Interaction of scorpion toxins with the sodium channel.

Scorpion toxins are miniproteins that have been characterized with regard to their molecular properties including their pharmacological action. On these bases, toxins specific to mammals or to insects have been described and within the first category alpha- and beta-toxins identified. These toxins were chemically modified and used as molecular probes of the sodium channel. Thus, the interaction of the toxins with the pharmacological target could be followed and a better definition of this target could be obtained.

The three-dimensional structure of scorpion neurotoxins.

The crystal and molecular structure of a toxin from the scorpion Centruroides sculpturatus has been solved by standard x-ray crystallographic methods at 3 A resolution. Subsequently the 3 A model has been refined and the resolution has been extended to 1.8 A using the gradient-curvature method. The final reliability index of 0.17 The structure has two and a half turns of alpha-helix, a three-strand stretch of antiparallel beta-sheet and several beta-turns. Three of the four disulfide bridges are found in close interaction with the alpha-helix and beta-sheet structures in what constitutes a ver

Pathophysiological Effects of the North Africa's Dangerous Scorpions and Antivenom Neutralization Efficacity: Unveiling the Spleen's Response.

The scorpion envenomations represent a serious public health issue, especially in North Africa. Their venom is known as a poison causing fat … ization effects of a rabbit-made antivenom on the spleen level against the North Africa's dangerous scorpions: Androctonus australis hector (Aah), Androctonus mauretanicus (Am), and Buthus occitanus (Bo) by the direct or cross-reactivity reaction. Firstly, the acute toxicity was assessed by determining the Median Lethal Dose (LD50) and Sublethal Dose (sLD), …

Toxic peptides and genes encoding toxin gamma of the Brazilian scorpions Tityus bahiensis and Tityus stigmurus.

Seven toxic peptides from the venom of Tityus bahiensis and Tityus stigmurus was isolated and sequenced, five of them to completion. The most abundant peptide from each of these two species of scorpion was 95% identical with that of toxin gamma from the venom of Tityus serrulatus. They were consequently named gamma-b and gamma-st respectively. The genes encoding these new gamma-like peptides were cloned and sequenced by utilizing oligonucleotides synthesized according to known cDNA sequences of toxin gamma, and amplified by PCR on templates of DNA purified from both T. bahiensis and T. stigmur

Purification and physiological characterization of neurotoxins from venoms of the scorpions centruroides sculpturatus and leiurus quinquestriatus.

Several different toxins having specific effects on the kinetics of sodium channels have been isolated from the venoms of two scorpions. A combination of two steps of ion-exchange chromatography has been used to purify these toxins, whose sizes and purities have been assayed by gel filtration, urea/sodium dodecyl sulfate/polyacrylamide gel electrophoresis, and isoelectric focusing. The actions of the toxins and their relative potencies have been determined by studying the modifications they produce in action potential shape, using the sucrose-gap method, and in ionic current kinetics, measured…

Beta Toxins Isolated from the Scorpion Centruroides hirsutipalpus (Scorpiones; Buthidae) Affect the Function of Sodium Channels of Mammals.

Scorpion venom toxins are important peptides being studied for their clinical significance. These peptides act by binding to ion channels in the membrane of nerve cells, causing the symptoms associated with scorpion stings (scorpionism). They principally affect the function of voltage-gated sodium channels (Nav) and are valuable for studying ion channels. Scorpions from the Buthidae family contain toxins that affect sodium channels and have a high affinity for mammalian channels. In this study, two sodium toxins isolated from the venom of the scorpion Centruroides hirsutipalpus, a member of th…

Scorpion Stings in Minas Gerais (Brazil): A Monocentric Retrospective Study Evaluating All Envenoming Cases of Local Scorpionism.

INTRODUCTION: We aim to provide better insights into the demographic, epidemiological, and seasonal characteristics of scorpion envenomation reported in the Rio Doce Valley, Brazil. METHODS: We conducted this monocentric retrospective descriptive study with data on Investigation Forms of Accidents by Venomous Animals, only envenoming cases of scorpion stings, between January 1, 2017 and December 31, 2020, belonging to the compulsory notification system of the Municipal Hospital of Governador Valadares. RESULTS: There were 3032 accidents, and the male-to-female ratio was 1.17:1 (male, 54%; fe
