Poor sleep and high rheumatoid arthritis risk: Evidence from large UK Biobank cohort.

OBJECTIVE: To evaluate the relationship between sleep behaviors and rheumatoid arthritis (RA) risk. METHODS: First, based on large-scale data from the UK Biobank, we analyzed the associations between multiple sleep factors and RA risk and calculated a poor sleep score (PSS) to assess their combined effect. Then we constructed genetic risk scores (GRS) based on a large genome-wide association study and assessed the interaction or combined effect of sleep and genetic factors on RA risk. Finally, we conducted a case-control study to explore the effect of RA on sleep. RESULTS: Sleep duration, ge…

Association between leukocyte telomere length and incident glaucoma: A prospective UK biobank study.

BACKGROUND: Leukocyte telomere length (LTL) has been associated with various diseases, including age-related eye diseases such as cataract and age-related macular degeneration. However, the role of LTL in the longitudinal development of glaucoma is still unknown. Here we prospectively evaluate the association of LTL with glaucoma incidence and related traits, in the UK Biobank cohort. METHODS: The study cohort included 419,603 participants with complete baseline data for glaucoma analyses. Multivariable Cox proportional hazards models were used to evaluate the association between LTL and the …

Defining a High-Quality Myalgic Encephalomyelitis/Chronic Fatigue Syndrome cohort in UK Biobank.

BACKGROUND: Progress in Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (ME/CFS) research is being slowed by the relatively small-scale studies being performed whose results are often not replicated. Progress could be accelerated by analyses of large population-scale projects, such as UK Biobank (UKB), which provide extensive phenotype and genotype data linked to both ME/CFS cases and controls. METHODS: Here, we analysed the overlap and discordance among four UKB-defined ME/CFS cohorts, and additional questionnaire data when available. RESULTS: A total of 5,354 UKB individuals were linked…

Diet quality patterns and chronic kidney disease incidence: a UK Biobank cohort study.

BACKGROUND: Only a few studies have investigated the role of diet on the risk of chronic kidney disease (CKD) in European populations and have mainly focused on the Mediterranean diet. This is the first study to evaluate the association between various diet quality indices and CKD incidence in British adults. OBJECTIVE: To study the relationship between a set of 6 different diet quality indices and CKD incidence among British adults. METHODS: A prospective cohort with 106,870 participants from the UK Biobank, followed from 2009 to 2012 through 2021. Food consumption was obtained from ≥2 24-h…

Defining left ventricular remodeling using lean body mass allometry: a UK Biobank study.

PURPOSE: The geometric patterns of ventricular remodeling are determined using indexed left ventricular mass (LVM), end-diastolic volume (LVEDV) and concentricity, most often measured using the mass-to-volume ratio (MVR). The aims of this study were to validate lean body mass (LBM)-based allometric coefficients for scaling and to determine an index of concentricity that is independent of both volume and LBM. METHODS: Participants from the UK Biobank who underwent both CMR and dual-energy X-ray absorptiometry (DXA) during 2014-2015 were considered (n = 5064). We excluded participants aged ≥ 70…

Genome-wide association study on pharmacological outcomes of musculoskeletal pain in UK Biobank.

The pharmacological management of musculoskeletal pain starts with NSAIDs, followed by weak or strong opioids until the pain is under control. However, the treatment outcome is usually unsatisfying due to inter-individual differences. To investigate the genetic component of treatment outcome differences, we performed a genome-wide association study (GWAS) in ~23,000 participants with musculoskeletal pain from the UK Biobank. NSAID vs. opioid users were compared as a reflection of the treatment outcome of NSAIDs. We identified one genome-wide significant hit in chromosome 4 (rs549224715, P = 3.…

Long-term cardiovascular outcomes of gestational diabetes mellitus: a prospective UK Biobank study.

BACKGROUND: Previous studies showed that gestational diabetes mellitus (GDM) can be a risk factor for subsequent atherosclerotic cardiovascular disease. However, there is a paucity of information regarding diverse cardiovascular outcomes in elderly women after GDM. In the current study, we examined whether women with a history of GDM have an increased risk for long-term overall cardiovascular outcomes. METHODS: Among the UK participants, we included 219,330 women aged 40 to 69 years who reported at least one live birth. The new incidence of diverse cardiovascular outcomes was compared accordi

UKB.COVID19: an R package for UK Biobank COVID-19 data processing and analysis.

COVID-19 caused by SARS-CoV-2 has resulted in a global pandemic with a rapidly developing global health and economic crisis. Variations in the disease have been observed and have been associated with the genomic sequence of either the human host or the pathogen. Worldwide scientists scrambled initially to recruit patient cohorts to try and identify risk factors. A resource that presented itself early on was the UK Biobank (UKBB), which is investigating the respective contributions of genetic predisposition and environmental exposure to the development of disease. To enable COVID-19 studies, UK…

Analysis of exome-sequenced UK Biobank subjects implicates genes affecting risk of hyperlipidaemia.

Rare genetic variants in LDLR, APOB and PCSK9 are known causes of familial hypercholesterolaemia and it is expected that rare variants in other genes will also have effects on hyperlipidaemia risk although such genes remain to be identified. The UK Biobank consists of a sample of 500,000 volunteers and exome sequence data is available for 50,000 of them. 11,490 of these were classified as hyperlipidaemia cases on the basis of having a relevant diagnosis recorded and/or taking lipid-lowering medication while the remaining 38,463 were treated as controls. Variants in each gene were assigned weig…

UACR as a clinical predictor of incident osteoarthritis: prospective evidence from the UK Biobank.

BACKGROUND: Osteoarthritis (OA) is a common joint disorder causing disability, marked by pain and stiffness, with current therapies only relieving symptoms. This highlights the need for early biomarkers. The urinary albumin-to-creatinine ratio (UACR) is an indicator of renal dysfunction and systemic inflammation, while OA is also closely associated with inflammatory processes. However, the direct relationship between UACR and OA risk remains unclear. We investigated the prospective association between UACR and incident OA. METHODS: We analyzed data from 445,860 UK Biobank participants without…
