Lgr5⁺ cells regulate small intestinal morphogenesis before villification.

Lgr5 marks both adult intestinal stem cells and embryonic intestinal stem/progenitor cells. However, the stemness properties and physiological roles of embryonic intestinal Lgr5⁺ cells prior to villification (PVLCs) remain largely unknown. In this study, we show that PVLCs in the embryonic small intestine exhibit region-specific stemness, with progressively enhanced stemness potential from the proximal to distal region. Through inducible cell ablation and gene knockout experiments, we demonstrate that PVLCs regulate small intestinal morphogenesis via Hedgehog signaling in a region-dependent ma…

Spatially Organized Human Ovarian Spheroids Instruct Endometrial Morphogenesis.

The human ovarian-endometrial axis operates through coordinated, dynamic endocrine signaling that cannot be faithfully reproduced by static hormone supplementation. While endometrial organoids (EMOs) respond to exogenous estradiol and progesterone, whether a spatially organized human ovarian endocrine unit can instruct epithelial morphogenesis through physiologically integrated signaling remains unknown. Here, we engineered a multilayered human follicle-like spheroid composed of primary granulosa cells and stromal-derived theca-like cells (SPHEGaT), recreating architectural and steroidogenic f…

Ctsk+ Osteoclasts Orchestrate Condylar Morphogenesis via Hypoxic Lysosome.

While the biological functions and molecular mechanisms of osteoclasts in condylar morphogenesis remain incompletely characterized, and the regulatory effects of hypoxic conditions within the deep condyle on osteoclast-mediated alveolar bone remodeling are poorly understood, this investigation systematically examines the hypoxia response mechanisms of cathepsin K-positive (ctsk+) osteoclasts and their specialized functions in condylar tissue organization. Genetic ablation of ctsk+ cells in diphtheria toxin receptor (DTR) transgenic mice resulted in pronounced accumulation of calcified cartilag…

Molecular Regulatory Mechanisms in Chicken Feather Follicle Morphogenesis.

In China, the sale of freshly slaughtered chickens is becoming increasingly popular in comparison with that of live chickens, and due to this emerging trend, the skin and feather follicle traits of yellow-feathered broilers have attracted a great deal of research attention. The feather follicle originates from the interaction between the epidermis and dermis in the early embryonic stage. Feather follicle morphogenesis is regulated by the Wnt, ectodysplasin (Eda), epidermal growth factor (EGF), fibroblast growth factor (FGF), bone morphogenetic protein (BMP), sonic hedgehog (Shh), Notch, and ot…

Nutritional control of thyroid morphogenesis through gastrointestinal hormones.

Developing animals absorb nutrients either through the placenta or from ingested food; however, the mechanisms by which embryos use external nutrients for individual organ morphogenesis remain to be elucidated. In this study, we assessed nutrient-dependent thyroid follicle morphogenesis in Xenopus laevis and investigated the role of secreted gastrointestinal (GI) hormones post-feeding. We found that feeding triggers thyroid follicle formation, and the thyroid cells showed transient inactivation of cell proliferation after feeding. In addition, the thyroid cells with multi-lumina were frequentl…

Hedgehog Signaling Pathway Orchestrates Human Lung Branching Morphogenesis.

The Hedgehog (HH) signaling pathway plays an essential role in mouse lung development. We hypothesize that the HH pathway is necessary for branching during human lung development and is impaired in pulmonary hypoplasia. Single-cell, bulk RNA-sequencing data, and human fetal lung tissues were analyzed to determine the spatiotemporal localization of HH pathway actors. Distal human lung segments were cultured in an air-liquid interface and treated with an SHH inhibitor (5E1) to determine the effect of HH inhibition on human lung branching, epithelial-mesenchymal markers, and associated signaling

Modelling Cellular Interactions and Dynamics During Kidney Morphogenesis.

