The Receptor for Advanced Glycation Endproducts (RAGE) Contributes to Severe Inflammatory Liver Injury in Mice.

Background: The receptor for advanced glycation end products (RAGE) is a multiligand receptor involved in a number of processes and disorders. While it is known that RAGE-signaling can contribute to toxic liver damage and fibrosis, its role in acute inflammatory liver injury and septic multiorgan failure is yet undefined. We examined RAGE in lipopolysaccharide (LPS) induced acute liver injury of D-galN sensitized mice as a classical model for tumor necrosis factor alpha (TNF-α) dependent inflammatory organ damage. Methods: Mice (Rage-/- and C57BL/6) were intraperitoneally injected with D-galN

Effects of epigallocatechin-3-gallate on the HMGB1/RAGE pathway in PM2.5-exposed asthmatic rats.

Current studies have shown that long-term exposure to fine particulate matter (PM2.5) can aggravate lung injury in asthmatic children. The HMGB1/RAGE pathway may play an important role, but few studies on the HMGB1/RAGE signaling pathway in PM2.5-induced asthma have been performed. Epigallocatechin-3-gallate (EGCG), which has antioxidant, anti-inflammatory and immunomodulatory effects, has not been examined in studies at home and abroad. In this study, we established an animal model of asthma and observed that the lung tissue was damaged, inflammatory cells infiltrated, bronchial wall thicknes

Expression pattern of RAGE and IGF-1 in the human fetal ovary and ovarian serous carcinoma.

The expression pattern of RAGE and IGF-1 proteins in different ovarian cell lineages was histologically analyzed in six fetal, nine adult human ovaries, and nine serous ovarian carcinomas (OSC) using immunohistochemical methods. Mild expression of IGF-1 in ovarian surface epithelium (Ose) and oocytes in the 15-week human ovaries increased to moderate or strong in the stromal cells, oocytes and follicular cells in week 22. Occasional mild RAGE expression was observed in Ose during week 15, while strong expression characterized primordial follicles in week 22. In the reproductive human ovary, IG

Expression of RAGE and HMGB1 in thymic epithelial tumors, thymic hyperplasia and regular thymic morphology.

Recently, a role of the receptor for advanced glycation endproducts (RAGE) in myasthenia gravis was described. RAGE and its ligand high mobility group box 1 (HMGB1) play key roles in autoimmunity and cancer. To test whether these molecules are involved in patients with thymic abnormalities we applied immunohistochemical analysis in 33 cases of thymic epithelial tumors, comprising 27 thymomas and 6 thymic carcinomas, and 21 nonneoplastic thymuses. Both molecules were detected in neoplastic epithelial cells: RAGE staining was most intense in WHO type B2 thymomas and thymic carcinomas (p<0.001).

Abeta-dependent Inhibition of LTP in different intracortical circuits of the visual cortex: the role of RAGE.

Oligomeric amyloid-beta (Abeta) interferes with long-term potentiation (LTP) and cognitive processes, suggesting that Abeta peptides may play a role in the neuronal dysfunction which characterizes the early stages of Alzheimer's disease (AD). Multiple lines of evidence have highlighted RAGE (receptor for advanced glycation end-products) as a receptor involved in Abeta-induced neuronal and synaptic dysfunction. In the present study, we investigated the effect of oligomeric soluble Abeta1-42 on LTP elicited by the stimulation of different intracortical pathways in the mouse visual cortex. A vari

The receptor for advanced glycation end products (RAGE) system in women with intraamniotic infection and inflammation.

OBJECTIVE: Receptor for advanced glycation end products (RAGE) is a multiligand cell-surface receptor part of the immunoglobulin superfamily with crucial roles in inflammation. S100A12/ENRAGE, a biomarker of amniotic fluid (AF) inflammation, is a ligand for RAGE. sRAGE, a competitive soluble RAGE receptor, inhibits RAGE ligands. Here we aimed to investigate the presence and changes in components of the RAGE system in women with intra-amniotic infection and inflammation. STUDY DESIGN: AF was retrieved by amniocentesis in 113 women stratified as follows: (1) positive AF culture (+AFC; GA = 27 [

Inhibition of the RAGE products increases survival in experimental models of severe sepsis and systemic infection.

