in pre- and post-otic placode crest cell populations that contribute to the pharyng…

Effects of chronic caffeine consumption in pregnant monkeys (Macaca fascicularis) on blood and urine clinical chemistry parameters.

Pregnancy requires a variety of physiological adaptations to create an environment for the optimal development of the fetus. The widespread consumption of the methylxanthines especially caffeine and to a lesser extent theophylline by pregnant women suggests that it is important to determine whether these methylxanthines may influence maternal physiology during pregnancy. Forty female monkeys (Macaca fascicularis), randomly divided into three groups, were exposed to caffeine in their drinking water (0, 0.15, or 0.35 mg/ml) before, during, and after pregnancy. This exposure resulted in a dose-re…

The Effects of the Fire Hose Square Knot Browser as a Foraging Enrichment Device on the Behavior of Captive Macaca fascicularis.

Effective management of captive M. fascicularis (long-tailed macaques) is crucial for maintaining high-quality research models, necessitating strategies to promote their welfare. This study evaluated the impact of a foraging enrichment device, the "fire hose square knot browser", on the behavior of 32 long-tailed macaques at the Primate Research Center of IPB University in Bogor, Indonesia. We observed and analyzed daily behaviors across various food types over 288 h using scan and instantaneous sampling methods. Statistical analyses, including ANOVA and Kruskal-Wallis tests, revealed signific

Use of operant performance to guide and evaluate medical treatment in an adult male cynomolgus macaque (Macaca fascicularis).

A 6-y-old male cynomolgus macaque presented with noticeable swelling of the left forearm and signs of discomfort, as indicated by nonuse of the arm even in a behavioral task that he previously had been well-motivated to perform. Examination under anesthesia revealed lacerations to the arm. Radiography of the forearm showed no fractures, indicating that the damage was limited to soft tissue. The daily operant behavioral session assessed the amount of force the monkey emitted when touching the screen with the affected arm and how long each touch was sustained. We then used these parameters (forc

Replacement of the condyle and ascending ramus by a modular endoprosthesis in Macaca fascicularis--part 1: a clinical and radiographic study.

PURPOSE: To evaluate the feasibility of replacing the condyle and ascending ramus with a novel modular endoprosthesis in Macaca fascicularis. MATERIALS AND METHODS: Eight male adult monkeys (Macaca fascicularis) were used in this study. The right condyle and ascending ramus posterior to the lower second molar were resected. An endoprosthesis consisting of 2 modules was inserted to replace the resected segment and fixed in place using polymethylmethacrylate bone cement (Palacos). Four monkeys were sacrificed at 3 months and another 4 at 6 months postoperatively. The operated side of the mandib…

Crypt architecture of tonsilla lingualis in the monkey, Macaca fascicularis. A correlated light- and scanning electron-microscopic study.

Crypts of the lingual tonsil were investigated in 10 male and female Macaca fascicularis by use of correlated light- and scanning-electron microscopy. Counting of crypt openings provided an estimate of the total number of respective crypto-lymphatic units, which were found to range from 20 to 39. Crypt openings appeared in three distinct morphological varieties, i.e. circular, oval or slit-like. Tonsillar units existed individually or were arranged in a rosary fashion below a slit-like mucosal fold serving as a common exit. Although the crypt epithelium was generally non-keratinized, individua…

Double-Chambered Right Ventricle and Intraventricular Thrombosis Mimicking Double-Chambered Right Ventricle in Cynomolgus Monkeys (Macaca fascicularis).

Reports of congenital heart disease in nonhuman primates are rare, and double-chambered right ventricle (DCRV), which is a rare congenital heart disease, in which an abnormal muscle bundle divides the right ventricle into 2 chambers (a proximal high-pressure chamber and a distal low-pressure chamber), has not been previously reported. We diagnosed DCRV antemortem in 2 monkeys bred at The Tsukuba Primate Research Center that presented with cardiac murmurs. Subsequent diagnostic evaluation confirmed DCRV in one case, with the other diagnosed with midventricular obstruction having a pathophysiolo

Comparison of Two Different Alfaxalone Concentrations Combined with Midazolam to Anesthetize Cynomolgus Macaques (Macaca fascicularis) for Plethysmography.

Plethysmography is employed in nonhuman primates (NHPs) to calculate respiratory minute volume and determine the exposure time required to deliver an aerosol at the target dose. Anesthetic drugs can impact breathing parameters like steady-state minute volume (SSMV) central to aerosol dosing. Alfaxalone-midazolam mixtures (AM) provide superior parameters for plethysmography in cynomolgus macaques. An obstacle to the use of AM is the volume required to anesthetize via intramuscular injection. A more concentrated formulation of alfaxalone will reduce injection volumes and refine AM protocols. The

Sudden death caused by Aeromonas hydrophila in a Crab-eating Macaque (Macaca fascicularis) associated with captivity stress.

Aeromonas hydrophila is a facultative anaerobic gram-negative bacterium regarded as an opportunistic pathogen in animals. A 17-year-old female crab-eating macaque (Macaca fascicularis) died after experiencing anorexia and depression for several days. The carcass was severely emaciated, and the sternum was exposed under subcutaneous lesions in the thorax. Many abnormal pathological lesions were found, including tracheal inflammation, pulmonary inflammatory emphysema, yellowish discoloration of the liver, enlargement of the gall bladder, necrosis of the heart, congested bilateral kidneys, and en…

Effect of Infant Presence on Social Networks of Sterilized and Intact Wild Female Balinese Macaques (Macaca fascicularis).

