Thyroid peroxidase autoantibodies of IgE class in thyroid autoimmunity.

Recently, we converted a thyroid peroxidase (TPO)-specific human autoantibody Fab (SP1.4) into an IgE molecule (IgE-SP1.4) which permits antigen capture via Fc epsilon receptors on B cells and presentation to T cells. An important question which arose was whether IgE class TPO autoantibodies are present in vivo. By ELISA, TPO autoantibodies of IgG1 and IgG4 subclasses, but not IgE, were readily detectable in patients' sera. However, such negative data were not definitive because high concentrations of IgG class TPO autoantibodies could obscure the presence of much lower concentrations of IgE c

Roles of efficient substrates in enhancement of peroxidase-catalyzed oxidations.

Efficient peroxidase substrates may have a critical role in the oxidation of secondary compounds by peroxidases. Hydrazines are often oxidized slowly by peroxidases due, in part, to hydrazine-dependent inactivation of these enzymes. Peroxidase-catalyzed oxidation of hydrazines may be dramatically affected by an efficient peroxidase substrate. We investigated this hypothesis in a model system using the well-known peroxidase substrate chlorpromazine (CPZ) and the hydrazine derivative isoniazid. CPZ stimulated isoniazid oxidation as measured by nitroblue tetrazolium (NBT) reduction and O2 consump…

Role of calcium in maintaining the heme environment of manganese peroxidase.

We previously demonstrated that manganese peroxidase from the white-rot fungus Phanerochaete chrysosporium was very susceptible to thermal inactivation due to the loss of calcium from the enzyme [Sutherland & Aust (1996) Arch. Biochem. Biophys. 332, 128-134]. The structural changes that occur during thermal inactivation and the release of calcium from manganese peroxidase have now been characterized. Thermal inactivation caused distinct alterations in the heme environment and slight changes in the overall protein structure, both of which were reversed upon reactivation of the enzyme with calci

Hepatic and erythrocytic glutathione peroxidase activity in liver diseases.

Hepatic and erythrocytic glutathione peroxidase activity, together with malondialdehyde levels, were determined as indicators of peroxidation in 83 patients from whom liver biopsies had been taken for diagnostic purposes. On histological study, the patients were classified into groups as minimal changes (including normal liver), steatosis, alcoholic hepatitis, hepatic cirrhosis, light to moderately active chronic hepatitis, and severe chronic active hepatitis. The glutathione peroxidase activity in erythrocytes showed no significant changes in any liver disease group. In the hepatic study, an

Diagnosis of dermatitis herpetiformis by an avidin-biotin-peroxidase method.

BACKGROUND AND DESIGN: Immunofluorescence detection of stippled IgA in dermal papillae has been considered the gold standard in the diagnosis of dermatitis herpetiformis (DH). We have developed an immunohistochemical technique using the avidin-biotin-peroxidase complex that is equally effective as direct immunofluorescence in detecting IgA. We retrospectively studied 43 paraffin-embedded biopsy specimens obtained from patients with DH and a variety of other diseases for the presence of IgA along the basement membrane zone. RESULTS: Eleven immunofluorescence-proved cases of DH were found posit

[Thyroid peroxidase (TPO) gene and pathogenic TPO mutation].

TPO is the key enzyme involved in the thyroid hormone synthesis. The human TPO (hTPO) gene locates on chromosome 2 and consists of 17 exons and 16 introns. Compared with other peroxidases, hTPO is 42% homologous with granulocyte myeloperoxidase. Thyroid cells contain multiple TPO mRNA transcripts of various size. However, the reason is unknown. TTF-1 and TTF-2 are known to regulate TPO gene expression. Moreover, the other factors are becoming clear to regulate it. Congenital TPO defects result in hypothyroidism and goiter. Recent studies clarify a human mutation causing TPO deficiency.

Bacillary haemoglobinuria diagnosis by the peroxidase-antiperoxidase (PAP) technique.

A peroxidase-antiperoxidase (PAP) technique was used to diagnose bacillary haemoglobinuria in formalin-fixed, paraffin-embedded liver tissues of cattle. The PAP method revealed Clostridium haemolyticum in the zone of liver necrosis characteristic of the disease and also in culture smears of this microorganism, but C. novyi type B, C. chauvoei, C. septicum and C. perfringens types B and C remained unstained by the PAP reaction. The PAP technique performed provides a specific, simple and rapid method to diagnose bacillary haemoglobinuria.

Determination of hemoglobin through its peroxidase activity on chlorpromazine.

Chlorpromazine is an excellent chromogen for determining micro-quantities of hemoglobin. The oxidation of chlorpromazine by peroxidase activity of hemoglobin is coupled to a non-enzymatic reaction of second-order. Kinetic analysis of the overall system leads to a discussion about the optimal assay conditions. Spectrophotometric progress curves for the accumulation of the chlorpromazine cation radical during the reaction have been obtained, and further analyzed by non-linear regression. The use of a linear calibration curve of the enzymatic reaction rate against hemoglobin concentration is prop

[Anti-thyroid peroxidase in non-neoplastic thyroid pathology].

Anti-thyroid peroxidase antibodies were prospectively assayed and compared with anti-microsome and anti-thyroglobulin antibodies in 203 patients (188 women, 15 men; mean age 42 +/- 14 years). These patients consulted for hyperthyroidism (n = 42, including 18 with Graves' disease), fumary hypothyroidism (n = 50, including 20 at the diagnosis stage), euthyroid diffuse or nodular goitre (n = 81) or benign euthyroid nodule (n = 14). Sixteen patients examined for fatigue, gynecomastia, menstrual disorders or overweight had normal thyroid function. Anti-thyroid peroxidase, anti-microsome and anti-th

Selenium and glutathione peroxidase levels in sickle cell anemia.