Kidney disease and renal disorders account for a significant proportion of health complications in mid-late adulthood worldwide. Many renal deficiencies are due to improper formation of the kidneys before birth, which are caused by disorders in the developmental process that arise from genetic and/or environmental factors. Mathematical modelling can help build on experimental knowledge to increase our understanding of the complexities of kidney organogenesis. In this paper, we present a discrete cell-based model of kidney development. Specifically, we model the tip of the developing ureteric t

PDGFRα: Expression and Function during Mitral Valve Morphogenesis.

Mitral valve prolapse (MVP) is a common form of valve disease and can lead to serious secondary complications. The recent identification of MVP causal mutations in primary cilia-related genes has prompted the investigation of cilia-mediated mechanisms of disease inception. Here, we investigate the role of platelet-derived growth factor receptor-alpha (PDGFRα), a receptor known to be present on the primary cilium, during valve development using genetically modified mice, biochemical assays, and high-resolution microscopy. While PDGFRα is expressed throughout the ciliated valve interstitium earl

Understanding kidney morphogenesis to guide renal tissue regeneration.

The treatment of renal failure has seen little change in the past 70 years. Patients with end-stage renal disease (ESRD) are treated with renal replacement therapy, including dialysis or organ transplantation. The growing imbalance between the availability of donor organs and prevalence of ESRD is pushing an increasing number of patients to undergo dialysis. Although the prospect of new treatment options for patients through regenerative medicine has long been suggested, advances in the generation of human kidney cell types through the directed differentiation of human pluripotent stem cells o

Mouse Crumbs3 sustains epithelial tissue morphogenesis in vivo.

The human apical protein CRB3 (Crb3 in mouse) organizes epithelial cell polarity. Loss of CRB3 expression increases the tumorogenic potential of cultured epithelial cells and favors metastasis formation in nude mice. These data emphasize the need of in vivo models to study CRB3 functions. Here, we report the phenotypic analysis of a novel Crb3 knockout mouse model. Crb3-deficient newborn mice show improper clearance of airways, suffer from respiratory distress and display perinatal lethality. Crb3 is also essential to maintain apical membrane identity in kidney epithelial cells. Numerous kidne

Epithelial inactivation of Yy1 abrogates lung branching morphogenesis.

Yin Yang 1 (YY1) is a multifunctional zinc-finger-containing transcription factor that plays crucial roles in numerous biological processes … tions. No evidence for a potential genetic interplay between murine Dicer and Yy1 genes during lung morphogenesis was observed. However, the cystic lung phenotype resulting from the epithelial inactivation of Dicer function mimics the Yy1 lung malformations with similar changes in Shh and Fgf10 expression. Together, our data demonstrate the crucial requirement …

Btbd7 regulates epithelial cell dynamics and branching morphogenesis.

During embryonic development, many organs form by extensive branching of epithelia through the formation of clefts and buds. In cleft formation, buds are delineated by the conversion of epithelial cell-cell adhesions to cell-matrix adhesions, but the mechanisms of cleft formation are not clear. We have identified Btbd7 as a dynamic regulator of branching morphogenesis. Btbd7 provides a mechanistic link between the extracellular matrix and cleft propagation through its highly focal expression leading to local regulation of Snail2 (Slug), E-cadherin, and epithelial cell motility. Inhibition expe…

Xmc mediates Xctr1-independent morphogenesis in Xenopus laevis.

In the frog, Xenopus laevis, fibroblast growth factor (FGF) signaling is required for both mesoderm formation and the morphogenetic movements that drive the elongation of the notochord, a dorsal mesodermal derivative; the coordination of these distinct roles is mediated by the Xenopus Ctr1 (Xctr1) protein: maternal Xctr1 is required for mesodermal differentiation, while the subsequent loss of Xctr1 promotes morphogenesis. The signaling cascade activated by FGF in the presence of Ctr1 has been well characterized; however, the Xctr1-independent, FGF-responsive network remains poorly defined. We …

Convergent signalling through Fgfr2 regulates divergent craniofacial morphogenesis.

