Some classical and nonclassical muscarinic agents were tested for their effects on potassium-evoked acetylcholine (ACh) release from rat neostriatal slices. Release was monitored by measuring endogenous ACh when acetylcholinesterase (AChE) was inhibited with physostigmine (30 microM) or by measuring endogenous choline when AChE activity was left intact. The classical antagonist, atropine (0.1-2 microM), induced an increase in release whether AChE activity was inhibited or intact. The putative M-1 selective antagonist, pirenzepine, had minimal effects over a broad concentration range (2-200 mic

We have previously proposed that the reciprocal hindlimb scratching (RHS) of mice elicited by intrathecal injection of muscarinic agents is mediated by M1 muscarinic receptors. In this study, benzhexol potently inhibited pilocarpine-induced RHS (ID50 = 0.5 ng), methoctramine did not fully block RHS and RS 86 neither produced nor blocked RHS. These data (1) differentiate the three muscarinic agents using this in vivo endpoint, (2) substantiate binding studies characterizing benzhexol and methoctramine as putative M2 and M2 antagonists, respectively, and (3) further support using RHS for studyin

Injection of bethanechol into the lateral cerebral ventricle of the rat induces a marked increase in drinking, within 30 min from administration. The response is dose-related, maximal water intake (6.1 +/- 0.55 mL; mean +/- s.e.) occurring at 10 micrograms of bethanechol. Peripheral administration of the agonist (up to 3 mg kg-1 i.p.) fails to elicit drinking. Among several specific antagonists only antimuscarinic drugs produced a significant inhibition of the response, suggesting that the compulsive drinking behaviour in the rat is caused by activation of central muscarinic receptors. The dri

The ability of chlorpromazine (CPZ) to inhibit muscarinic-cholinergic secretory events was studied in vitro in rat parotid acinar cells. CPZ inhibited carbachol-induced amylase release in a dose-dependent fashion but had no effect on that elicited by isoproterenol. The inhibition of parotid protein synthesis induced by carbachol, but not that induced by A23187, was blocked by CPZ. CPZ exhibited a dose-dependent inhibition of [3H] quinuclidinyl benzilate (QNB) binding to muscarinic receptors, and altered the KD of the receptor for the ligand. These results are consistent with an ability of CPZ

The effect of hydrogen ion concentration on ligand binding to muscarinic acetylcholine receptors was studied in membranes isolated from rat brainstem. The binding of [3H]methylscopolamine was constant between pH 7 and 10. The affinity, but not the number, of [3H]methylscopolamine binding sites decreased below pH 7; at pH 4 little binding was detected. When brainstem membranes were incubated at various pH levels from 3 to 11 for 1 h and then returned to pH 8, [3H]methylscopolamine binding affinity was restored to control levels. Carbamylcholine binding affinity was also depressed in media of lo

In microsomes of bovine adrenal medulla, there were two affinity binding sites for carbamylcholine with a high and a low affinity. Copper (1-10 microM) largely enhanced the affinity of carbamylcholine at the low affinity binding site, with a slight increase in the affinity at the high affinity binding site. On the other hand, copper slightly decreased the binding affinity of pirenzepine and atropine. Thus, low concentrations of copper modulate the muscarinic receptors in the adrenal medulla by selectively increasing agonist affinity.

In these experiments the effect on the cat's salivation of the choline ester, carbachol, the anticholinesterase, eserine, and the muscarinic ganglionic stimulants, McN-A-343 and AHR-602, injected into the cerebral ventricles was investigated and compared. Carbachol and eserine injected intracerebroventricularly (ICV) produced dose-related salivation. On the other hand, ICV McN-A-343 and AHR-602 evoked inconsistent salivation when injected in large doses. The antimuscarinic drug, atropine, injected into the cerebral ventricles abolished the salivation caused by carbachol and eserine similarly i

Postmortem human brain samples were taken from non-neurological controls as well as demented subjects who died with Alzheimer's disease (AD), multi-infarct dementia (MID), or a combination of AD and MID dementia (MIXED). Choline acetyltransferase (ChAT) activity was measured radiometrically using [1-14C]acetyl-coenzyme A as the substrate, muscarinic binding was assayed with [3H]quinuclidinyl benzilate, and the proportion of binding associated with high affinity agonist sites was measured by carbamylcholine displacement of the radioligand. Relative to control, ChAT activity was significantly re

