Chondroitin sulfate proteoglycans (CSPGs) are involved in several processes relevant to recovery of function after CNS damage. They restrict axon regeneration through their presence in glial scar tissue and plasticity through their presence in perineuronal nets (PNNs), affect memory through their effect on dendritic spines, and influence the inflammatory reaction. Much of our knowledge of these CSPG effects comes from digestion of their glycosaminoglycan chains by the enzyme chondroitinase ABC (ChABC). ChABC after spinal cord injury permits some axon regeneration and greatly increases plastici

Perineuronal nets (PNNs) are specialized extracellular matrix aggregates surrounding distinct neuronal populations and regulating synaptic functions and plasticity. Previous findings showed robust PNN decreases in amygdala, entorhinal cortex and prefrontal cortex of subjects with schizophrenia (SZ), but not bipolar disorder (BD). These studies were carried out using a chondroitin sulfate proteoglycan (CSPG) lectin marker. Here, we tested the hypothesis that the CSPG aggrecan, and 6-sulfated chondroitin sulfate (CS-6) chains highly represented in aggrecan, may contribute to these abnormalities.

The rate of cognitive decline in Alzheimer's disease (AD) varies considerably from person to person. Numerous epidemiological studies point to the protective effects of cognitive, social, and physical enrichment as potential mediators of cognitive decline in AD; however, there is much debate as to the mechanism underlying these protective effects. The retrosplenial cortex (RSC) is one of the earliest brain regions with impaired functions during AD pathogenesis, and its activity is affected by cognitive, social, and physical stimulation, making it a particularly interesting region to investigat

Chondroitin sulfate (CS) plays essential roles in neurodevelopment and brain aging. Alterations in glycosaminoglycan composition on the neuronal cell surface and within the extracellular matrix (ECM) contribute to the progression of tauopathies, including Alzheimer's disease (AD). Disease-associated changes in CS sulfation patterns have been linked to tau pathology, and administration of CS-specific antibodies has been shown to improve cognitive function in AD model mice. CSGALNACT1 catalyzes the initial step of CS chain elongation by transferring N-acetylgalactosamine residues to the linkage

In humans, early-life adversity (ELA) predisposes individuals to neuropsychiatric conditions, with different types of adverse experiences linked more strongly to certain maladaptive outcomes. Maternal separation with early weaning (MSEW) and limited bedding and nesting (LBN) are rodent models that expose litters to very different postnatal manipulations, but the effect of these paradigms on offspring through differences in maternal care and neuronal activation remains incompletely explored. To address this gap, we directly compared behavior of dams in MSEW and LBN conditions to control conditi

Cocaine and methamphetamine use disorders pose public health concerns due to the high propensity for relapse driven by persistent drug-associated memories. Output from the medial prefrontal cortex (mPFC) regulates relapse and is controlled by parvalbumin (PV) interneurons, which are surrounded by perineuronal nets (PNNs) that support precise PV neuron firing. We previously showed that removal of mPFC PNNs, which reduces PV firing, disrupted reconsolidation of a cocaine self-administration memory when rats were given a novel, but not familiar, memory retrieval session. Ketamine rapidly suppress

PURPOSE: This study investigated the effects of fluoxetine on noise-induced injuries to the cochlea and auditory nerve, with a focus on its impact on perineuronal nets (PNNs) and gene expression changes in the ventral cochlear nuclei (VCN).
METHODS: Sprague-Dawley rats were exposed to white noise at 115 dB SPL for 3 hours per day over 3 weeks. After measuring auditory brainstem response (ABR) thresholds, rats were treated with fluoxetine (10 mg/kg) for 19 days. Four groups were included (vehicle, fluoxetine, noise + vehicle, and noise + fluoxetine; n = 10 per group). ABR measurements, analysi

Neuroinflammation, characterized by activation of glial cells, plays a critical role in central nervous system disorders. However, the precise mechanisms of neuroinflammation contributing to cognitive impairment remain elusive. Perineuronal nets (PNNs) are extracellular matrixes that envelop the cell bodies and dendrites of parvalbumin (PV)-positive interneurons and may be mediated by apolipoprotein E (ApoE) gene. To investigate whether disruption of PNNs associated with ApoE is implicated in neuroinflammation-induced cognitive impairment, we established a neuroinflammation model by administer

Rett Syndrome (RTT) is a rare X-linked neurodevelopmental disorder, mainly caused by mutations in the MECP2 gene. Reduction in monoamine levels in RTT patients and mouse models suggested the possibility to rescue clinical phenotypes through antidepressants. Accordingly, we tested mirtazapine (MTZ), a noradrenergic and specific-serotonergic tetracyclic antidepressant (NaSSA). In previous studies, we showed high tolerability and significant positive effects of MTZ in male Mecp21m1.1Bird-knock-out mice, adult female Mecp2tm1.1Bird-heterozygous (Mecp2+/-) mice, and adult female RTT patients. Existing work on drug-induced synaptic changes has shown that the expression of perineuronal nets (PNNs) at the cerebellar cortex can be regulated by cocaine-related memory. However, these studies on animals have mostly relied on limited manually-driven procedures, and lack some more rigorous statistical approaches and more automated techniques. In this work, established methods from computer vision and machine learning are considered to build stronger evidence of those previous findings. To that end, an image descriptor is designed to characterize PNNs images; unsupervised learning (clusterin

