Differential regulation of IgG-NMO autoantibodies on S100Beta protein and disability in relapsing neuromyelitis optica.

Neuromyelitis optica (NMO), an uncommon central nervous system (CNS) demyelinating disease, produces transverse myelitis and severe optic neuritis. IgG-NMO autoantibody, a specific immunoglobulin binding aquaporin-4 water channel protein, confirms that NMO is a different entity to multiple sclerosis. Parallel to cytokine down-regulations found in serum of relapsing-NMO (rNMO) patients, it has been reported that IgG-NMO may also confer a worse course of the disease in r-NMO Caribbean patients. In this study, we were interested in exploring the influence of IgG-NMO autoantibody on S100beta level

Neuropsychological function in children with cyanotic heart disease undergoing corrective cardiac surgery: effect of two different rewarming strategies.

OBJECTIVE: Hypothermia conventionally used in cardiopulmonary bypass necessitates rewarming to normothermic temperatures, which has been shown to be associated with neuropsychological injury. We studied the effects of two different rewarming strategies on postoperative neuropsychological function in cyanotic paediatric patients undergoing elective primary intracardiac repair of tetralogy of Fallot with the aid of cardiopulmonary bypass. METHODS: This was a randomised clinical study undertaken in the cardiothoracic centre of a tertiary level referral and teaching hospital. Eighty children, age

Developmental anomalies of the enteric nervous system in normoganglionic segments of bowel from rats with total colonic aganglionosis.

Polysialyated neural cell adhesion molecule (PSA-NCAM) is a marker for immature neurons and S100 beta is a known marker for enteric glia. The aim of this study was to determine the maturation of the enteric nervous system (ENS) in normoganglionic (Ng) bowel from rats with total colonic aganglionosis (TCA). Ng ileum was obtained from TCA rats (spotting lethal: mutant, sl/sl: n = 15) at 10, 19, and 24 days of age (n = 5 for each age). Normal control Ng ileum was obtained from wild type littermates (+/+, n = 25) at 10, 19, 24, 30, and 60 days of age (n = 5 for each age). Specimens were studied wi

A glial-derived protein, S100B, in neonates and infants with congenital heart disease: evidence for preexisting neurologic injury.

UNLABELLED: The glial-derived protein S100B is a serum marker of cerebral ischemia and correlates with negative neurological outcome after cardiopulmonary bypass (CPB) in adults. We sought to characterize the S100B release pattern before and after CPB in neonates and infants with congenital heart disease and correlate it with surgical mortality. Serum was collected before surgery and at 24 postoperative h from 109 neonates and infants with congenital heart disease. All patients had presurgical transthoracic echocardiograms and CPB with or without hypothermic circulatory arrest. S100B concentra

Bergmann glial S100B activates myo-inositol monophosphatase 1 and Co-localizes to purkinje cell vacuoles in SCA1 transgenic mice.

Spinocerebellar ataxia-1 (SCA1) is a late onset neurodegenerative disease caused by the expansion of a polyglutamine repeat within ataxin-1 protein. The toxic effects triggered by mutant ataxin-1 result in degeneration of the neurons in cerebellum, brain stem and spinocerebellar tracts. The targeted overexpression of mutant ataxin-1 in cerebellar Purkinje cells (PCs) of the SCA1 transgenic mice results in the formation of cytoplasmic vacuoles in PCs. These vacuoles appear early on before the onset of behavioral abnormalities. Interestingly, we found that vacoules contain S100B and vimentin pro

The label-free electrochemical S100B-TBI immunosensor for sensitive detection of S100B protein as a biomarker for traumatic brain injury using FFT-SWV voltammetry.

S100B protein is an established biomarker related to prognosis in traumatic brain injury (TBI). Developing sensitive methods for the serum S100B quantification is clinically valuable. This paper proposes a simple label-free electrochemical TBI-immunosensor for the timely detection of S100B in the sera of TBI patients. In the first step of bioelectrode fabrication, gold nanoparticles-reduced graphene oxide (AuNPs-rGO) nanocomposite was layered onto a glassy carbon electrode (GCE) to improve the amplification of electrochemical signals. The anti-S100B was immobilized onto a modified-GCE via self

Longitudinal Kinetics of Cerebrospinal Fluid and Serum S100B in Patients with Delayed Cerebral Ischemia After Aneurysmal Subarachnoid Hemorrhage.

S100 calcium-binding protein B (S100B) has been proposed as a biomarker of brain injury after aneurysmal subarachnoid hemorrhage (aSAH), although the clinical utility of serial measurements remains uncertain. This prospective observational study investigated whether longitudinal serum and cerebrospinal fluid (CSF) S100B concentrations are associated with delayed cerebral ischemia (DCI), inflammatory activation, and neurological outcome. Twenty-six consecutive patients with aSAH underwent serial serum and CSF S100B measurements throughout hospitalization. Associations between S100B concentratio

Integrated Multi-Omics and Radiogenomic Analysis Identifies S100B and ITGB5 as Complementary Candidate Biomarkers of Glioblastoma Heterogeneity.

