Magnetosome-incorporated silk fibroin-based magnetic hydrogel for efficient tumor magnetic hyperthermia.

We developed a magnetosome-incorporated silk fibroin-based magnetic hydrogel (MSMH) for efficient tumor magnetic hyperthermia. The MSMH was fabricated by integrating biologically synthesized magnetosomes with glycidyl methacrylate-modified silk fibroin (SFGMA), enabling injectable deployment and rapid photo-crosslinking. The as-fabricated MSMH exhibits excellent magnetothermal conversion performance, achieving prominent temperature rises under alternating magnetic fields (AMF). MSMH can also serve as a T₂-weighted MRI contrast agent (r2 = 173.57 mM-1 s-1 at 0.5 T), enabling imaging-guided ther

Silk fibroin microneedles loaded with epigallocatechin gallate mitigate atrazine-induced testicular toxicity.

Atrazine (ATZ), a commonly used herbicide, disrupts male endocrine systems, impacting reproductive health. Epigallocatechin gallate (EGCG) possesses potent antioxidant properties and shows promise in alleviating testicular dysfunction induced by endocrine disruptors. However, its clinical utility is hindered by poor physicochemical stability and low oral bioavailability. Herein, we first developed a silk fibroin microneedles (MNs) patch loaded with EGCG, enabling EGCG to directly target the testes through transdermal administration. Our findings revealed that EGCG-loaded silk fibroin microneed

A recombinant chimeric spider pyriform-aciniform silk with highly tunable mechanical performance.

Spider silks are natural protein-based biomaterials which are renowned for their mechanical properties and hold great promise for applications ranging from high-performance textiles to regenerative medicine. While some spiders can produce several different types of silks, most spider silk types - including pyriform and aciniform silks - are relatively unstudied. Pyriform and aciniform silks have distinct mechanical behavior and physicochemical properties, with materials produced using combinations of these silks currently unexplored. Here, we introduce an engineered chimeric fusion protein con

Unveiling the Rational Development of Stimuli-Responsive Silk Fibroin-Based Ionogel Formulations.

We present an approach for the rational development of stimuli-responsive ionogels which can be formulated for precise control of multiple unique ionogel features and fill niche pharmaceutical applications. Ionogels are captivating materials, exhibiting self-healing characteristics, tunable mechanical and structural properties, high thermal stability, and electroconductivity. However, the majority of ionogels developed require complex chemistry, exhibit high viscosity, poor biocompatibility, and low biodegradability. In our work, we overcome these limitations. We employ a facile production pro

Luminal endothelialization of small caliber silk tubular graft for vascular constructs engineering.

The constantly increasing incidence of coronary artery disease worldwide makes necessary to set advanced therapies and tools such as tissue engineered vessel grafts (TEVGs) to surpass the autologous grafts [(i.e., mammary and internal thoracic arteries, saphenous vein (SV)] currently employed in coronary artery and vascular surgery. To this aim, in vitro cellularization of artificial tubular scaffolds still holds a good potential to overcome the unresolved problem of vessel conduits availability and the issues resulting from thrombosis, intima hyperplasia and matrix remodeling, occurring in au

Evaluation of Neurosecretome from Mesenchymal Stem Cells Encapsulated in Silk Fibroin Hydrogels.

Physical and cognitive disabilities are hallmarks of a variety of neurological diseases. Stem cell-based therapies are promising solutions to neuroprotect and repair the injured brain and overcome the limited capacity of the central nervous system to recover from damage. It is widely accepted that most benefits of different exogenously transplanted stem cells rely on the secretion of different factors and biomolecules that modulate inflammation, cell death and repair processes in the damaged host tissue. However, few cells survive in cerebral tissue after transplantation, diminishing the thera

Membrane-reinforced three-dimensional electrospun silk fibroin scaffolds for bone tissue engineering.

Electrospun silk fibroin (SF) scaffolds have drawn much attention because of their resemblance to natural tissue architecture such as extracellular matrix, and the biocompatibility of SF as a candidate material to replace collagen. However, electrospun scaffolds lack the physical integrity of bone tissue scaffolds, which require resistance to mechanical loadings. In this work, we propose membrane-reinforced electrospun SF scaffolds by a serial process of electrospinning and freeze-drying of SF solutions in two different solvents: formic acid and water, respectively. After wet electrospinning f

Silk sericin ameliorates wound healing and its clinical efficacy in burn wounds.

