Systematic evaluation of phenolic compounds and protective capacity of a new mulberry cultivar J33 against palmitic acid-induced lipotoxicity using a simulated digestion method.

This research aimed to investigate the protective effects of a new mulberry cultivar J33 with simulated gastrointestinal digestion against palmitic acid (PA)-induced lipotoxicity. LC-MS analysis revealed that the contents of four flavonoid glycosides (quercetin rhamnosylhexoside hexoside, quercetin rhamnosylhexoside, quercetin hexoside, kaempferol rhamnosylhexoside) increased after digestion. Besides, mulberry digest (MBD) at 0.5-2 mg/mL significantly reduced PA-induced lipotoxicity in human hepatocytes, while mulberry extract without digestion (MBE) showed no protection. Further investigation…

Modulation of postprandial lipaemia by a single meal containing a commonly consumed interesterified palmitic acid-rich fat blend compared to a non-interesterified equivalent.

PURPOSE: Interesterification of palm stearin and palm kernal (PSt/PK) is widely used by the food industry to create fats with desirable functional characteristics for applications in spreads and bakery products, negating the need for trans fatty acids. Previous studies have reported reduced postprandial lipaemia, an independent risk factor for CVD, following interesterified (IE) palmitic and stearic acid-rich fats that are not currently widely used by the food industry. The current study investigates the effect of the most commonly consumed PSt/PK IE blend on postprandial lipaemia. METHODS: A

Enhancement of Agkistrodon piscivorus piscivorus venom phospholipase A2 activity toward phosphatidylcholine vesicles by lysolecithin and palmitic acid: studies with fluorescent probes of membrane structure.

The activity of phospholipase A2 from snake venom to hydrolyze bilayers of phosphatidylcholines is greatly enhanced by the presence of the hydrolysis products, lysolecithin and fatty acid, in the bilayer. The fluorescence of several probes of membrane structure was used to monitor changes in bilayer physical properties during vesicle hydrolysis. These changes were compared to emission spectra and fluorescence polarization results occurring upon direct addition of lysolecithin and/or fatty acid to the bilayer. The excimer to monomer ratio of 1,3-bis(1-pyrene)propane was insensitive to vesicle h

Differential effects of stearic, palmitic and oleic acid enriched diets on gut microbiome, bile acid and cholesterol metabolism in mildly hypercholesterolemic post-menopausal females: A secondary analysis of a randomized controlled trial.

BACKGROUND: Stearic acid (18:0), a saturated fatty acid (SFA), does not raise plasma LDL cholesterol concentrations compared with palmitic acid (16:0), and has similar effects to oleic acid (18:1), but underlying mechanisms remain unclear. OBJECTIVES: To determine if the hypocholesterolemic effects of dietary 18:0 and 18:1 relative to 16:0 are mediated by alterations in gut microbiome, bile acid (BA), and cholesterol metabolism. METHODS: Secondary analysis of a randomized controlled crossover trial in mildly hypercholesterolemic postmenopausal females (n=17) who consumed isocaloric diets enr…

Ascorbic acid and ascorbyl palmitate-loaded liposomes: Development, characterization, stability evaluation, in vitro security profile, antimicrobial and antioxidant activities.

The objective of this work was to prepare and characterize liposomes containing co-encapsulated ascorbic acid (AA) and ascorbyl palmitate (AP), as well as to evaluate their stability, cytotoxicity, antioxidant, and antimicrobial activity. Through the pre-formulation studies, it was possible to improve the formulation, as leaving it more stable and with a greater antioxidant activity, resulting in a formulation designated LIP-AAP, with 161 nm vesicle size, 0.215 polydispersity index, -31.7 mV zeta potential, and pH of 3.34. Encapsulation efficiencies were 37% for AA and 79% for AP, and the cont

Establishment of an Adult Medaka Fatty Liver Model by Administration of a Gubra-Amylin-Nonalcoholic Steatohepatitis Diet Containing High Levels of Palmitic Acid and Fructose.

Among lifestyle-related diseases, fatty liver is the most common liver disease. To date, mammalian models have been used to develop methods for inhibiting fatty liver progression; however, new, more efficient models are expected. This study investigated the creation of a new model to produce fatty liver more efficiently than the high-fat diet medaka model that has been used to date. We compared the GAN (Gubra-Amylin nonalcoholic steatohepatitis) diet, which has been used in recent years to induce fatty liver in mice, and the high-fat diet (HFD). Following administration of the diets for three

Sparstolonin B suppresses free fatty acid palmitate-induced chondrocyte inflammation and mitigates post-traumatic arthritis in obese mice.

Abnormal lipid metabolism, such as systemic increased free fatty acid, results in overproduction of pro-inflammatory enzymes and cytokines, which is crucial in the development of obesity-related osteoarthritis (OA). However, there are only a few drugs that target the lipotoxicity of OA. Recent researches have documented that the traditional Chinese medicine, Sparstolonin B (Ssn B), exerted anti-inflammatory effects in various diseases, but not yet in OA. On the basis of this evidence, our works purposed to evaluate the effect of Ssn B on free fatty acid (FFA) palmitate (PA)-stimulated human os

Palmitic acid, but not high-glucose, induced myocardial apoptosis is alleviated by N‑acetylcysteine due to attenuated mitochondrial-derived ROS accumulation-induced endoplasmic reticulum stress.