Levels of plasma selenium (Se) and glutathione peroxidase were measured in 20 sickle cell anemia (SCA) patients not in crisis and in 14 nonanemic control subjects. The results show that the levels of Se and glutathione peroxidase were significantly (p less than 0.005) lower than those of controls in both plasma and whole blood. These data are consistent with the previous reports that there is increased oxidative stress in SCA. Low blood Se levels and glutathione peroxidase activity observed in this research suggest that a weakened antioxidant potential may be associated with SCA patients. The

Glutathione oxidation during peroxidase catalysed drug metabolism.

The peroxidase catalyzed oxidation of certain drugs in the presence of glutathione (GSH) resulted in extensive oxidation to oxidized glutathione (GSSG). Extensive oxygen uptake ensued and thiyl radicals could be trapped. Only catalytic amounts of drugs were required indicating a redox cycling mechanism. Active drugs included phenothiazines, aminopyrine, p-phenetidine, acetaminophen and 4-N,N-(CH3)2-aminophenol. Other drugs, including dopamine and alpha-methyl dopa, did not catalyse oxygen uptake, nor were GSSG or thiyl radicals formed. Instead, GSH was depleted by GSH conjugate formation. Drug

Cytochemical demonstration of 'platelet' peroxidase at the light microscope level.

A reliable and reproducible modified method was developed for the cytochemical demonstration of the so-called 'platelet' peroxidase at the light microscopy (LM) level. Interestingly, not only normal platelets and megakaryocytes showed the peroxidase activity, but also normal lymphocytes. As with the ultrastructural platelet peroxidase (PPO) method, myeloperoxidase (MPO) was also demonstrated in granulocytes. Peroxidase activity was exhibited by blast cells of acute myeloid leukaemia, including megakaryoblasts of megakaryoblastic leukaemia, by hairy cells of hairy cell leukaemia, by centroblast

Immunological reaction to horseradish peroxidase of the palatine tonsil of the rabbit.

Detection of the immune response to external antigen of the palatine tonsil of the rabbit is an interesting problem. Employing horseradish peroxidase (HRP) as an external antigen, the reactions of a perfusion sensitized group and those of a group sensitized by intravenous injection were examined. The results were as follows: 1. Hypertrophy of the palatine tonsil was recognized only in the perfusion sensitized group. 2. The serum titer level of the antibody to HRP was recognized to be high in the intravenous injection sensitized group, which was confirmed by the production of antibody in the sp

Platelet peroxidase of circulating thrombocytes in acquired refractory anaemias.

The platelet peroxidase (PPO) content of circulating thrombocytes was determined in 10 healthy controls and in 18 cases of acquired refractory anaemia (2 with refractory anaemia, 4 with sideroblastic refractory anaemia, 8 with refractory anaemia with excess of blasts, 3 with refractory anaemia in transformation and 1 with chronic myelomonocytic leukaemia). The thrombocytes of the controls were invariably PPO-positive. No peroxidase deficiency was found in the 4 patients with sideroblastic refractory anaemia. Of the remaining 14 cases PPO-positive and PPO-negative thrombocytes coexisted in 8. O

Platelet peroxidase deficiency in a case of myelodysplastic syndrome with myelofibrosis.

Morphological and functional abnormalities of the megakaryocytic series have been well described in myelodysplastic syndromes. Platelet peroxidase has always been demonstrated in abnormal megakaryocytes and early megakaryoblasts in such syndromes. We have studied a case of myelodysplastic syndrome with marked morphological abnormalities of megakaryocytes in which ultrastructural studies showed the coexistence of platelet peroxidase positive and platelet peroxidase negative megakaryocytes. This enzymatic deficiency was confirmed by the ultrastructural study of circulating platelets. This case a

Glutathione peroxidase and glutathione S-transferase activity of platelets.

A low Se intake in dietetically treated patients with phenylketonuria (PKU) or maple syrup urine disease (MSUD) leads to a marked reduction of the platelet glutathione peroxidase activity (GSHPx). The mean value amounted to 2.0 U/10(11) platelets with t-butyl hydroperoxidase (t-BOOH) (2.2 U/10(11) with H2O2) in patients and 5.8 U/10(11) with t-BOOH (5.4 U/10(11) with H2O2) in the control children. After Se supplementation with yeast rich in Se (dose: 135 micrograms Se/m2) the GSHPx activities rapidly increased. They reached a plateau after 2-3 weeks and remained there during the following 15-2

Olfactory receptor cell staining using horseradish peroxidase.

Iontophoretic injection of horseradish peroxidase into severed olfactory nerve fascicles has been used to stain salamander olfactory receptor cell somata, their associated nerves, and their axonal terminations in the glomerular layer of the olfactory bulb. This technique gives homogeneous, Golgi-like staining of individually identifiable receptor cells and has permitted preliminary mapping of the topographical relationship between the loci of receptors in the olfactory mucosa and sites of their termination in the glomerular layer in the olfactory bulb.

[Superoxide dismutase, catalase and peroxidase of propionic acid bacteria].

The respiration of different species of propionic acid bacteria was found to be resistant to the inhibiting action of cyanide. Propionibacterium globosum, P. globosum (emm), P. vannielii, P. shermanii, P. petersonii, P. pentosaceum and P. raffinosaceum were shown to contain superoxide dismutase, catalase and peroxidase. The highest activity of catalase and superoxide dismutase was found in P. globosum and P. vannielii, the lowest one in P. shermanii and P. raffinosaceum. Peroxidase was detected in propionic bacteria for the first time.
