Energy Stress-Induced Neuroprotection Against Ferroptosis in Dopaminergic Neurons.

Ferroptosis, an iron-dependent form of regulated necrosis, is implicated in the pathogenesis of Parkinson's disease (PD). We studied the influence of energy stress on ferroptosis in differentiated dopaminergic neurons (LUHMES). Glucose deprivation conferred protection against ferroptosis induced by erastin or arachidonic acid plus iron by reducing lipid peroxidation. Glucose withdrawal did not protect against RSL3-induced ferroptosis, suggesting that direct GPX4 inhibition cannot be reversed by metabolic modulation. The expression of ferroptosis markers ACSL4, GPX4, xCT, and TFRc remained unal

Mitophagy bridges glucose metabolism, inflammation and neuroprotection in astrocytes.

Mitochondria regulate ATP production, calcium buffering, and apoptotic signaling, and clearing dysfunctional mitochondria by mitophagy is essential for cellular homeostasis. While PINK1-dependent mitophagy is well-characterized in neurons, its function in glial cells such as astrocytes is less understood. Our study demonstrates that PINK1-mitophagy in astrocytes occurs faster and with less spatial restriction compared to neurons. This pathway was specifically regulated in astrocytes by the glycolytic enzyme, HK2 (hexokinase 2), which forms a glucose-dependent complex with PINK1 following mitoc

Proven Neonatal Neuroprotection Strategies: Magnesium Sulfate and Therapeutic Hypothermia.

Neurodevelopmental impairments follow both term and preterm birth, and include cerebral palsy (CP), cognitive dysfunction, and sensory impairment (blindness and deafness). Proven neuroprotective strategies include antepartum magnesium sulfate (MgSO4) administration prior to anticipated preterm delivery, delayed cord clamping, and therapeutic hypothermia for term and late preterm infants. MgSO4 administration prior to anticipated preterm delivery has been shown to reduce the risk of death, cerebral palsy and major neurodevelopmental disorders, whereas therapeutic hypothermia in term and later p

Remyelination and neuroprotection translational trials: Lessons from optic neuritis.

Multiple sclerosis disease-modifying treatments have reduced inflammatory activity but have limited impact on progressive neurodegeneration. In this context, remyelination and neuroprotection appear as key therapeutic goals. The visual pathway offers an attractive translational model, as optic neuritis recapitulates the demyelinating and neurodegenerative features of multiple sclerosis while providing accessible functional and structural readouts through visual evoked potentials and optical coherence tomography. Over the past two decades, several clinical trials have evaluated remyelinating an…

Exploring Hypoxia-Induced Neuroprotection Mechanisms in Post-Stroke Recovery.

Stroke recovery is a multifaceted process influenced by various neuroprotective mechanisms that support long-term rehabilitation. Recent studies on hypoxia-induced neuroprotection have shown promising potential in enhancing stroke recovery through adaptive cellular responses. Hypoxic conditioning in techniques of passive intermittent hypoxic exposures (IHE) or intermittent hypoxic-hyperoxic exposures (IHHE), which alternates between low and normal/high oxygen levels, is emerging as a novel complementary therapy that may improve post-stroke outcomes by promoting neuroprotection, neurogenesis, a…

Antioxidant Bioactive Agents for Neuroprotection Against Perinatal Brain Injury.

Physiological oxidative stress plays a pivotal role in supporting proper growth and development. While moderate oxidative stress is essential for activating key metabolic pathways and maintaining normal cellular signaling, excessive production of reactive oxygen species (ROSs) can overwhelm the immature antioxidant systems of newborns, potentially leading to cellular damage and impaired physiological function. This vulnerability is particularly pronounced in the central nervous system, where limited detoxification capacity exacerbates the risk of oxidative damage, following hypoxic-ischemic ev

Glyburide confers neuroprotection against age-related macular degeneration (AMD).

Glyburide, a sulfonylurea drug used to treat type 2 diabetes, boasts neuroprotective effects by targeting the sulfonylurea receptor 1 (SUR1) and associated ion channels in various cell types, including those in the central nervous system and the retina. Previously, we demonstrated that glyburide therapy improved retinal function and structure in a rat model of diabetic retinopathy. In the present study, we explore the application of glyburide in non-neovascular ("dry") age-related macular degeneration (AMD), another progressive disease characterized by oxidative stress-induced damage and neuro

Emerging Gene Therapy Technologies for Retinal Ganglion Cell Neuroprotection.

Optic neuropathies encompass a breadth of diseases that ultimately result in dysfunction and/or loss of retinal ganglion cells (RGCs). Although visual impairment from optic neuropathies is common, there is a lack of effective clinical treatments. Addressing a critical need for novel interventions, preclinical studies have been generating a growing body of evidence that identify promising new drug-based and cell-based therapies. Gene therapy is another emerging therapeutic field that offers the potential of specifically and robustly increasing long-term RGC survival in optic neuropathies. Gene

Exploring common genetic contributors to neuroprotection from amyloid pathology.

Preclinical Alzheimer's disease describes some individuals who harbour Alzheimer's pathologies but are asymptomatic. For this study, we hypothesized that genetic variation may help protect some individuals from Alzheimer's-related neurodegeneration. We therefore conducted a genome-wide association study using 5 891 064 common variants to assess whether genetic variation modifies the association between baseline beta-amyloid, as measured by both cerebrospinal fluid and positron emission tomography, and neurodegeneration defined using MRI measures of hippocampal volume. We combined and jointly a

Neurologic Dysfunction and Neuroprotection in Transcatheter Aortic Valve Implantation.

Transcatheter aortic valve implantation (TAVI) is a fast-growing procedure. Expanding to low-risk patients, it has surpassed surgical aortic valve implantation in frequency and has been associated with excellent outcomes. Stroke is a devastating complication after transcatheter aortic valve implantation. Silent brain infarcts identified by diffusion-weighted magnetic resonance imaging are present in most patients following TAVI. Postoperative delirium and cognitive dysfunction are common neurologic complications. The stroke and silent brain infarcts are likely caused by particulate emboli rele