INTRODUCTION: The receptor for advanced glycation end products (RAGE), a multi-ligand member of the immunoglobulin superfamily, contributes to acute and chronic disease processes, including sepsis. METHODS: We studied the possible therapeutic role of RAGE inhibition in the cecal ligation and puncture (CLP) model of polymicrobial sepsis and a model of systemic listeriosis using mice genetically deficient in RAGE expression or mice injected with a rat anti-murine RAGE monoclonal antibody. RESULTS: The 7-day survival rates after CLP were 80% for RAGE-/- mice (n = 15) (P < 0.01 versus wild-type)

RAGE signaling in acute myocardial infarction: sRAGE as a dynamic biomarker and emerging therapeutic target.

The receptor for advanced glycation end products (RAGE) is a multiligand pattern-recognition receptor involved in inflammation, oxidative stress, and cardiometabolic disease. In acute myocardial infarction (AMI), activation of the AGE-RAGE signaling axis contributes to ischemia-reperfusion injury and post-infarction cardiac remodeling. Its soluble isoform (sRAGE) has emerged as a potential circulating biomarker related to RAGE pathway activity. Experimental studies support a role for RAGE in amplifying myocardial injury through inflammatory and oxidative mechanisms. In clinical settings, circu

The RAGE Pathway in Skin Pathology Development: A Comprehensive Review of Its Role and Therapeutic Potential.

The receptor for advanced glycation end-products (RAGE), a member of the immunoglobulin superfamily, is expressed in various cell types and mediates cellular responses to a wide range of ligands. The activation of RAGE triggers complex signaling pathways that drive inflammatory, oxidative, and proliferative responses, which are increasingly implicated in the pathogenesis of skin diseases. Despite its well-established roles in conditions such as diabetes, cancer, and chronic inflammation, the contribution of RAGE to skin pathologies remains underexplored. This review synthesizes current finding

Normalized intraplaque hemorrhage signal on MP-RAGE as a marker for acute ischemic neurological events.

PURPOSE: This study sought to validate whether the signal intensity ratio (SIR) of carotid intraplaque hemorrhage (IPH) was associated with acute ischemic neurologic events. METHODS: A retrospective review was completed of consecutive patients that underwent neck magnetic resonance angiography using magnetization prepared-rapid gradient echo (MP-RAGE) and T1-CUBE sequences between 2017 and 2020. Patients with magnetic resonance evidence of IPH were included. SIRs were measured by comparing the maximum IPH signal with the mean intramuscular signal from the adjacent sternocleidomastoid. The Roof is Leaking and a Storm is Raging: Repairing the Blood-Brain Barrier in the Fight Against Epilepsy.

A large body of evidence that has accumulated over the past decade strongly supports the role of both blood-brain barrier (BBB) dysfunction and perivascular inflammation in the pathophysiology of epilepsy. Recent preclinical studies indicate that prolonged seizure- or brain injury-induced BBB dysfunction and subsequent perivascular inflammation may play an important role in post-traumatic epileptogenesis. In turn, perivascular inflammation can further sustain BBB dysfunction. In genetic epilepsies, such as tuberous sclerosis complex and other related epileptogenic developmental pathologies, th

Characterization of advanced glycation end products and their receptor (RAGE) in an animal model of myocardial infarction.

Circulating advanced glycation end products (AGE) and their receptor, RAGE, are increased after a myocardial infarction (MI) episode and seem to be associated with worse prognosis in patients. Despite the increasing importance of these molecules in the course of cardiac diseases, they have never been characterized in an animal model of MI. Thus, the aim of this study was to characterize AGE formation and RAGE expression in plasma and cardiac tissue during cardiac remodeling after MI in rats. Adult male Wistar rats were randomized to receive sham surgery (n = 15) or MI induction (n = 14) by lef

Periodontal disease and type 2 diabetes mellitus: is the HMGB1-RAGE axis the missing link?

