Agenesis of the corpus callosum: a study of the frequency of associated malformations.

Review of 11 cases of agenesis of the corpus callosum studied at our institution revealed a high incidence of associated anomalies. None patients had associated malformations of the central nervous system, 6 involving the pyramidal system. Eight cases were associated with malformations in the rest of the body. Review of completely examined cases from the literature yielded 47 examples of associated malformations. These were varied and without consistent pattern, with the possible exception of facial abnormalities.

[Maternal serum screening: possibilities of using in the complex diagnosis of fetal malformation].

Along with ultrasonography, maternal serum screening is an initial stage of a diagnostic process of genetically determined fetal malformation. Based on the performed clinical and statistical study, the author estimated the values of alpha-fetoprotein and beta-subunit of chorionic gonadotropin if the fetus has malformations of the central nervous system, heart, anterior abdominal wall, and urinary tract and Down's syndrome. The informative value of alpha-fetoprotein was confirmed in malformations of the anterior abdominal wall (gastroschisis, omphalocele) and the central nervous system (hydroce

Exploration of treatment strategies for cerebral cavernous malformations: two case reports on non-resection treatment and literature review.

BACKGROUND: Cavernous malformations are common vascular abnormalities of the central nervous system, but cavernous malformations of the cerebral aqueduct are rare. The choice of treatment is influenced by various factors. CASE DESCRIPTION: We report two cases of midbrain cavernous malformations. Both cases involved midbrain lesions obstructing the cerebral aqueduct, leading to obstructive hydrocephalus. The primary symptoms and complaints of the patients were related to hydrocephalus. Prior to surgery, patients underwent comprehensive imaging evaluations and received endoscopic third ventricu

Congenital malformations in Shimla.

Congenital malformations were studied over a period of five years in 10,100 consecutive births including still births at Indira Gandhi Medical College, Shimla. Out of these, 180 babies had one or the other congenital malformations and the overall incidence was 1.78%. Amongst the 311 still born babies 47 had congenital malformations indicating that the incidence of congenital malformations was much higher in still born babies (15.1%) as compared to the live born babies (1.3%). The malformations involving the central nervous system were the commonest (40%) followed by musculoskeletal system (23.

[Congenital anomalies reflected in materials of obstetric ward of the Hospital of Internal Affairs in Warsaw during the period 1988-96].

A congenital anomaly consists of a departure from the normal anatomic architecture of an organ or system. Malformations can be considered as the result of a developmental arrest of the primordium (incomplete morphogenesis), redundant morphogenesis, or aberrant morphogenesis. Congenital malformations establish more and more percentage of reason of incidence and morbidity in newborn and young children. It is also great family and social problem. The subjects studied were all cases of abnormality identified pre- or postnatally. The commonest system malformations were urogenitally tract, central n

New insight into DAVF pathology-Clues from meningeal immunity.

In recent years, with the current access in techniques, studies have significantly advanced the knowledge on meningeal immunity, revealing that the central nervous system (CNS) border acts as an immune landscape. The latest concept of meningeal immune system is a tertiary structure, which is a comprehensive overview of the meningeal immune system from macro to micro. We comprehensively reviewed recent advances in meningeal immunity, particularly the new understanding of the dural sinus and meningeal lymphatics. Moreover, based on the clues from the meningeal immunity, new insights were propose

Mutations in α- and β-tubulin encoding genes: implications in brain malformations.

The tubulin gene family is mainly expressed in post-mitotic neurons during cortical development with a specific spatial and temporal expression pattern. Members of this family encode dimeric proteins consisting of two closely related subunits (α and β), representing the major constituents of microtubules. Tubulin genes play a crucial role in the mechanisms of the Central Nervous System development such as neuronal migration and axonal guidance (axon outgrowth and maintenance). Different mutations in α/β-tubulin genes (TUBA1A, TUBA8, TUBB2A, TUBB4A, TUBB2B, TUBB3, and TUBB) might alter the dyna

Introduction: Brain malformations.

This issue of the American Journal of Medical Genetics Seminar Series Part C is dedicated to congenital brain malformations with a special focus on the molecular mechanisms underlying this fascinating, and often complex, group of developmental brain disorders. As with most genetic disorders, the past few years have witnessed a dramatic leap in our understanding of the molecular basis of these malformations that include both constitutional and post-zygotic (or mosaic) genetic aberrations. This is best exemplified by the recent identification of mutations within components of the PI3K-AKT-mTOR p

Radiogenomics of congenital brain malformations: Linking embryology, genetics, and imaging.

PURPOSE: Congenital brain malformations are structural anomalies present at birth stemming from underlying genetic mutations or prenatal disruptions. The increased use of advanced genomic sequencing has led to major breakthroughs in pediatric neurogenetics; however, progress in unraveling the genetic basis of many central nervous system (CNS) malformations has lagged. This gap is partly due to the complexity of brain development and challenges like establishing the genetic culprit in somatic mosaicism. This review aims to integrate embryology, genetics, and neuroimaging to provide a practical

Arnold-Chiari malformation and associated anomalies in a dicephalic newborn calf.

A full-term female newborn calf of the Japanese black short-horn breed which was two-headed (dicephalus) and had two incomplete necks was carefully dissected. The calf was one of twins, the other being a normal male. There was no record of reproductive problems with the cow. The left and right heads were of almost the same size and shape. Each head had a nose, mouth, eyeballs, and ears. The brains were very congested. That is to say, in both the left and right brains, the longitudinal fissure was unclear, and these was narrower pattern in the cerebral gyri. The surfaces of the two brains displ

French protocol for diagnosis and management of type 1 interferonopathies.

Type I interferonopathies are rare genetic diseases characterised by excessive production or signalling of type I interferons (IFN-I), which are key cytokines in the antiviral response. These conditions lead to inappropriate activation of IFN-I pathway, even in the absence of viral stimulation. Over thirty monogenic conditions have been identified, with Aicardi-Goutières syndrome being the most common. The genes involved often relate to the metabolism of intracellular nucleic acids, their detection and signalling pathways, contributing to excessive IFN-I production or signalling. Features usua

JAK Inhibition in Aicardi-Goutières Syndrome: a Monocentric Multidisciplinary Real-World Approach Study.

The paradigm type I interferonopathy Aicardi-Goutières syndrome (AGS) is most typically characterized by severe neurological involvement. AGS is considered an immune-mediated disease, poorly responsive to conventional immunosuppression. Premised on a chronic enhancement of type I interferon signaling, JAK1/2 inhibition has been trialed in AGS, with clear improvements in cutaneous and systemic disease manifestations. Contrastingly, treatment efficacy at the level of the neurological system has been less conclusive. Here, we report our real-word approach study of JAK1/2 inhibition in 11 patients

DNA damage contributes to neurotoxic inflammation in Aicardi-Goutières syndrome astrocytes.

Aberrant induction of type I IFN is a hallmark of the inherited encephalopathy Aicardi-Goutières syndrome (AGS), but the mechanisms triggering disease in the human central nervous system (CNS) remain elusive. Here, we generated human models of AGS using genetically modified and patient-derived pluripotent stem cells harboring TREX1 or RNASEH2B loss-of-function alleles. Genome-wide transcriptomic analysis reveals that spontaneous proinflammatory activation in AGS astrocytes initiates signaling cascades impacting multiple CNS cell subsets analyzed at the single-cell level. We identify accumulati
