Immunohistochemical localization of HLA-DR-positive cells in unerupted and erupted normal and carious human teeth.

Class II major histocompatibility complex (MHC) antigen-expressing cells are generally associated with the early phase of the immune response. We have studied the distribution of class II-expressing cells in developing, normal, and carious human teeth to clarify when human pulp acquires an immunologic defense potential and how this reacts to dental caries. Antigen-expressing cells were identified immunohistochemically by means of HLA-DR monoclonal antibody. In the pulp of unerupted developing teeth, numerous HLA-DR-positive cells were distributed mainly in and around the odontoblast layer. In

Histone methyltransferase NSD1 maintains Egr1 expression to orchestrate primary/reactionary dentinogenesis.

Dentinogenesis is a highly coordinated process orchestrated by odontoblasts that is not only critical for tooth development, but also essential for pulp vitality after injury. Although the signaling pathways regulating this process have been well characterized, it remains elusive how dentinogenesis is epigenetically regulated during tooth development and adulthood. Here, we show that nuclear receptor binding SET domain protein 1 (NSD1), a histone methyltransferase catalyzing the dimethylation of histone H3 at lysine 36 (H3K36me2), exhibits dynamic spatiotemporal expression patterns during odon

Transcriptional repression of the Dspp gene leads to dentinogenesis imperfecta phenotype in Col1a1-Trps1 transgenic mice.

Dentinogenesis imperfecta (DGI) is a hereditary defect of dentin, a calcified tissue that is the most abundant component of teeth. Most commonly, DGI is manifested as a part of osteogenesis imperfecta (OI) or the phenotype is restricted to dental findings only. In the latter case, DGI is caused by mutations in the DSPP gene, which codes for dentin sialoprotein (DSP) and dentin phosphoprotein (DPP). Although these two proteins together constitute the majority of noncollagenous proteins of the dentin, little is known about their transcriptional regulation. Here we demonstrate that mice overexpre

Constitutive stabilization of ß-catenin in the dental mesenchyme leads to excessive dentin and cementum formation.

Wnt/ß-catenin signaling plays an important role in morphogenesis and cellular differentiation during development. Essential roles of Wnt/ß-catenin signaling in tooth morphogenesis have been well known, but the involvement of Wnt/ß-catenin signaling in the dental hard tissue formation remains undefined. To understand roles of Wnt/ß-catenin signaling in dentin and cementum formation, we generated and analyzed the conditional ß-catenin stabilized mice in the dental mesenchyme. The OC-Cre;Catnb(lox(ex3)/+) mice exhibited malformed teeth characterized by aberrantly formed dentin and excessively dep

The effect of human umbilical cord mesenchymal stem cells combined with concentrated growth factor on repairing necrotic pulp caused by dental caries.

This study investigated the impact of combining human umbilical cord mesenchymal stem cells (hUC-MSCs) with concentrated growth factor (CGF) on regenerating necrotic pulp. Ten-month-old male Bama miniature pigs were divided into control and caries groups. The experimental teeth were randomly divided into three groups: caries untreated, Ca(OH)2, and engineering dental pulp-like tissue (EDPT). hUC-MSCs and CGF scaffold were combined to construct EDPT, and the histological structure was observed. Odontoblasts and dental pulp cells were counted in each group. The results showed that hUC-MSCs adher

Immunohistochemical localization of LIM mineralization protein 1 in pulp-dentin complex of human teeth with normal and pathologic conditions.

LIM mineralization protein 1 (LMP-1) is an essential positive regulator of osteoblast differentiation and maturation and bone formation. However, the expression and distribution of LMP-1 in human teeth are largely unknown. The aim of this study was to detect the expression of LMP-1 in normal healthy human teeth and human teeth with various pathologic conditions. LMP-1 expression was determined by immunohistochemistry in all of the samples including 12 healthy teeth, 10 teeth with caries, 12 teeth with pulpitis, and 4 teeth with pulp calcification. We found that LMP-1 was expressed primarily in

Miniaturization: How many cells are needed to build a tooth?

BACKGROUND: Organs that develop early in life, and are replaced by a larger version as the animal grows, often represent a miniature version of the adult organ. Teeth constituting the first functional dentition in small-sized teleost fish, such as medaka (Oryzias latipes), are examples of such miniature organs. With a dentin cone as small as the size of one human cell, or even smaller, these teeth raise the question how many dentin-producing cells (odontoblasts) are required to build such a tooth, and whether this number can be as little as one. RESULTS: Based on detailed observations with tr

The effect of TGF-beta 2 on dentin apposition and hardness in transgenic mice.