Fibroblast growth factor receptor 2 (Fgfr2) has two splice variants IIIb and IIIc, which are unique in function and localization. Signalling through Fgfr2IIIb controls epithelial-mesenchymal interactions, which regulate morphogenesis during the development of several organs including the palate and tooth. In this study, we confirm that molar tooth development in Fgfr2IIIb(-/-) mice is arrested early in development and that the molar teeth of Fgf10(-/-) mice develop through all the normal stages of morphogenesis. We show that the molar phenotype of Fgfr2IIIb(-/-) mice is, in part, owing to redu…

Branching Morphogenesis of Mouse Embryonic Submandibular Epithelia Cultured under Three Different Conditions: (mouse submandibular gland/epithelial branching/morphogenesis/collagenase/heparitinase/heparin/Matrigel).

To investigate how the mesenchyme interacts with the epithelium, we employed three different culture systems: System A, in which intact submandibular gland rudiments at the mid 13-day stage were cultured on Millipore filters; System B, in which the 13-day epithelium and mesenchyme were separated once with dispase, recombined again, and cultured on the filter; System C, in which the separated 13-day epithelium was clotted with Matrigel and cultured with the mesenchyme across the filter or in the presence of EGF instead of the mesenchyme. In Systems A and B, 13-day epithelia expanded and produce

Engineered Development: Directed Morphogenesis of an Embryonic Heart Tube.

Morphogenesis is a major developmental process that shapes organs in utero through coordinated structural deformations. Outside of simple cellular or animal models, limited models of morphogenesis exist due to the complexity of hierarchical structural change. In development, the embryonic heart undergoes a complex morphogenesis process involving looping, chamber growth, and septation, transforming the primordial tube to an adult four-chambered organ. An in vitro model of looping could provide unique insights into this phase of development and improve understanding of congenital defect generati…

HsfA-driven gene transcription regulates fungal morphogenesis and pathogenicity.

Aspergillus flavus, a ubiquitous filamentous fungus, severely compromises global food safety and public health by producing carcinogenic aflatoxins. Heat shock factor 1 (HSF1) orchestrates stress responses in eukaryotes, yet the functional role and regulatory mechanisms of its homolog, HsfA, in A. flavus remain elusive. Here, we demonstrate that A. flavus encodes two hsfA copies, whose knockdown or expression of a dominant-negative variant abrogates spore germination, a prerequisite for fungal development and colonization. Through integrated reverse genetics, ChIP-qPCR, electrophoretic mobilit

Hedgehog-Wnt Crosstalk Orchestrates Submandibular Gland Morphogenesis in Mice.

The development of the mouse submandibular gland (SMG) is a classic model for branching morphogenesis, orchestrated by key signaling pathways such as Hedgehog (Hh) and Wnt. While Sonic Hedgehog (Shh) is known to be essential for early SMG development, its downstream mechanisms and potential crosstalk with other pathways remain unclear. This study investigates the functional interaction between Hh and Wnt signaling during SMG morphogenesis. Using both in vivo (pharmacological inhibition in pregnant mice) and ex vivo (organ culture) models, we inhibited Hh signaling with vismodegib from embryoni…

Gonadal morphogenesis and sexual differentiation in rhea (Rhea americana).

The rhea (Rhea americana) is a docile and highly adaptable ratite with notable zootechnical potential for the production of meat, eggs, feathers, and leather. Beyond its economic relevance, captive breeding plays an important role in conservation. However, information on its embryonic and gonadal development remains limited-knowledge that is essential for improving reproductive management. This study aimed to characterize gonadal differentiation during artificial incubation, focusing on developmental stages at days 9, 12, 15, 18, 21, and 24. We provide a detailed morphological and cytological

Morphogenesis and morphometry of brain folding patterns across species.

Evolutionary adaptations associated with the formation of a folded cortex in many mammalian brains are thought to be a critical specialization associated with higher cognitive function. The dramatic surface expansion and highly convoluted folding of the cortex during early development is a theme with variations that suggest the need for a comparative study of cortical gyrification. Here, we use a combination of physical experiments using gels, computational morphogenesis, and geometric morphometrics to study the folding of brains across different species. Starting with magnetic resonance image…