Intense muscarinic stimulation of the monkey ciliary muscle causes long-lasting muscarinic subsensitivity. This could be due to changes in number or affinity of muscarinic receptors which would cause a threshold elevation detectable in vivo. Since plasma levels of the agonists causing contraction in vivo were not available, the accommodation response to systemic muscarinic agents in subsensitized eyes was compared with that in the normal fellow eyes, usually seven days after a single subsensitizing dose of 100 micrograms carbachol to cornea. Subsensitivity was present whether the agonist teste

In order to assess a possible role for copper as a regulator of muscarinic receptors in vitro, the receptor was characterized in rats made copper deficient by a dietary regimen. In forebrain regions there was a decrease in both the affinity of the receptors for [3H]-1-quinuclidinyl benzilate and the density of receptors in the copper-deficient animals compared with control animals. Copper treatment in vitro of homogenates from deficient animals did not reverse the in vivo effects on antagonist binding but, rather, decreased receptor occupancy and ligand affinity in a manner similar to copper t

The effects of trimethyl tin on passive and active avoidance behavior, hippocampal muscarinic receptors and hippocampal cell destruction were examined in male rats. The animals were intubated with 18 mumoles/kg (3.5 mg/kg) of TMT hydrochloride or vehicle. When tested two weeks later treated animals exhibited marked deficits in retention of passive avoidance and extinction of active avoidance tasks. Receptor binding analysis, using 3H-QNB, revealed a significant decrease (21%) in muscarinic receptor density in the hippocampus. Histological examination of the hippocampus revealed a concomitant l

The binding properties of (-)-[3H]quinuclidinyl benzilate and [3H] N-methylscopolamine to muscarinic acetylcholine receptors have been investigated in rat brain homogenates. The binding of both antagonists demonstrated high affinity and saturability. Analysis of the binding data resulted in linear Scatchard plots. However, (-)-[3H]quinuclidinyl benzilate showed a significantly higher maximal binding capacity than that of [3H]N-methylscopolamine. Displacement of both ligands with several muscarinic receptor antagonists resulted in competition curves in accordance with the law of mass-action for

Intracellular recordings were made from neurones in the myenteric plexus of the guinea-pig ileum in vitro. Muscarinic depolarizations were evoked by brief (1-500 ms) ionophoretic applications of acetylcholine (ACh) or other agonists. Nicotinic responses to ACh evoked by the same ionophoretic pulse had short latencies and rapid rise times, indicating close proximity of the ionophoretic pipette to the neurone membrane. The time course (duration several seconds) of the muscarinic depolarization was independent of the identity of the agonists applied (ACh, methacholine, carbachol, oxotremorine). H

The contracted segment of terminal large intestine in both Hirschsprung's disease in humans and in the piebald-lethal strain of mice with congenital megacolon may result from muscarinic cholinergic receptor denervation supersensitivity. This hypothesis was tested by direct binding studies using the muscarinic receptor ligand, tritiated quinuclidinyl benzilate. There were no differences in either the receptor density, dissociation constant, or agonist/antagonist Hill coefficients in the proximal, middle, or distal colon of mice with congenital megacolon as compared with normal littermates. The ligand binding properties of muscarinic receptors present in rat adenohypophysis were studied using the tritiated antagonists N-methyl-4-piperidyl benzilate, 3-quinuclidinyl benzilate, and methylscopolamine. Equilibrium analysis of antagonists binding as well as competition experiments with several agonists were used to probe the nature of binding sites. The nature of antagonist binding in the adenohypophysis points to heterogeneity of binding sites, in contrast to other brain regions, in which homogeneous populations of muscarinic receptors were detected. Some of the binding properties, e

1 Responses of single neurones in isolated superior cervical ganglia of the rat to muscarinic agonists were recorded with intracellular microelectrodes. 2 (+/-)-Muscarine (1 to 10 microM) and methylfurmethide (1 to 3 microM) produced reversible membrane depolarizations (less than or equal to 15 mV) accompanied by a fall in input conductance and an increased tendency toward repetitive spike discharges. The spike configuration was unchanged. 3 Analysis of steady-state current/voltage curves revealed the most consistent muscarinic effect to be a large reduction (approximately 50% at 10 microM mus…