Pharmacological inhibition of reactive oxygen species (ROS) is a potential strategy to prevent diabetes-induced cardiac dysfunction. This study was designed to investigate precise effects of antioxidant N‑acetylcysteine (NAC) in alleviating diabetic cardiomyopathy (DCM). Echocardiography and histologic studies were performed 12 weeks after streptozocin injection. Protein levels involved in endoplasmic reticulum stress (ERS) and apoptosis were analyzed by western blotting in diabetic hearts or high-glucose (HG, 30 mM)- and palmitic acid (PA, 300 μM)-cultured neonatal rat cardiomyocytes (NRCMs).…

Synthesis, characterisation, and in vitro assessment of hyaluronic acid-coated solid lipid nanoparticles entrapping dexamethasone palmitate designed for intra-articular administration.

Osteoarthritis (OA) is the major cause of pain and disability worldwide, particularly affecting the knee joint. Although intra-articular (IA) treatments including corticosteroids and hyaluronic acid (HA), are commonly used to manage symptoms, their therapeutic effects are often short-lived, necessitating repeated injections. In this study, HA-coated solid lipid nanoparticles (HA-DXP-SLNs) entrapping with the glucocorticoid prodrug, dexamethasone palmitate (DXP) was developed as dual-function nanocarrier system for IA delivery. Using a scalable microfluidics platform, monodisperse SLNs of ∼90 n

Low-Level Saturated Fatty Acid Palmitate Benefits Liver Cells by Boosting Mitochondrial Metabolism via CDK1-SIRT3-CPT2 Cascade.

Saturated fatty acids (SFAs) (the "bad" fat), especially palmitate (PA), in the human diet are blamed for potential health risks such as obesity and cancer because of SFA-induced lipotoxicity. However, epidemiological results demonstrate a latent benefit of SFAs, and it remains elusive whether a certain low level of SFAs is physiologically essential for maintaining cell metabolic hemostasis. Here, we demonstrate that although high-level PA (HPA) indeed induces lipotoxic effects in liver cells, low-level PA (LPA) increases mitochondrial functions and alleviates the injuries induced by HPA or he

Activation of the Nrf2/ARE signaling pathway protects against palmitic acid-induced renal tubular epithelial cell injury by ameliorating mitochondrial reactive oxygen species-mediated mitochondrial dysfunction.

Chronic kidney disease (CKD) is often accompanied by dyslipidemia, and abnormal lipid metabolism in proximal tubule cells is considered closely related to the dysfunction of proximal tubule cells and eventually leads to accelerated kidney damage. Nuclear factor E2-related factor 2 (Nrf2), known as a redox-sensitive transcription factor, is responsible for regulating cellular redox homeostasis. However, the exact role of Nrf2 in dyslipidemia-induced dysfunction of proximal tubule cells is still not fully elucidated. In the present study, we showed that palmitic acid (PA) induced mitochondrial d…

Short-Chain Fatty Acids and Palmitate Induce Distinct Metabolic and Phenotypic Signatures in Normal and Ischemic Skeletal Muscle Microvascular Endothelial Cells.

Background: Palmitate, a long-chain fatty acid, is well known to be a significant risk factor for cardiovascular diseases. In our current study, we wanted to determine whether palmitate treatment further aggravates ischemic endothelial cell (EC) injury and can serve as an in vitro model that emulates diabetic peripheral artery disease (diabetic-PAD). Short-chain fatty acid (SCFA) treatment was used as an additional comparator for palmitate-induced vascular dysfunction in normal or ischemic ECs in vitro. Methods: Hypoxia serum starvation (HSS) was used as an in vitro model for PAD. Cell surviva

Oral retinyl palmitate or retinoic acid corrects mucosal IgA responses toward an intranasal influenza virus vaccine in vitamin A deficient mice.

Vitamin A deficiency (VAD) is a leading cause of pediatric morbidity and mortality due to infectious diseases. Recent pre-clinical studies have revealed that VAD impairs mucosal IgA-producing antibody forming cell (AFC) responses toward a paramyxovirus vaccine in the upper respiratory tract (URT), thus impeding a first line of defense at the pathogen's point-of-entry. The studies described here tested the hypothesis that VAD may also impair immune responses after FluMist vaccinations. Results show that (i) IgA-producing antibody forming cells (AFCs) are significantly reduced following FluMist

Palmitic acid induced the onset of lipotoxicity in a HepaSH-on-chip model with raised of H2O2 and IL-6, and altered p38/MAPK & JAK/STAT pathways.

Hepatic inflammation induced by lipotoxicity is a key event in the progression of liver steatosis to non-alcoholic steatohepatitis (NASH). To characterize this process, we investigated the response of HepaSH cells-a novel mature human hepatocyte derived from chimeric mice-to a weak but chronic palmitic acid (PA) exposure, a prevalent dietary fatty acid. To mimic the hepatic microphysiological environment, cells were cultured using organ-on-chip technology and exposed to PA for two weeks. Functional assays revealed no changes in glucose or lactate metabolism. Likewise, PA exposure did not alter…

Liberation of 14CO2 from 14C-fatty acids by riboflavin-deficient sucking rat pups: a study of 14C-Octanoate and 14C-palmitate oxidation in vivo.

To test the hypothesis that impaired fatty acid oxidation in the mitochondria of riboflavin-deficient rats may be reflected in impaired rates of fatty acid oxidation to carbon dioxide in vivo, the conversion of 14C-labelled fatty acids to 14CO2 was measured in rat pups. Sucking pups were selected, since whole body energy expenditure was reduced by riboflavin deficiency in this age group. A gas-flow apparatus was designed, to permit the collection of expired CO2 over a 4 hr period, in pups maintained at constant temperature. 14C-octanoate and 14C-palmitate, given by intraperitoneal injection, w
