An outbreak of Brucella melitensis in a family was studied. From the fourteen family members who ate unpasteurized goat cheese nine became ill. Patients included four females and five males of 8 to 75 years old. In seven of the patients the diagnosis was confirmed by positive blood culture for B. melitensis biovar 1. A 50-year-old female, heterozygous for beta-thalassaemia was found to have a lytic lesion surrounded by osteosclerotic tissue in the 1st lumbar vertebra. The role of antibody and cell-mediated immunity in the resistance to Japanese encephalitis virus (JEV) infection was studied in adult mice. Passively transferred antibodies obtained up to 2 weeks after primary infection protected the recipient mice against a challenge infection with JEV. An automated assay of anti-P1 allohemagglutinins has been carried out on 133 P2 subjects: 13 with acute hepatic distomiasis (Fascioliasis), 20 with hydatidosis and 100 healthy blood donors. The effect of the application of a single oral dose of 100 mg of three mucolytic agents (N-acetylcysteine, carbocysteine and Na-2-mercaptoethane sulphonate) on the ultrastructure of the tracheal epithelium of healthy rabbits was studied. Due to the function of oral mucolytics the goblet cells were overstimulated and injured. Na-2-mercaptoethane sulphonate was the least irritating substance. Status asthmaticus in children is a life-threatening condition that calls for a comprehensive set of therapeutic measures, viz. evacuation of tracheobronchial secretions, parenteral drug administration and improvement of alveolar gas exchange. In the authors' experience, that therapeutic procedure can be much improved by bronchial lavage with mesna. This is illustrated by the good results obtained in fourteen patients with status asthmaticus and in another forty-four cases of global respiratory insufficiency. Antibody titers to common enterobacterial antigen (CA) were determined in 141 controls and in acute serum specimens from 206 patients with bacteremia caused by gram-negative organisms. Levels of antibody to CA ranged from 1:160 to 1:2,560 in 95% of control subjects. These levels did not differ significantly from those in acute serum specimens from bacteremic patients with "nonfatal underlying diseases." Patients with more severe underlying diseases, "ultimately fatal underlying diseases," tended to have lower titers of antibody to CA. Human antibody to CA was predominantly of the 19S variety.

Brucellosis is a significant health problem in countries where control of zoonoses is inadequate. During 1993-98, we analysed sera and cultures from 792 suspected brucellosis patients who presented with histories of fever, chills, night sweating, weakness, malaise and headache to the referral hospital in Yazd. Conventional indirect haemagglutination test was performed in rhesus monkey sera (collected from Plasmodium knowlesi infected animals) with and without prior treatment of sera with 2-mercapto-ethanol (2-ME). Surprisingly, many sera samples showed significant enhancement of final titre with 2-ME. The structural polypeptides of accepted species and recently isolated members of the genus Orthopoxvirus have been examined by SDS-polyacrylamide gel electrophoresis. The viruses shared many polypeptides but some differences were found. The viruses could be divided into a vaccinia group (including buffalopox, 'Lenny' and MK-10), an ectromelia group (including elephant virus and Moscow virus), cowpox, camelpox and monkeypox. DNA is widely used in plant genetic and molecular biology studies. In this chapter, we describe how to extract DNA from wheat tissues. The tissue samples are ground to disrupt the cell wall.
