Visualization of Mouse Choroidal and Retinal Vasculature Using Fluorescent Tomato Lectin Perfusion.

PURPOSE: To develop a reliable and simplified method to assess choroid and retinal vasculature on whole mount and cross sections in mice using tomato lectin (TL; Lycopersicon esculentum). METHODS: Albino mice (n = 27) received 1 mg/mL of TL (conjugated to Dylight-594) intravascularly through the tail vein, jugular vein, or cardiac left ventricle. Whole mounts of the retina and choroid were evaluated using fluorescence microscopy. Perfusion with GSL-IB4 conjugated to Dylight-594 and fluorescein isothiocyanate was performed to compare against labeling with TL. Co-labeling of choroidal endotheli

Crystal structure and glycan-binding ability of galectin domain of OsGALT1.

N- linked glycan structures vary across different organisms, and Lewis A glycan represents a unique N-glycoform in plants. The GT31 enzyme GALT1 catalyzes Lewis A glycan synthesis by transferring galactose from UDP-galactose to the terminal GlcNAc of N-glycan branches. Structurally, the plant GALT1 protein is characterized by the presence of both a galactosyltransferase catalytic domain and a galectin-like domain. However, the specific ligands recognized by its plant-specific galectin domain, as well as the functional role of this domain in catalysis, remain poorly understood. In this study, w

DVL, a Lectin from Dioclea violacea Seeds, Disturbs the Proteomic Profile of Candida krusei, Leading to Cell Death.

Background/Objectives Plant lectins have emerged as potential antifungal molecules, where the carbohydrate recognition domain (CRD) is possibly the main mode of action of these proteins. Previously, we saw that the lectin extracted from the seeds of Dioclea violacea (DVL) has anti-candida activity against Candida krusei cells by acting to inhibit ergosterol biosynthesis, cell wall deformation, and deregulation of the redox system. Methods We have now confirmed this anti-candida activity by proteomic analysis, with the expression of proteins that show us how C. krusei cells respond to this trea…

Kaposi's sarcoma in AIDS: basement membrane and endothelial cell markers in late-stage lesions.

Early-stage lesions of Kaposi's sarcoma (KS) are composed of single-layered, highly flattened cells lining collagen bundles, whereas late-stage lesions contain densely packed, spindle-shaped cells. We examined the progression of KS lesions in oral mucosa and lymph nodes from patients with AIDS, using antibodies specific for blood vascular endothelial cells (Factor VIII-related antigen) and their basement membrane (Type IV collagen and laminin). In addition, the plant lectin Ulex europaeus, which selectively stains blood vessels, was also used. In early-stage KS lesions, fibronectin, laminin an

A comparison of established human lymphoma lines by flow cytometry: quantitation of Ricinus communis agglutinin binding and the effect of specific glycosidases.

Two established cell lines of human B-cell lymphomas derived from Burkitt lymphomas and their Epstein-Barr virus-transformed counterparts were analyzed with respect to their ability to bind the beta-galactoside-specific lectin Ricinus communis agglutinin (RCA). Native and sialidase- as well as sialidase-beta-galactosidase-treated cells were compared. The method for the quantitative determination of average numbers of binding sites and of apparent affinity constants was flow cytometry with fluorescence-labeled lectin. Although with native cells there was no significant deviation of the values f

Cross-Talk Between a Nematode Effector-Targeted Metabolic Enzyme and a Lectin Receptor Kinase Regulates Wheat Immunity.

The cereal cyst nematode (Heterodera avenae) is an obligate plant parasite that severely threatens wheat production. Previous studies have shown that its secreted effector HaGLAND5, suppresses host immunity and promotes nematode parasitism by enhancing acetylation at lysine 39 of TaEMB3003, the E2 subunit of the wheat pyruvate dehydrogenase complex. However, how this molecular interaction precisely regulates the immune response remains unclear. In this study, we identified a wheat L-type lectin receptor kinase, TaLecRK-S.4, and demonstrated its interaction with TaEMB3003. Genetic analyses show

Novel biopesticide based on a spider venom peptide shows no adverse effects on honeybees.

