Peptidylarginine deiminase inhibitor treatment is associated with increased phage susceptibility in Erwinia amylovora.

Understanding the phage resistance mechanisms in Erwinia amylovora is crucial for improving phage therapy applications targeting this plant pathogen, which is responsible for fire blight disease in apple and pear orchards. This study investigated E. amylovora phage resistance in the presence of peptidylarginine deiminase (PAD) inhibitors, which have been found in other studies to prevent the conversion of arginine to citrulline as a post-translational modification that may initiate outer membrane vesical (OMV) formation. Infection of the E. amylovora Ea6-4 strain by ϕEa21-4 phages, in the pres

Patulin Inhibition of Specific Apple Microbiome Members Uncovers Hanseniaspora uvarum as a Potential Biocontrol Agent.

Penicillium expansum is a major postharvest pathogen of apples, causing loss in fruits through tissue damage, as well as in apple products due to contamination with the mycotoxin patulin. During infections, patulin is a cultivar-dependent virulence factor that facilitates apple lesion development. Patulin also has characterized antimicrobial activity and is important for inhibiting other competitive phytopathogens, but the role of this inhibitory activity has not been investigated in the context of the apple microbiome. In our current study, we isolated 68 apple microbiota and characterized th

[Construction of a high-yield platform for phlorizin based on Nicotiana benthamiana].

Phlorizin has antioxidant and anti-aging pharmacological activities, which render it a highly promising candidate for applications in functional foods and the pharmaceutical industry. However, the incomplete elucidation of phlorizin biosynthesis in plants has constrained its large-scale production and application. Lithocarpus litseifolius, which contains a notably higher level of phlorizin than most plant species, serves as an ideal model for identifying key enzymes involved in phlorizin biosynthesis. The genes MdDBR, MdCHS, and MdMYB10 from Malus domestica Borkh. and LlP2'GT from L. litseifol…

Identification of novel virulence factors in Erwinia amylovora through temporal transcriptomic analysis of infected apple flowers under field conditions.

The enterobacterial pathogen Erwinia amylovora uses multiple virulence-associated traits to cause fire blight, a devastating disease of apple and pear trees. Many virulence-associated phenotypes have been studied that are critical for virulence and pathogenicity. Despite the in vitro testing that has revealed how these systems are transcriptionally regulated, information on when and where in infected tissues these genes are being expressed is lacking. Here, we used a high-throughput sequencing approach to characterize the transcriptome of E. amylovora during disease progression on apple flower

Antifungal Activity of Saponins from the Fruit Pericarp of Sapindus mukorossi against Venturia inaequalis and Botrytis cinerea.

The antifungal activity of an aqueous extract (AE) and the solid fraction of a chloroform-methanol fruit pericarp extract (CME) of Sapindus mukorossi resolved in water was tested for the first time against Venturia inaequalis and Botrytis cinerea-two important fungal pathogens worldwide. In the greenhouse, a CME (1% vol/vol) spray significantly reduced V. inaequalis symptoms and sporulation (99%) on apple seedling leaves (P ≤ 0.05). In field trials, applications of AE (1% vol/vol) reduced the disease severity of B. cinerea on grape, on average, by 63%. Extracts were fractionated by high-perfor

Lipid transfer protein sensitization in an apple-allergic patient: a case report from northern Europe.

We describe a case of a woman who developed three separate episodes of urticaria and ana-phylaxis during exercise after consuming an apple, with immunological evidence that nonspecific lipid transfer proteins (LTP) may have been responsible for these reactions. LTP sensitivity can cause life-threatening allergies and anaphylaxis. Although LTP sensitization is common in Mediterranean countries, the frequency of knowledge and diagnoses is increasing in Europe. Despite the geographic differences, LTP allergy should be kept on sight when facing severe anaphylaxis after consuming LTP-included food.

Investigating the in-vitro and in-vivo flavour release from 21 fresh-cut apples.

In-vitro and in-vivo flavour release from 21 different apple cultivars was studied using proton transfer reaction time-of-flight mass spectrometry (PTR-ToF-MS) with a focus on the relationship between texture and volatile organic compound (VOC) emission. Generally, firm-juicy cultivars had a shorter time to first swallow (Tswal) and a higher number of swallows (Nswal), while softer-mealy cultivars had a longer Tswal and a lower Nswal. Firm-juicy cultivars containing high VOC concentrations had a short time to maximum intensity (Tmax) owing to a shorter Tswal and a higher Nswal as juice was rel

Growth of Listeria monocytogenes within a caramel-coated apple microenvironment.

UNLABELLED: A 2014 multistate listeriosis outbreak was linked to consumption of caramel-coated apples, an unexpected and previously unreported vehicle for Listeria monocytogenes. This outbreak was unanticipated because both the pH of apples (<4.0) and the water activity of the caramel coating (<0.80) are too low to support Listeria growth. In this study, Granny Smith apples were inoculated with approximately 4 log10 CFU of L. monocytogenes (a cocktail of serotype 4b strains associated with the outbreak) on each apple's skin, stem, and calyx. Half of the apples had sticks inserted into the core

Dynamic changes of the ethylene biosynthesis in 'Jonagold' apple.

In this study, the short-term and dynamic changes of the ethylene biosynthesis of Jonagold apple during and after application of controlled atmosphere (CA) storage conditions were quantified using a systems biology approach. Rapid responses to imposed temperature and atmospheric conditions were captured by continuous online photoacoustic ethylene measurements. Discrete destructive sampling was done to understand observed changes of ethylene biosynthesis at the transcriptional, translational and metabolic level. Application of the ethylene inhibitor 1-methylcyclopropene (1-MCP) allowed for the

Factors affecting appearance of stink bug (Hemiptera: Pentatomidae) injury on apple.

