Lyso-phosphatidylethanolamine primes the plant immune system and promotes basal resistance against hemibiotrophic pathogens.

BACKGROUND: Lyso-phosphatidylethanolamine (LPE) is a natural phospholipid that functions in the early stages of plant senescence. Plant innate immunity and early leaf senescence share molecular components. To reveal conserved mechanisms that link-up both processes, we tried to unravel to what extent LPE coordinates defense response and by what mode of action. RESULT: We found that LPE-treatment induces signaling and biosynthesis gene expression of the defensive hormone salicylic acid (SA). However, jasmonic acid and ethylene triggered gene induction levels are indistinguishable from the contr

Plant-based porcine reproductive and respiratory syndrome virus VLPs induce an immune response in mice.

Porcine reproductive and respiratory syndrome virus (PRRSV) significantly affects the swine industry worldwide. An efficient, protective vaccine is still lacking. Here, we report for the first time the generation and purification of PRRSV virus like particles (VLPs) by expressing GP5, M and N genes in Nicotiana silvestris plants. The particles were clearly visible by transmission electron microscopy (TEM) with a size of 60-70 nm. Hydrodynamic diameter of the particles was obtained and it was confirmed that the VLPs had the appropriate size for PRRS virions and that the VLPs were highly pure. B

Secreted Effector Proteins of Poplar Leaf Spot and Stem Canker Pathogen Sphaerulina musiva Manipulate Plant Immunity and Contribute to Virulence in Diverse Ways.

Fungal effectors play critical roles in manipulating plant immune responses and promoting colonization. Sphaerulina musiva is a heterothallic ascomycete fungus that causes Septoria leaf spot and stem canker disease in poplar (Populus spp.) plantations. This disease can result in premature defoliation, branch and stem breakage, increased mortality, and plantation failure. However, little is known about the interaction between S. musiva and poplar. Previous work predicted 142 candidate secreted effector proteins in S. musiva (SmCSEPs), 19 of which were selected for further functional characteriz

Ralstonia solanacearum type III effector RipV2 encoding a novel E3 ubiquitin ligase (NEL) is required for full virulence by suppressing plant PAMP-triggered immunity.

Ralstonia solanacearum causes bacterial wilt disease in a broad range of plants, primarily through type Ⅲ secreted effectors. However, the R. solanacearum effectors promoting susceptibility in host plants remain limited. In this study, we determined that the R. solanacearum effector RipV2 functions as a novel E3 ubiquitin ligase (NEL). RipV2 was observed to be locali in the plasma membrane after translocatio into plant cells. Transient expression of RipV2 in Nicotiana benthamiana could induce cell death and suppress the flg22-induced pathogen-associated molecular pattern (PAMP)-triggered immun

Use of Yoghurt Enhanced with Volatile Plant Oils Encapsulated in Sodium Alginate to Increase the Human Body's Immunity in the Present Fight Against Stress.

(1) Background: The COVID-19 pandemic and the imposition of strict but necessary measures to prevent the spread of the new coronavirus have been, and still are, major stress factors for adults, children, and adolescents. Stress harms human health as it creates free radicals in the human body. According to various recent studies, volatile oils from various aromatic plants have a high content of antioxidants and antimicrobial compounds. An external supply of antioxidants is required to destroy these free radicals. The main purpose of this paper is to create a yoghurt with high antioxidant capaci

Pathogen Effector-Mediated Reprogramming of Plant Alternative Splicing: From Immune Regulation to Crop Disease Resistance.

Plants have evolved sophisticated immune systems to defend against diverse pathogens, whereas pathogens deploy effectors to manipulate host cellular processes for successful infection. Recent studies have revealed that pathogen effectors can target host RNA processing, particularly pre-mRNA alternative splicing (AS), to reshape transcript isoform profiles and modulate plant immunity. However, the common principles and specific differences among pathogen effector-mediated regulation of host AS and their impacts on plant immunity have not been systematically summarized. In this review, we summar…

Medicinal and Edible Plants for the Management of Inflammatory Bowel Disease: Synergistic Modulation of the Microbiota-Metabolite-Immune Axis.

Inflammatory bowel disease (IBD) is a chronic condition driven by the intertwined interplay of gut dysbiosis, immune perturbation, and metabolic imbalance. Current single-target therapies often fail to durably restore homeostasis in this complex ecosystem, and have thereby increased interest in diet-based holistic interventions. Plants traditionally used as both food and medicine, such as Crataegus pinnatifida, Ganoderma lucidum, and Codonopsis pilosula, are natural products that possess both nutritional and therapeutic attributes. The use of such plants is commonly referred to as medicinal fo

An AMA1/MSP119 Adjuvanted Malaria Transplastomic Plant-Based Vaccine Induces Immune Responses in Test Animals.

Malaria is a tropical human disease, caused by protozoan parasites, wherein a significant number of the world's population is at risk. Annually, more than 219 million new cases are reported. Although there are prevention treatments, there are no highly and widely effective licensed anti-malarial vaccines available for use. Opportunities for utilization of plant-based vaccines as novel platforms for developing safe, reliable, and affordable treatments offer promise for developing such a vaccine against malaria. In this study, a Malchloroplast candidate vaccine was designed, composed of segments

Plant immune responses - from guard cells and local responses to systemic defense against bacterial pathogens.