Periodontitis is a major chronic inflammatory disease associated with increased production of numerous proinflammatory cytokines, which leads to the destruction of the periodontal tissue and ultimately loss of teeth. Periodontitis has powerful and multiple influences on the occurrence and severity of systemic conditions and diseases, such as diabetes mellitus, cardiovascular disease and respiratory disease. Meanwhile, diabetes is associated with increased prevalence, severity and progression of periodontal disease. There is also abundant evidence showing that diabetes plays important etiologic

The behavioral organization, temporal characteristics, and diagnostic concomitants of rage outbursts in child psychiatric inpatients.

Angry outbursts, sometimes called rages, are a major impetus for the psychiatric hospitalization of children. In hospitals, such outbursts are a management problem and a diagnostic puzzle. Among 130 4- to 12-year-olds successively admitted to a child psychiatry unit, those having in-hospital outbursts were likely to be younger, have been in special education, have had a preadmission history of outbursts, and to have a longer hospital stay. Three subsets of behaviors, coded as they occurred in 109 outbursts, expressed increasing levels of anger; two other subsets expressed increasing levels of

HMGB1-Induced Metabolic Rewiring of Macrophages via RAGE/MAPK Fosters Endocrine Resistance in Breast Cancer.

Endocrine resistance remains a critical challenge in hormone receptor-positive (HR+) breast cancer. The impact of the immune microenvironment on endocrine therapy efficacy is increasingly recognised, but its mechanisms are not fully understood. Here, we investigated that nuclear HMGB1 expression in tumour cells was closely associated with endocrine therapy resistance in HR+ breast cancer using tumour genomic databases. Specially, tumours with high HMGB1 expression induced M2-like macrophage polarisation, which was related to HMGB1 paracrine signalling. Mechanistically, HMGB1 bound to the RAGE

Alogliptin attenuates diabetes-associated cognitive impairment in rats via HMGB1/RAGE/TLR4-NFκB pathway modulation.

Chronic hyperglycemia associated with diabetes mellitus (DM) induces systemic metabolic disturbances that progressively promote microvascular and macrovascular complications including the impairment of neuronal integrity and cognitive function. Accumulating evidence has supported that cognitive impairment and Type 2 Diabetes Mellitus (T2DM) share common pathological mechanisms. The current investigation explored potential benefits of alogliptin, a dipeptidyl peptidase IV (DPP-4) inhibitor, on diabetes induced cognitive impairment by revealing its role in attenuating neuroinflammatory pathways.

Rosiglitazone alleviates Parkinson's disease phenotypes of Drosophila via dCTP-mediated inhibition of AGE-RAGE pathway.

BACKGROUND: Parkinson's disease (PD) is an aging-related neurodegenerative disorder, which threatens the health and life of millions of patients worldwide, but remains incurable. Developing novel interventive strategy is urgent yet unmet. In recent years, it was well accepted that pathogenesis of PD closely correlated with aberrant glucolipid metabolism. The antidiabetic drug, Rosiglitazone (Rosi), one typical PPARγ agonist was found to exert neuroprotective effect. However, whether Rosi can rescue the PD-related phenotypes and the underlying mechanisms remain to be elucidated. METHODS: In pr

Decreased adipokine CTRP4 in CAD patients: CTRP4 attenuates atherosclerosis via inhibition of RAGE and TLR4.

BACKGROUND: C1q/TNF-related proteins (CTRPs) belong to the adipokine family. Here, we aimed to assess the relation of CTRP4 levels in serum and perivascular adipose tissue (PVAT) with coronary artery disease (CAD), and investigate the effect of CTRP4 on atherosclerosis and the underlying mechanisms. METHODS: CTRP4 levels were examined in serum and epicardial adipose tissue (a major PVAT) from patients with CAD. Atherosclerotic lesions were analysed in CTRP4‒/‒/ApoE‒/‒ and ApoE‒/‒ mice. The paracrine effects of CTRP4 on atherosclerosis were tested by supplementation with CTRP4, either via PVAT
