Bacillus subtilis spores displaying Toxoplasma gondii GRA12 induce immunity against acute toxoplasmosis.

BACKGROUND: Toxoplasma gondii (T. gondii) is a widely prevalent intracellular parasite that infects almost all warm-blooded animals and caus … nstructed by fusing the T. gondii GRA12 protein to the B. subtilis coat protein B (CotB). rBS-GRA12 spores were identified by PCR, western blotting, immunofluorescence assays, amylase activity, and ultrastructural analysis. Immunological experiments were then conducted to assess the immunoprotective effects of rBS-GRA12. Groups of mice immunized with rBS …

UV-Induced Spectral and Morphological Changes in Bacterial Spores for Inactivation Assessment.

The ability to detect and inactivate spore-forming bacteria is of significance within, for example, industrial, healthcare, and defense sectors. Not only are stringent protocols necessary for the inactivation of spores but robust procedures are also required to detect viable spores after an inactivation assay to evaluate the procedure's success. UV radiation is a standard procedure to inactivate spores. However, there is limited understanding regarding its impact on spores' spectral and morphological characteristics. A further insight into these UV-induced changes can significantly improve the…

Effects of Different Bacteriostats on the Dynamic Germination of Clostridium perfringens Spores.

Bacteriostats, as chemical substances that inhibit bacterial growth, are widely used in the sterilization process; however, their effects on spindle spores are unclear. In this study, the effects of bacteriostats, including nine commonly used food additives and four detergents, on the growth of Clostridium perfringens spores were investigated. The results showed that 0.07‱ ethylenediaminetetraacetate had a good inhibitory effect on C. perfringens spore growth, and the spore turbidity decreased by 4.8% after incubation for 60 min. Furthermore, 0.3‱ tea polyphenols, 0.8‱ D-isoascorbic acid, and …

Establishing Sterility Assurance for Bacillus canaveralius 29669 Spores Under High Heat Exposure.

The ever-increasing complexity in critical spacecraft hardware and materials has led to the development of new microbial reduction procedure … ralius 29669 to high-temperature, short-duration infrared temperatures. While B. canaveralius 29669 spores can survive microbial heat reduction processes above 200°C, we found evidence suggesting that the 500°C for 0.5 s temperature sterilization specification for Planetary Protection should be updated. This research presents spore survival data and a co …

Aerosol and Surface Deposition Characteristics of Two Surrogates for Bacillus anthracis Spores.

Spores of an acrystalliferous derivative of Bacillus thuringiensis subsp. kurstaki, termed Btcry-, are morphologically, aerodynamically, and structurally indistinguishable from Bacillus anthracis spores. Btcry- spores were dispersed in a large, open-ended barn together with spores of Bacillus atrophaeus subsp. globigii, a historically used surrogate for Bacillus anthracis Spore suspensions (2 × 1012 CFU each of B. atrophaeus subsp. globigii and Btcry-) were aerosolized in each of five spray events using a backpack misting device incorporating an air blower; a wind of 4.9 to 7.6 m s-1 was also …

Dormant Bacillus spores protect their DNA in crystalline nucleoids against environmental stress.

Bacterial spores of the genera Bacillus and Clostridium are extremely resistant against desiccation, heat and radiation and involved in the spread and pathogenicity of health relevant species such as Bacillus anthracis (anthrax) or Clostridium botulinum. While the resistance of spores is very well documented, underlying mechanisms are not fully understood. In this study we show, by cryo-electron microscopy of vitreous sections and particular resin thin section electron microscopy, that dormant Bacillus spores possess highly ordered crystalline core structures, which contain the DNA, but only i…

Role of YpeB in cortex hydrolysis during germination of Bacillus anthracis spores.

The infectious agent of the disease anthrax is the spore of Bacillus anthracis. Bacterial spores are extremely resistant to environmental stresses, which greatly hinders spore decontamination efforts. The spore cortex, a thick layer of modified peptidoglycan, contributes to spore dormancy and resistance by maintaining the low water content of the spore core. The cortex is degraded by germination-specific lytic enzymes (GSLEs) during spore germination, rendering the cells vulnerable to common disinfection techniques. This study investigates the relationship between SleB, a GSLE in B. anthracis,…

Portable, quantitative detection of Bacillus bacterial spores using surface-enhanced Raman scattering.

Portable rapid detection of pathogenic bacteria such as Bacillus is highly desirable for safety in food manufacture and under the current he … rate that SERS is able to detect and quantify rapidly the dipicolinate (DPA) biomarker for Bacillus spores at 5 ppb (29.9 nM) levels which are significantly lower than those previously reported for SERS and well below the infective dose of 10(4)B. anthracis cells for inhalation anthrax. Finally we show the potential of multivariate data analysis to impro …

Dueling in the lung: how Cryptococcus spores race the host for survival.