Successful treatment of drug addiction is hampered by high relapse rates during periods of abstinence. Neuroadaptation in the medial prefrontal cortex (mPFC) is thought to have a crucial role in vulnerability to relapse to drug seeking, but the molecular and cellular mechanisms remain largely unknown. To identify protein changes that contribute to relapse susceptibility, we investigated synaptic membrane fractions from the mPFC of rats that underwent 21 days of forced abstinence following heroin self-administration. Quantitative proteomics revealed that long-term abstinence from heroin self-ad

Cystatin-related epididymal spermatogenic (CRES) is a member of a reproductive subgroup of family 2 cystatins and part of a functional amyloid-containing extracellular matrix (ECM) with host defense functions in the epididymal lumen. CRES and the endogenous epididymal amyloid matrix exhibit high plasticity, forming distinct amyloid structures, with antimicrobial functions tailored to specific bacterial strains. We recently demonstrated that CRES/CRES amyloid is also present in the mammalian brain ECM. In this study, we utilized a CRES knockout (KO) mouse model to determine if the loss of CRES

Hind limb locomotor activity spontaneously recovers after complete spinal transection (CST) in neonatal rats, but the mechanisms underlying the recovery are poorly understood. The perineuronal net (PNN) surrounding the neuronal cell bodies comprises an extracellular matrix that regulates neuronal plasticity during development. Here, we examined the expression of chondroitin sulfate (CS), a major component of the PNN, on motoneurons after CST in neonatal rats, and compared it with that in juvenile rats, in which hindlimb locomotor activity does not recover spontaneously. The spinal cord was tra

The qualitative and quantitative distribution pattern of hyaluronan (HA), a component of the extracellular matrix (ECM), was studied in the frog central nervous system by using a highly specific HA probe and digital image analysis. HA reaction was observed in both the white and the gray matter, showing a very intense staining around the perikarya and dendrites in the perineuronal net (PN). In the telencephalon, strong reaction was found in different parts of the olfactory system, in the pallium, and in the amygdala. In the diencephalon, intensive staining was found in the nucleus of Bellonci,

OBJECTIVE: Alterations in the somatostatin (SST) and parvalbumin (PVALB) classes of inhibitory GABAergic interneurons in the dorsolateral prefrontal cortex (DLPFC) are thought to contribute to the core cognitive impairments of schizophrenia. Both classes of interneurons are composed of multiple subtypes, but the distinguishing features and relative proportions of these subtypes have not been determined in the DLPFC. The aim of this study is to provide a comprehensive analysis of these subtypes from young and middle-age adults with no known psychiatric disorders.
METHODS: The authors used sing

Neuroinflammation underlies many neurodegenerative disorders and is orchestrated by interactions between microglia, astrocytes, and neurons. While cytokines, chemokines, and damage-associated molecular patterns (DAMPs) are established neuroimmune mediators, extracellular matrix (ECM)-derived protein fragments, collectively termed matrikines, have received little attention in the central nervous system (CNS). Emerging evidence suggests that matrikines constitute a distinct and functionally important class of neuroimmune modulators. This review focuses on six CNS-relevant matrikines: type I coll

Slight and hidden hearing loss in children have been linked to cognitive and social difficulties, and yet the neurobiological mechanisms behind these issues remain poorly understood. Most animal models focus on severe hearing loss, leaving the effects of hidden or slight hearing loss largely unexplored. To uncover the neural mechanisms connecting slight/hidden hearing loss to cognitive and social challenges, we induced hearing loss in young (4-week-old) Wistar rats through noise exposure. We then examined cognitive function (object recognition test) and social behavior (juvenile play behavior

Reciprocal pathways connecting the cerebellum to the prefrontal cortex provide a biological and functional substrate to modulate cognitive functions. Dysfunction of both medial prefrontal cortex (mPFC) and cerebellum underlie the phenotypes of several neuropsychiatric disorders that exhibit comorbidity with substance use disorder (SUD). In people with SUD, cue-action-reward associations appears to be particularly strong and salient, acting as powerful motivational triggers for craving and relapse. Studies of cue reactivity in human with SUD have shown cerebellar activations when drug-related c

Brain oscillations are associated with specific cognitive and sensory processes. How age-related hearing loss (presbycusis) alters cortical oscillations is unclear. Altered inhibitory neurotransmission and temporal processing deficits contribute to speech recognition impairments in presbycusis. Specifically, age-related reduction in parvalbumin positive interneurons and perineuronal nets in the auditory cortex predicts a reduction in gamma oscillations that may lead to a decline in temporal precision and attention. To test the hypothesis that resting and evoked gamma oscillations decline with

Early-life adversity alters inhibitory circuit maturation across developing brain systems, but most studies have focused on corticolimbic regions involved in emotional and cognitive regulation. Whether early adversity also reshapes inhibitory extracellular matrix architecture in primary sensory cortex remains less clear. Here, we examined the effects of maternal separation on parvalbumin-positive (PV+ ) interneurons and Wisteria floribunda agglutinin-labeled perineuronal nets (PNNs) in the mouse primary somatosensory barrel cortex (S1BF), a highly organized sensory region in which laminar posi