Transforming growth factor beta, TGF-beta, is expressed during tooth formation and can induce pre-odontoblast differentiation and formation of functional odontoblast-like cells in vitro. In addition, exogenous TGF-beta can increase reparative dentin formation, presumably by acting on odontoblasts. In this study, we examined the tooth phenotype of transgenic mice, in which TGF-beta 2 expression is directed by the osteocalcin promoter. Previous studies have shown that these mice have a bone phenotype that resembles that of human osteoporosis, including the existence of spontaneous fractures. Mic

Dental cells express factors that regulate bone resorption.

Odontoblasts and osteoblasts produce similar highly mineralized extracellular matrices. In bone, osteoblasts/stromal cells regulate osteoclast (ocl) formation and bone resorption by producing factors like osteoprotegerin (OPG), osteoclast differentiating factor (ODF/RANKL), and macrophage colony-stimulating factor (M-CSF) that interact with hematopoietic ocl precursor cells. Using odontoblast and pulp cell lines, we detected a constitutive expression of OPG, RANKL, and M-CSF mRNA in both cell types. OPG and RANKL proteins were also detectable. In vivo, RANKL and OPG were localized to odontobla

On the repair of the dentine barrier.

The overall aim of this thesis was to study some aspects of the repair of the dentine barrier, especially in conjunction with dental pulp capping. Understanding the events leading to the healing of the dentine and pulp, and hence successfully preserving the vitality and functions of the tooth, would lead to a scientific basis for a less invasive treatment of pulp exposures than performing root canal treatments. The surfaces of the body have physiological barrier functions aimed at protecting the body from external noxious agents. In the tooth, the odontoblasts, which line the outermost part of

Human pulpal response to citric acid cavity cleanser.

Treatment of cavities with citric acid cavity cleanser, also used as an etchant, before restoration with zinc oxide-eugenol cement, was toxic to the human dental pulp in comparison to sham-treated controls. The cleanser caused a significantly (P less than .01) greater intensity and incidence (P less than .01) of inflammation. The added insult from cleanser pretreatment caused destruction of the odontoblast processes withnin the cut dentinal tubules, and an increase in diameter of the tubules. These effects on dentin and odontoblasts probably contributed to the observed increase in adverse pulp

The Critical Role of MMP13 in Regulating Tooth Development and Reactionary Dentinogenesis Repair Through the Wnt Signaling Pathway.

Matrix-metalloproteinase-13 (MMP13) is important for bone formation and remodeling; however, its role in tooth development remains unknown. To investigate this, MMP13-knockout (Mmp13 -/- ) mice were used to analyze phenotypic changes in the dentin-pulp complex, mineralization-associated marker-expression, and mechanistic interactions. Immunohistochemistry demonstrated high MMP13-expression in pulp-tissue, ameloblasts, odontoblasts, and dentin in developing WT-molars, which reduced in adults, with human-DPC cultures demonstrating a >2000-fold increase in Mmp13-expression during mineralization.

Slc26a2-mediated sulfate metabolism is important in tooth development.

The sulfate transporter gene SLC26A2 is crucial for skeletal formation, as evidenced by its role in diastrophic dysplasia, a type of skeletal dysplasia in humans. Although SLC26A2-related chondrodysplasia also affects craniofacial and tooth development, its specific role in these processes remains unclear. In this study, we explored the pivotal roles of SLC26A2-mediated sulfate metabolism during tooth development. We found that Slc26a2 was predominantly expressed in dental tissues, including odontoblasts and ameloblasts. Slc26a2 knockout (Slc26a2-KO-Δexon2) mice exhibited distinct craniofacial

Tooth development disorders in infants of rat dams exposed to 2,3,7,8-tetrachlorodibenzo-p-dioxin and protective role of tocopherol and acetylsalicylic acid.

Aryl hybrocardon receptor (AhR) activation plays a key role in the pathomechanism of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)-induced defective spatial structure of teeth caused by disordered collagen synthesis. The aim of this study was to identify the influence of dioxins present in female Buffalo rats on the dental structure of their offspring's in the neonatal period and the potential of α-tocopherol and acetylsalicylic acid in curbing post-dioxin hard tissue defects. Research material consisted of molar teeth (n=40) of rat pups which had been given a single dose of TCDD and were then tr