Three main types of stink bug injury have been reported on the surface of apple fruit: (1) discolored dots, (2) discolored dots with depressions, and (3) discolored dots with discolored depressions. From 2005 to 2008, studies were performed to determine the factors linked to the variations in the appearance of adult stink bug injury on apple fruit observed at harvest. The main factors tested were type of feeding injury (i.e., mechanical versus breakdown and removal of tissues), time between injury and evaluation, cultivar, fruit maturity, and relative exposure to sunlight and UV radiation. No

Cyanide Metabolism in Apple Embryos During Dormancy Breakage Is Modulated by NO or HCN Application.

Hydrogen cyanide (HCN) is an N-containing gasotransmitter with a regulatory function in plants and animals. Despite its toxicity, HCN acts as a signalling molecule in plants, influencing development, pathogen defence, and stress responses, with particular emphasis on biotic stress and seed dormancy/germination. Around 3000 species across > 130 families produce HCN from cyanogenic compounds. This study investigated how nitric oxide (NO) and HCN impact the activity of enzymes involved in HCN metabolism during dormancy breakage of apple (Malus domestica Borkh.) embryos. Based on earlier findings …

Light-emitting diode white light suppresses ethylene biosynthesis in apple fruit.

As a typical climacteric fruit, the apple (Malus domestica) ripening is predominantly regulated by ethylene. Light-emitting diode (LED) white light, an energy-efficient and widely promoted light source, can significantly inhibit ethylene biosynthesis during apple ripening; however, its underlying regulatory mechanism remains unclear. This study demonstrates that LED white light (LWL) inhibits ethylene production by enhancing spermidine (Spd) production, which competes with ethylene for the common precursor S-adenosylmethionine (SAM). Mechanistically, LWL induces the transcription factor ELONGA…

Seasonal Xylem Sap Acidification Is Governed by Tree Phenology, Temperature and Elevation of Growing Site.

pH of xylem sap (pHx) was determined in three trees (Malus domestica (apple tree), Picea abies and Pinus cembra) in response to seasonal changes. Conifer trees from lowland (600 m) were compared to trees growing at the alpine timberline (1950 m a.s.l.). Xylem sap was extracted with a Scholander pressure bomb and pHx was measured with a pH microsensor. In all species, pHx changed markedly with season. In spring, pHx was acidic; during winter, the pHx was more alkaline. In apple trees, the pHx did not show a significant correlation with temperature but was rather affected by developmental stage.…

The MdERF1B-miR482c-MdCCR4-MdTLP1b module regulates apple resistance to Colletotrichum gloeosporioides.

Glomerella leaf spot (GLS), incited by Colletotrichum gloeosporioides, poses a significant threat to apple (Malus domestica) production. microRNAs (miRNAs) are critical regulators of plant-pathogen crosstalk. However, the underlying mechanism through which miR482 regulates apple resistance to GLS remains unclear. Here, we show that overexpression of miR482c compromises resistance against C. gloeosporioides, while silencing miR482c confers resistance to C. gloeosporioides. 5'RACE assays in apple and co-expression assays in Nicotiana benthamiana confirmed that miR482c targets and cleaves the MdC…

Scaling of Floral Display and Mechanical/Hydraulic Support Across Twelve Crabapple Cultivars.

Flowers of the Rosaceae, with their distinct pedicels and well-differentiated floral organs, provide an ideal model for investigating biomass allocation trade-offs associated with the balance between reproductive investment and mechanical/hydraulic support. However, the scaling relationship between supportive structures (e.g., pedicels) and floral display structures (e.g., petals) remains largely unresolved. We examined 12 crabapple (Malus) cultivars, measuring their pedicel length, pedicel fresh mass, corolla fresh mass, corolla area, and total flower fresh mass. Scaling relationships among t…

Molecular insights and integrated management of Glomerella leaf spot in apple.

Apple (Malus × domestica) is one of the world's premier fruit crops. Apple Glomerella leaf spot (GLS) is caused by Colletotrichum spp., jeopardizing global apple supply and incurring millions of dollars in annual losses. The development of resistant apple varieties has evolved from conventional selection breeding to deliberate breeding strategies that incorporate statistical analysis and experimental design, and now increasingly integrate genetic and genomic data. Innovative methods like marker-assisted selection and genetic editing have facilitated the development of cultivars resistant to GL…

Behavioral evidence for fruit odor discrimination and sympatric host races of Rhagoletis pomonella flies in the Western United States.

The recent shift of Rhagoletis pomonella (Diptera: Tephritidae) from its native host downy hawthorn, Crataegus mollis, to introduced domesticated apple, Malus domestica, in the eastern United States is a model for sympatric host race formation. However, the fly is also present in the western United States, where it may have been introduced via infested apples within the last 60 years. In addition to apple, R. pomonella also infests two hawthorns in the West, one the native black hawthorn, C. douglasii, and the other the introduced English ornamental hawthorn, C. monogyna. Here, we test for beh…

Synergistic Light-Harvesting and Heat Transfer in Platinum-Confined Gold Nanocages for Sensitive Photothermal Lateral Flow Immunoassay.

Photothermal lateral flow immunoassay (LFIA) is a promising point-of-care testing platform renowned for its high sensitivity and quantitative capability. However, its development is constrained by the limited performance of available photothermal nanotags. Herein, platinum-confined gold nanocages (PANCs) were designed as efficient photothermal nanotags. The mesoporous structure of PANCs enhanced light-harvesting by prolonging the optical path while facilitating heat transfer via their high specific surface area. The confined platinum nanoparticles (PtNPs) played dual roles, including enhancing