Glioblastoma (GBM) is characterized by marked intratumoral heterogeneity, diffuse invasion, and a profoundly immunosuppressive tumor microenvironment. In this study, we applied an integrative multi-omics and radiogenomic framework to identify candidate biomarkers reflecting complementary dimensions of GBM heterogeneity. Public bulk transcriptomic datasets were integrated to define GBM-related differentially expressed genes, which were intersected with curated microbiota-associated gene sets as a hypothesis-generating screening strategy. Machine-learning models with SHAP interpretation were use

A Colon-Targeted Oral Nanosystem Disrupts the Inflammatory Loop in Enteric Glia to Alleviate Ulcerative Colitis.

The gut's inflammation is governed by the enteric nervous system, where enteric glial cells (EGCs) serve as essential intermediaries between the nervous and immune systems. During inflammation, elevated levels of S100 calcium-binding protein B (S100B) from hyperactive EGCs initiate a proinflammatory cascade by inducing the excessive production of reactive oxygen species (ROS) and proinflammatory molecules, including S100B itself, thus establishing a detrimental feedback loop. Herein, we develop a S100B inhibitor pentamidine (PTM)-loaded olsalazine-based nanoneedle, Zn2(Olsa)/PTM, to break this

Significant correlation between serum biomarkers and outcome in neonatal hypoxic-ischaemic encephalopathy.

AIM: Associations between serum biomarkers S100 calcium-binding protein B (S100B) and neuron-specific enolase (NSE) in neonates with hypoxic-ischaemic encephalopathy (HIE) offer contradicting data in regard to neurocognitive outcome. The aim of our study was to provide another dataset to answer this question if S100B or NSE correlates to outcome in neonatal HIE. In addition, we investigate whether amplitude-integrated EEG (aEEG) or magnetic resonance imaging (MRI) also has predictive value. METHODS: In neonates with HIE born in Vorarlberg, Austria, (n = 34) from 2012to 2020, NSE and S100B ser

The Effects of Transcranial Direct Current Stimulation and Exercise on Salivary S100B Protein Indicated Blood-Brain Barrier Permeability: A Pilot Study.

OBJECTIVE: This study aimed to assess the effect of transcranial direct current stimulation (tDCS) and exercise on blood-brain barrier (BBB) permeability in humans as assessed through the quantification of the salivary protein biomarker S100B. It was hypothesized that active tDCS would induce a significant increase in salivary S100B concentration when compared with sham stimulation and no stimulation. It also was hypothesized that the increase in salivary S100B concentration would be greater after active tDCS and exercise than after tDCS or exercise alone. MATERIALS AND METHODS: A total of 13

S100B urine levels are gestational age, birthweight, delivery mode and gender-dependent in healthy newborns.

OBJECTIVES: The inclusion of neurobiomarkers (NB) into routine clinical practice remains an unresolved challenge. Among the actions recommended by leading international institutions, a key priority is the establishment of reference curves during the perinatal period. The aim of the study was to define urinary reference ranges during the perinatal period for S100B, a well-established biomarker of central nervous system (CNS) development and injury. METHODS: Between May 2015 and January 2020, we conducted a prospective observational study in 2,168 healthy preterm and term newborns. Standard cli

S100B Mitigates Cytoskeletal and Mitochondrial Alterations in a Glial Cell Model of Autosomal Recessive Spastic Ataxia of Charlevoix-Saguenay.

Autosomal Recessive Spastic Ataxia of Charlevoix-Saguenay (ARSACS) is an early-onset neurological disorder caused by mutations in the SACS gene, resulting in the loss of sacsin function. Sacsin is a multidomain protein that plays key roles in chaperone regulation, protein quality control, and neurofilament dynamics. Sacsin deficiency leads to disruption of intermediate filament and mitochondrial networks. S100B, a multifunctional brain-enriched protein, exhibits protective neuroprotective functions that include chaperone activity and interactions with filament proteins and mitochondria. In thi

Role of S100β in Unstable Angina Pectoris: Insights from Quantitative Flow Ratio.

BACKGROUND AND OBJECTIVE: Disturbed autonomic nervous system (ANS) may promote inflammatory, immune, and oxidative stress responses, which may increase the risk of acute coronary events. S100β has been proposed as a biomarker of neuronal injury that would provide an insightful understanding of the crosstalk between the ANS, immune-inflammatory cells, and plaques that drive atherosclerosis. This study investigates the correlation between S100β, and functional coronary stenosis as determined by quantitative flow ratio (QFR). METHODS: Patients with unstable angina pectoris (UAP) scheduled for co