Evidence is accumulating that commonly used pesticides are linked to decline of pollinator populations; adverse effects of three neonicotinoids on bees have led to bans on their use across the European Union. Developing insecticides that pose negligible risks to beneficial organisms such as honeybees is desirable and timely. One strategy is to use recombinant fusion proteins containing neuroactive peptides/proteins linked to a 'carrier' protein that confers oral toxicity. Hv1a/GNA (Galanthus nivalis agglutinin), containing an insect-specific spider venom calcium channel blocker (ω-hexatoxin-Hv

Insecticidal activity of scorpion toxin (ButaIT) and snowdrop lectin (GNA) containing fusion proteins towards pest species of different orders.

BACKGROUND: The toxicity of a fusion protein, ButalT/GNA, comprising a venom toxin (ButaIT) derived from the red scorpion, Mesobuthus tamulus (F.), and Galanthus nivalis agglutinin (GNA), was evaluated under laboratory conditions against several pest insects. Insecticidal activity was compared with SFI1/GNA, a fusion comprising a venom toxin (SFI1) derived from the European spider Segestria florentina (Rossi) and GNA, which has been previously demonstrated to be effective against lepidopteran and hemipteran pests, and to GNA itself. RESULTS: Injection assays demonstrated that both fusion prot

The Golgi apparatus of goblet cells in the mouse descending colon: three-dimensional visualization using a confocal laser scanning microscope.

The three-dimensional structure of the Golgi apparatus was studied in goblet cells in lectin-stained sections of the mouse descending colon by using a confocal laser scanning microscope. In the lower part of the crypt, the Golgi apparatus formed a dome- or globe-like structure in the supranuclear region. The wall of the dome had some holes, one of which usually faced toward the nucleus and others toward the apical cytoplasm. Mucous granules seemed to be initially released into the interior of the dome and transported toward the apical cytoplasm through the holes. In the upper part of the crypt

Lectin binding to olfactory system in a shark, Scyliorhinus canicula.

Lectin histochemical studies were performed on the olfactory system of Scyliorhinus canicula to identify specific glycoconjugates on the cell surface of primary olfactory neurons. The olfactory receptor cells, the olfactory nerve fibers and their terminals in the bulbs were labelled with SBA, BSA-I and BSA-I-B4. The lectin staining patterns indicate that the membranes of small-spotted catshark olfactory neurons had glycoproteins with alpha-galactose residues. This carbohydrate moiety could be related to modulation of the cell-cell interactions in the olfactory system.

Jack fruit lectin binding pattern in benign and malignant lesions of the breast.

N-acetyl D-galactosamine specific lectin was isolated from Jack fruit (Artocarpus integrifolia) and conjugated to horse radish peroxidase type VI. The purified conjugate was used for the study of tissue binding properties on benign and malignant lesions of the breast using diaminobenzidine as substrate on dewaxed tissue sections. Forty mammary carcinomas, 10 cystic hyperplasias of the breast and 10 normal breast tissues were used for the study. Neoplastic cells showed increased affinity to the lectin. The lectin binding was focally strong in neoplastic cells compared to the normal as well as t

Regional differences in normal and cancer-associated glycoconjugates of the human colon.

Because inherent regional differences in colonic epithelium may determine the biologic behavior of tumors originating from different sites, and inasmuch as colonic epithelial cells secrete mucins that may reflect the state of cell differentiation, colonic goblet cell mucin was analyzed with the use of fluorescein isothiocyanate-conjugated lectins and fluorescence microscopy in normal fetal and adult mucosa and in cancers of the proximal and distal colon. In the adult proximal colon only the goblet cell mucin in the upper portion of the crypts was specifically labeled by the lectin Dolichos bif

The significance of lectin receptors for the evaluation of hormone dependence in breast cancer.