Contraception is increasingly used to control wild animal populations. However, as reproductive condition influences social interactions in primates, the absence of new offspring could influence the females' social integration. We studied two groups of wild macaques (Macaca fascicularis) including females recently sterilized in the Ubud Monkey Forest, Indonesia. We used social network analysis to examine female grooming and proximity networks and investigated the role of infant presence on social centrality and group connectivity, while controlling for the fertility status (sterilized N = 14, …

Detection of tuberculosis in cynomolgus macaques (Macaca fascicularis) using a supplementary Monkey Interferon Gamma Releasing Assay (mIGRA).

Cynomolgus monkeys (Macaca fascicularis; MF) are commonly used as nonhuman primate models for pharmaceutical product testing. In their habitat range, monkeys have close contact with humans, allowing the possibility of bidirectional transmission of tuberculosis (TB) between the two species. Although the intradermal tuberculin skin test (TST) is used for TB detection in MF, it has limitations. Herein, we established the mIGRA, combining human QuantiFERON-TB Gold-Plus and monkey IFN-γ ELISApro systems, and used it to investigate 39 captive MF who were cage-mates or lived in cages located near a m…

Morphometrics and pelage characterization of longtailed macaques (Macaca fascicularis) from Pulau Bintan, Indonesia; Singapore; and Southern Vietnam.

Cynomolgus (or longtailed) macaques (Macaca fascicularis) are used extensively as laboratory animals in biomedical research. Their use in Singapore, an emerging biomedical hub in Southeast Asia, is now increasing widely, with research subjects currently originating from Singapore, Vietnam, and Pulau Bintan, Indonesia. Limited data exist on the genetic and phenotypic polymorphisms and phylogenetic relationships of these groups, and the animals are used as research subjects without regard to potential differences or homogeneity. Here we characterize their phenotypes by using established primatol…

Postanesthesia death and suspected peracute endotoxic shock due to Pseudomonas putida in a cynomolgous macaque (Macaca fascicularis).

An adult male cynomolgous macaque (Macaca fascicularis) died suddenly after anesthesia for a positron emission tomography scan. Bacteriologic culture of the mucopurulent secretions recovered from the endotracheal tube yielded heavy growth of Pseudomonas putida, a known endotoxin producer. Histologically, the lungs had severe, diffuse perivascular edema and neutrophils marginating to the endothelium. The sudden death and the pathologic findings were consistent with peracute endotoxic shock. Numerous environmental swab specimens of the surgical suite and equipment were submitted for bacteriologi…

Cynomolgus monkeys (Macaca fascicularis) in preclinical immune function safety testing: development of a delayed-type hypersensitivity procedure.

A delayed-type hypersensitivity (DTH) test commonly used for humans was adapted for use with cynomolgus monkeys (Macaca fascicularis). Pilot experiments showed naive animals had poor response rates and inconsistent reactivity to the antigens. In an exploratory phase, it was determined that monkeys could be experimentally sensitized by immunization with commercially available antigens. Animals were then sensitized with various concentrations of diphtheria and tetanus toxoids, Candida, and Trichophyton in the dose-response phase. Antigens were injected intradermally (i.d.) 3 times over a 7-day p…

Immunohistochemical characterization of integrin alfa 6 in uterus and cervix of the cynomolgus monkeys (Macaca fascicularis) with MfPV3 infection.

BACKGROUND: Cervical cancer is an abnormal growth of cervical tissue epithelial cells due to persistent human papilloma virus (HPV) infection. Cynomolgus monkeys (Macaca fascicularis) can be naturally and spontaneously infected with M. fascicularis Papillomavirus Type 3 (MfPV3), a virus that is phylogenetically closely related to human oncogenic HPV (HPV-16 and HPV-34), and therefore a potentially beneficial for modeling HPV disease. This study aims to evaluate the expression of the integrin alpha 6 (ITGα6) receptor in cynomolgus monkeys spontaneously infected with MfPV3, which this receptor a…

Possible role of peripheral CD14low monocytes in the development of collagen-induced arthritis in cynomolgus monkeys (Macaca fascicularis).

The changes in levels of peripheral major lymphocyte subsets were monitored with 10 adult cynomolgus monkeys (5 females and 5 males) during the 9 weeks after immunization with chick type-II collagen in Freund's complete adjuvant. Three females and 3 males developed overt arthritis determined by swelling of small joints and increase of plasma alkaline phosphatase as well as C-reactive protein. An increase of CD16+ NK cells was observed in four non-arthritis-developed monkeys (two females and two males). There was no significant difference in the fluctuation pattern of CD4+ T cell, CD8+ T cell a

Ultrastructure of the primate carotid body: a morphometric study of the glomus cells and nerve endings in the monkey (Macaca fascicularis).

The carotid body of the monkey (Macaca fascicularis) was studied at both the light and electron microscopic levels in an effort to provide a detailed quantitative characterization of this chemoreceptor organ in the primate. Structurally, the monkey carotid body was organized into lobules of from three to eight glomus cells (in section) and their ensheathing supporting cells. Interspersed among the lobules was abundant connective tissue stroma, fibroblasts and mast cells. Fenestrated capillaries, small arterioles and venules also permeated the organ. Each supporting cell partially ensheathed ab…