When plants are infected by pathogens two distinct responses can occur, the early being a local response in the infected area, and later a systemic response in non-infected tissues. Closure of stomata has recently been found to be a local response to bacterial pathogens. Stomata closure is linked to both salicylic acid (SA), an essential hormone in local responses and systemic acquired resistance (SAR), and absisic acid (ABA) a key regulator of drought and other abiotic stresses. SAR reduces the effects of later infections. In this review we discuss recent research elucidating the role of guar

Arabidopsis BRCA2 and RAD51 proteins are specifically involved in defense gene transcription during plant immune responses.

Systemic acquired resistance (SAR) is a plant immune response associated with both transcriptional reprogramming and increased homologous DNA recombination (HR). SNI1 is a negative regulator of SAR and HR, as indicated by the increased basal expression of defense genes and HR in sni1. We found that the sni1 phenotypes are rescued by mutations in BREAST CANCER 2 (BRCA2). In humans, BRCA2 is a mediator of RAD51 in pairing of homologous DNA. Mutations in BRCA2 cause predisposition to breast/ovarian cancers; however, the role of the BRCA2-RAD51 complex in transcriptional regulation remains unclear

Potyviral genome-linked protein targets NbMYB330 to attenuate hydrogen-peroxide-induced antiviral immunity in plants.

MYB transcription factors play crucial roles in orchestrating plant immunity against various pathogens. However, their regulation during virus infection and how viruses counteract these defenses remain poorly understood. In this study, we demonstrated that turnip mosaic virus (TuMV) viral genome-linked protein (VPg) interacts with Nicotiana benthamiana MYB330 (NbMYB330), an R2R3-MYB transcription factor, in the nucleus. NbMYB330 expression was activated at the early stage of TuMV infection, and NbMYB330 positively regulated host antiviral defense. NbMYB330 directly bound to the promoter region…

Immune-enhancing activity of a plant-derived acid polysaccharide LCAP on influenza and respiratory syncytial virus vaccines.

Polysaccharides, given their immunomodulating properties, biocompatibility, and biodegradability, have been extensively studied for their potential as vaccine adjuvants. The structural characterization of polysaccharides and their application as vaccine adjuvants have garnered significant attention. In this study, an acidic polysaccharide, LCAP, was isolated from a natural plant, and its immune-enhancing activity as a vaccine adjuvant was investigated. LCAP was obtained through aqueous extraction, protein removal, and column chromatography purification, with a purity of approximately 94%. The

CsWRKY33 Integrates Immune Signalling and Metabolic Reprogramming to Enhance Tea Plant Resistance Against Colletotrichum camelliae.

Tea anthracnose, caused by Colletotrichum camelliae, poses a serious threat to tea yield and quality. While certain transcription factors have been implicated in stress responses, the core transcriptional networks governing immunity in tea plants remain largely elusive. In this study, we identify CsWRKY33 as a central regulator that orchestrates immune responses and metabolic reprogramming during C. camelliae infection in tea plants. Transcriptome analysis revealed that CsWRKY33 is significantly induced upon fungal challenge and functions downstream of both pattern-triggered immunity (PTI) and

Protein nanoparticles assemble in plants, display antigenic viral peptides, and produce an epitope-specific immune response.

Porcine reproductive and respiratory syndrome (PRRS) is one of the major diseases affecting the global swine industry. Current methods to control this viral disease are insufficient or pose safety concerns; therefore, there is a need for a safer and more effective vaccine against PRRS. In this study, we designed a chimeric nanoparticle vaccine through genetic fusion of an epitope composed of portions from two PRRS viral proteins, M and GP5, with lumazine synthase from Aquifex aeolicus. Transient expression in the leaves of Nicotiana benthamiana plants resulted in soluble levels around 0.18 mg·

The Ralstonia solanacearum type III effector RipAY targets plant redox regulators to suppress immune responses.

The subversion of plant cellular functions is essential for bacterial pathogens to proliferate in host plants and cause disease. Most bacterial plant pathogens employ a type III secretion system to inject type III effector (T3E) proteins inside plant cells, where they contribute to the pathogen-induced alteration of plant physiology. In this work, we found that the Ralstonia solanacearum T3E RipAY suppresses plant immune responses triggered by bacterial elicitors and by the phytohormone salicylic acid. Further biochemical analysis indicated that RipAY associates in planta with thioredoxins fro

Protective immunity of plant-produced African horse sickness virus serotype 5 chimaeric virus-like particles (VLPs) and viral protein 2 (VP2) vaccines in IFNAR-/- mice.

Next generation vaccines have the capability to contribute to and revolutionise the veterinary vaccine industry. African horse sickness (AHS) is caused by an arbovirus infection and is characterised by respiratory distress and/or cardiovascular failure and is lethal to horses. Mandatory annual vaccination in endemic areas curtails disease occurrence and severity. However, development of a next generation AHSV vaccine, which is both safe and efficacious, has been an objective globally for years. In this study, both AHSV serotype 5 chimaeric virus-like particles (VLPs) and soluble viral protein