The aim of this study was to evaluate the effect of silk sericin, a protein from silkworm cocoon, on scratch wound healing in vitro. For applicable result in clinical use, we also study the efficacy of sericin added to a standard antimicrobial cream, silver zinc sulfadiazine, for open wound care in the treatment of second-degree burn wounds. In vitro scratch assays show that sericin at concentration 100 μg/mL can promote the migration of fibroblast L929 cells similar to epidermal growth factor (positive control) at 100 μg/mL. After 1 day of treatment, the length of scratch in wounds treated wi

Biomimetic nanofibrous scaffolds: preparation and characterization of chitin/silk fibroin blend nanofibers.

Electrospinning of chitin/silk fibroin (SF) blend solutions in 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) was investigated to fabricate a biomimetic nanostructured scaffolds for tissue engineering. The morphology of the electrospun chitin/SF blend nanofibers was investigated with a field emission scanning electron microscope (FE-SEM). The average diameters of chitin/SF blend fibers decreased from 920 to 340nm, with the increase of chitin content in blend compositions. The miscibility of chitin/SF blend fibers was examined by solution viscosity measurement. The chitin and SF were immiscible in th

Scalable Mie-Silk textile for dual-mode personal cooling indoors and outdoors.

Radiative cooling textiles are typically designed for specific indoor or outdoor conditions, limiting their environmental adaptability. Here, we report a continuously spun molecular interface-engineered silk (Mie-Silk) that integrates radiative and conductive cooling for effective body temperature regulation in diverse settings. The Mie-Silk has a natural silk yarn core with high mid-infrared (MIR) emissivity and a surface layer of ZrO2 nanoparticles (NPs) enabling strong solar scattering based on the Mie scattering principle. Natural silk fibroin acts as a robust binder, anchoring the NPs on

Translating silk protein hierarchical structure into graphitic order by FIB-induced pyrolysis.

Silk fibers are protein-based materials with a highly ordered hierarchical architecture extending from β-sheet-rich molecular domains and aligned polypeptide chains to nanofibrillar bundles and microscopic fibers, collectively giving rise to their exceptional mechanical performance. This intrinsic multiscale organization also offers a powerful, yet largely unexplored, platform for templating ordered carbon materials with programmable nanoscale anisotropy. During our nanoscale structural analysis, Bombyx mori (B. mori) silk fibers were unintentionally carbonized while being sectioned into thin

Manufacturing Silk Fibroin Hollow Nanoyarns as Fundamental Units for Advanced Medical Textiles.

Nature-inspired designs aim to replicate the hierarchical structure observed in native tissues. Fibers serve as fundamental modular units, enabling the fabrication of complex architectures for the engineering of medical textiles and tissue equivalents. However, synthetic yarns lack inherent biological cues to support tissue integration, leading to a growing interest in yarns derived from natural materials. Here, we describe for the first time the fabrication of hollow nanoyarns from pure silk fibroin (SF) using an advanced funnel electrospinning process. This yielded long SF nanoyarns, spannin

Silk Sericin in Dermatological Diseases: From Preclinical Studies to Future Clinical Applications.

Biomaterials in medicine are becoming more widespread as a single or complementary treatment option. One such biomaterial is silk, comprised of two primary proteins: fibroin and sericin. Recently, sericin's anti-inflammatory, antioxidant, moisturizing, elastase- and tyrosinase-inhibiting properties have been widely investigated. Sericin biomaterials are already used in wound healing and bone tissue engineering. Additionally, there are promising results for its usefulness in many other applications. This review focuses on sericin use in dermatological diseases, above all in atopic dermatitis an

Diagnosing plague in 17th century camelids from Romania along historical Silk Routes.

Six camels exhumed from a 17th-century Silk Route site in Romania, along with negative controls, were blindly investigated via dental pulp paleometagenomics and paleoproteomics for traces of Yersinia pseudotuberculosis complex including the plague agent Yersinia pestis. Specific reads were detected in sample R04 (one read) and R05 (two reads) which also yielded a 16S rRNA guanine transferase specific for Y. pestis and one other Y. pseudotuberculosis complex peptide. Taken together, these validated data diagnosed plague in these ancient camels which likely participated in plague dissemination a

Silk scaffolding drives self-assembly of functional and mature human brain organoids.

Human pluripotent stem cells (hPSCs) are intrinsically able to self-organize into cerebral organoids that mimic features of developing human brain tissue. These three-dimensional structures provide a unique opportunity to generate cytoarchitecture and cell-cell interactions reminiscent of human brain complexity in a dish. However, current in vitro brain organoid methodologies often result in intra-organoid variability, limiting their use in recapitulating later developmental stages as well as in disease modeling and drug discovery. In addition, cell stress and hypoxia resulting from long-term
