Intra-strain genetic heterogeneity in Toxoplasma gondii ME49: Oxford Nanopore long-read sequencing reveals copy number variation in the ROP8-ROP2A locus.

BACKGROUND: Toxoplasma gondii is an important pathogen and model organism for studying mechanisms of immune evasion and defense. Within the … mous variants included genes associated with T. gondii life cycle (AP2X-8) and virulence in vivo (6-phosphogluconate dehydrogenase). A 13 kb expansion in the ROP8-ROP2A virulence locus increased the copy number of ROP2A-ROP8 genes in isolates 2000B and 2020T from three to six. CONCLUSIONS: We provide an improved T. gondii ME49 reference genome and demonstrate the potentially co …

Impaired carbohydrate metabolism of polymorphonuclear leukocytes in glycogen storage disease Ib.

This study measures hexose monophosphate (HMP) shunt activity, glycolytic rate, and glucose transport in PMN and lymphocytes of patients with glycogen storage disease (GSD) type Ib as compared with controls and with GSD Ia patients. HMP shunt activity and glycolysis were significantly lower in intact PMN cells of GSD Ib patients as compared with GSD Ia patients and with controls. These activities were above normal levels in disrupted GSD Ib PMN. HMP shunt activity and glycolytic rates in lymphocytes were similar in all three groups studied. The rate of 2-deoxyglucose transport into GSD Ib PMN

Uptake and incorporation of glucose and mannose by whole cells of Bacteroides thetaiotaomicron.

Glucose uptake by whole-cell suspensions of the obligate anaerobe Bacteroides thetaiotaomicron was two- to fourfold higher under aerobic conditions than during incubation under atmospheres of N(2) or H(2) gas. The O(2)-stimulated uptake activity was lost rapidly (>70% in 5 h) when cell suspensions were incubated aerobically, but this loss was prevented by the addition of crude catalase. Catalase had no apparent effect on cell viability during these incubations. Glucose uptake activity was strongly inhibited by a 10-fold excess of mannose or galactose but not by methyl-alpha-d-glucoside, fructo

Effects of enzyme induction therapy on glucose and drug metabolism in obese mice model of non-insulin dependent diabetes mellitus.

We evaluated the effects of phenobarbital, an inducer, on plasma glucose and serum immunoreactive insulin levels and on hepatic glucose and drug metabolism using an animal model of non-insulin dependent diabetes mellitus. Genetically obese (ob/ob) mice, characterized by hyperglycaemia, hyperinsulinaemia, fatty liver and obesity were selected. The impairment of diabetic state with age was associated with increased activities of NADPH producing enzymes, whereas mixed function oxidase system remained unaltered. Phenobarbital reduced serum immunoreactive insulin and plasma glucose levels and decre

6PGD Upregulation is Associated with Chemo- and Immuno-Resistance of Renal Cell Carcinoma via AMPK Signaling-Dependent NADPH-Mediated Metabolic Reprograming.

BACKGROUND: The essential role of 6-phosphogluconate dehydrogenase (6PGD), the enzyme catalyzing the oxidative pentose phosphate pathway, in tumor growth and metabolism has garnered attention in recent years. In this work, we are the first to demonstrate that aberrant activation of 6PGD is a feature in renal cell carcinoma (RCC) and is critically involved in renal carcinogenesis and chemo- and immuno-resistance. MATERIALS AND METHODS: 6PGD expression and activity were systematically analyzed in normal and malignant renal cells and tissues. The roles of 6PGD and its downstream mechanism were i…

Prediction of the halothane (Hal) genotypes of pigs by deducing Hal, Phi, Po2, Pgd haplotypes of parents and offspring: results from a large-scale practice in Swedish breeds.

Results from a large-scale study, comprising 75 different breeding herds, are reported on predicting the halothane (Hal) genotypes of individual pigs by making use of the known close linkage between Hal and three electrophoretic blood marker loci (Phi, Po2, Pgd). The parents haplotypes (involving Hal and marker loci) were determined from the HAL phenotypes (halothane test results) and marker loci phenotypes of their offspring in the first one or two litters studied. In subsequent litters of the Hal-marker loci haplotyped parents, the offspring's expected Hal genotypes could be predicted on the

Since the liver is the central organ for me … were significantly increased alongside a paradoxical decrease in fructose-1,6-diphosphate (FDP). 6-phosphogluconate dehydrogenase (G6PD) was increased, and malic acid was elevated. Membrane characteristics and metabolic properties of glucose-6-phosphate dehydrogenase deficient red cells.

Two Finnish variants of reduced erythrocyte glucose-6-phosphate dehydrogenase (G-6-PD) activity were studied. The G-6-PD Espoo variant is characterized by severe enzyme deficiency which is normally non-haemolytic although primaquine sensitive. The other variant, G-6-PD Helsinki, in which the enzyme activity is moderately reduced, is associated with chronic haemolytic anaemia. The activity of the pentose phosphate pathway was not stimulated by methylene blue in G-6-PD Espoo cells, whereas in normal and G-6-PD Helsinki cells there were increases in shunt activity of 64.5- and 5.3-fold, respectiv

Characteristics and significance of the reverse glucose-6-phosphate dehydrogenase reaction.

Glucose-6-phosphate dehydrogenase (G-6-PD) is the first enzyme of the hexose monophosphate pathway, and this important reaction is often considered to be irreversible. However, its apparent irreversibility is caused by the rapid removal of the immediate product, 6-phosphoglucono-delta-lactone. We have now investigated the reverse G-6-PD reaction, namely, the oxidation of reduced nicotinamide-adenine dinucleotide phosphate (NADPH) by 6-phosphoglucono-delta-lactone to form glucose-6-phosphate and nicotinamide-adenine dinucleotide phosphate (NADP). The substrate of the reaction, 6-phosphoglucono-

Hemolytic anemia in hereditary pyrimidine 5'-nucleotidase deficiency. II. Effect of pyrimidine nucleotides and their derivatives on glycolytic and pentose phosphate shunt enzyme activity.

We evaluated the glycolytic intermediate concentrations from the erythrocytes of a patient with hereditary pyrimidine 5'-nucleotidase (P5'N) deficiency. Conclusive evidence for a metabolic block was not found. We evaluated the effects of the pyrimidine (cytidine and uridine) tri- and diphosphate nucleotides (CTP, CDP, UTP, UDP) and the choline and ethanolamine derivatives of CDP (CDP-choline, CDP-ethanolamine) on the activities of key enzymes of the Embden-Meyerhof pathway. Identification of blood groups and isoenzymic phenotypes from blood stains, heart blood and nervous tissues after transfusion.

A man aged 42 years died from a hemorrhage resulting from stab wounds in his neck, chest and abdomen. The MN blood group and PGM1 phenotype of the victim's heart blood were inconsistent with those of the blood stains around the victim. The victim was transfused with 4000 ml of blood during the 9 hours before his death. Because PGM1 phenotypes obtained from blood stains at the scene of the crime and from the weapon corresponded with those from the victim's dental pulp, peripheral nerve and bone marrow, the victim's blood groups and isoenzymic phenotypes could be determined accurately.