Benign and malignant breast diseases were histochemically investigated with regard to their secretory function. Thereby, the labelled lectin from peanut (peanut agglutinin = PNA) which possesses a high affinity for D-galactosyl-(1-3)-N-acetyl-D-galactosamine, showed a binding pattern partly indicating disturbed secretory cell mechanisms. Further studies with a labelled antibody directed against a milk fat globule membrane glycoprotein, which was isolated by PNA affinity chromatography from human milk, confirmed the PNA receptor as a marker of a milk protein in breast carcinomas. Additionally,

Protein O-linked glycosylation in the plant pathogen Ralstonia solanacearum.

Ralstonia solanacearum is one of the most lethal phytopathogens in the world. Due to its broad host range, it can cause wilting disease in many plant species of economic interest. In this work, we identified the O-oligosaccharyltransferase (O-OTase) responsible for protein O-glycosylation in R. solanacearum. An analysis of the glycoproteome revealed that 20 proteins, including type IV pilins are substrates of this general glycosylation system. Although multiple glycan forms were identified, the majority of the glycopeptides were modified with a pentasaccharide composed of HexNAc-(Pen)-dHex(3),

Localization of rabies virus glycoprotein into the endoplasmic reticulum produces immunoprotective antigen.

Rabies virus surface glycoprotein (rabies G-protein) with (G+RS) and without (G-RS) endoplasmic reticulum retrieval signal was expressed and characterized in tobacco plants. Transgenically expressed rabies G-protein was estimated at 0.015-0.38 % of total leaf protein. The relative migration of the rabies G-protein on SDS-PAGE was at the position, as anticipated for the viral coat protein (~66 kDa). Immunolocalization by confocal microscopy established that immunoprotective G+RS expressed in tobacco was primarily confined to ER. G+RS showed binding to Con A lectin and was susceptible to N-glyco

Glycan Signatures on Neutrophils in an Equine Model for Autoimmune Uveitis.

Glycosylation of surface proteins is a crucial post-translational modification that reflects the activation status of neutrophils, the predo … rom healthy horses and the potential changes in surface glyco-signatures in ERU. Using 35 different plant lectins, we outlined a profile of surface-exposed glycan moieties on equine neutrophils and detected significantly increased O-glycosylation in a diseased state through Jacalin (JAC) binding via flow cytometry. Cytochemical study of the germinal membrane of the Echinococcus granulosus cyst.

The present cytochemical study was undertaken to provide more information on the localization of enzymatic and glycoconjugates in the germinal membrane of the Echinococcus granulosus cyst. The distinctive distribution of binding sites for two lectins (peanut agglutinin and Dilochos biflorus agglutinin) in the germinal membrane are described. An investigation is made of the distribution and specific activity of adenosine triphosphatase, alkaline phosphatase and acid phosphatase. The results suggest that cells located in the deeper layer of the germinal membrane are intrinsic in the cellular dif

Ultrastructural lectinocytochemistry of fowl plague virus-infected and uninfected MDCK cells.

Using horseradish peroxidase (HRP)-conjugated lectins for pre-embedding labelling we have shown differences in ultrastructural localization of saccharides in cell compartments of fowl plague (FP) virus-infected and uninfected MDCK cells. Lectinochemical staining of the cell compartments in the case of FP virus-infected MDCK cells was less intensive as compared with uninfected cells. Also certain differences in the staining of subcompartments of cell organells were seen. Staining of uninfected cells with Pisum sativum agglutinin (PSA)-HRP revealed an extensive visualization of Golgi complex, ma

Immunohistochemical examination of the relationship between two types of mucous neck cell and the intermediate cells in the rat fundic gland.

It is generally accepted that the mucous neck cells are the precursors of chief cells, and that they are converted to chief cells via intermediate cells. We reported previously that two types of mucous neck cell are present in the region close to the cardia of the rat fundic gland. Using immunoelectron microscopy and lectin histochemistry, we examined whether the two histochemically distinct types of mucous neck cell are converted to chief cells via intermediate cells by the same process. Intermediate cells were positive for immunostaining with an antibody raised against neutral mucin but nega