Many human fungal pathogens infect people when they are inhaled as spores. Despite the serious impact of fungal spores on human health, little is known about their basic properties or how they interact with the host. This is particularly true for Cryptococcus neoformans, a human fungal pathogen that causes more than 600,000 deaths annually. Spores of C. neoformans have not been well characterized previously because of technical challenges in isolating them; however, recent advances in spore isolation have lead to the first direct analyses of spores. Novel insights into the spore-host interacti…

Immunization against anthrax using Bacillus subtilis spores expressing the anthrax protective antigen.

Protective immunity to anthrax can be achieved by antibodies raised against the secreted protective antigen (PA) and this forms the basis of the current acellular vaccines for human use. Bacillus subtilis spores have previously been used for delivery of heterologous antigens by the oral and nasal routes and their intrinsic heat-stability make them attractive vaccine vehicles. In this study we have expressed PA, or segments of PA, in B. subtilis using two strategies. First, display on the spore coat, and second, in the germinated spore (or vegetative cell). Using parenteral delivery we show tha…

[Efficient killing of anthrax spores using aqueous and alcoholic peracetic acid solutions].

We analysed the sporicidal effect of different concentrations of aqueous and alcoholic peracetic acid (PAA) solutions on anthrax spores in suspension and germ carrier tests. In activation of anthrax spores in suspension assays was achieved in less than 2 min using 1% PAA solution and in less than 3 min using 0.5% PAA solution, respectively. In contrast, in germ carrier as says, a test under practical conditions, spores on 38% of the germ carriers survived treatment with 1% PAA solution for 15 min. The use of PAA in 80% ethyl alcohol outclassed the sporicidal effect of aqueous PAA solutions in …

In vitro selection of DNA aptamers to anthrax spores with electrochemiluminescence detection.

Systematic evolution of ligands by exponential enrichment (SELEX) was used to select and PCR amplify DNA sequences (aptamers) capable of binding to and detecting nonpathogenic Sterne strain Bacillus anthracis spores. A simplified affinity separation approach was employed, in which autoclaved anthrax spores were used as the separation matrix. An aptamer-magnetic bead-electrochemiluminescence (AM-ECL) sandwich assay scheme was devised for detecting anthrax spores. Using a low SELEX DNA to spore ratio (154 ng DNA/10(6) spores), at least three distinct populations of single-stranded DNA aptamers, …

The role of airborne fungal spores from garbage dumps in respiratory diseases.

In recent years, there has been a growing global, regional and local concern about the potential health hazards associated with air pollution. Great attention has so far been paid to pollution associated with oxides of sulphur, CO, nitrogen oxides, O(3), trace organics, selected trace metals like lead, and miscelleneous suspended particulate matter. But, apart from these much talked about pollutants, urban air pollution associated with fungal spores from garbage pose a major environmental human health problem in most urban centres in Africa. Garbage probably accounts for most of the fungal spo…

Buoyant density heterogeneity in spores of Bacillus subtilis: biochemical and physiological basis.

The biochemical and physiological basis of density heterogeneity in Renografin of Bacillus subtilis W23 spores was determined by analysis of metals, macromolecules, and dipicolinic acid in the two density classes of the population. Germination rate and heat resistance were measured in both density classes. Atomic absorption spectrophotometry revealed that heavy spores (density = 1.335 g/ml) have 30% more calcium than light spores (density = 1.290 g/ml). Other metals found in greater amounts in heavy spores were manganese and potassium. However, light spores had more sodium than heavy spores. T…

-aminobutyric acid as a required germinant for mutant spores of Bacillus megaterium.

Germinated spores of Bacillus megaterium QM B1551 were irradiated with ultraviolet light, and spore-forming survivors were screened for germination requirements. Spore strains which failed to germinate in a variety of defined solutions germinative for spores of the parent strain were obtained. Mutant spores germinated readily in solutions containing yeast extract or one of numerous complex preparations. gamma-Aminobutyric acid, obtained from yeast extract by column chromatography, was shown to be required for germination by the mutant spores. gamma-Aminobutyric acid and l-alanine at final conc…

Urea-mercaptoethanol-soluble protein from spores of Bacillus thuringiensis and other species.

Treatment with urea-mercaptoethanol of purified spores of Bacillus thuringiensis, other Bacillus species, and Clostridium roseum solubilizes a protein fraction between 5 and 12% of the dry weight of the spores. This fraction behaves identically to the crystal protein of B. thuringiensis on acrylamide-gel electrophoresis. The protein from all of the Bacillus species shows partial homology with crystal protein, using the Ouchterlony immunodiffusion technique. A further fraction, similar in amount, can be removed from spores of B. thuringiensis by the addition of sodium lauryl sulfate to the urea…
