Immunofluorescent staining of Treponema pallidum and Treponema pertenue in tissues fixed by formalin and embedded in paraffin wax.

The main problems in identifying Treponema pallidum in tissues are optical definition contrast, and specificity. In general, fluorochrome staining provides optical definition and contrast superior to that obtained by ordinary tinctorial staining, and in theory improved resolution. Specificity is lacking however, as with other stains. In contrast, immunofluorescence should combine the optical advantages of fluorochrome staining with the immunological advantages of specificity. Since the validity of such staining depends in part upon the integrity of the antigenic components of the micro-organis

Metabolic engineering of cereal lipids: from omega-3 fatty acids to wax esters and pheromones.

Cereals are emerging as attractive platforms for the sustainable production of high-value lipids through metabolic engineering. Although plant lipids play essential biological roles and have considerable economic value, their conventional production from natural sources is often limited by sustainability, scalability and cost. Recent advances in synthetic biology enable the reprogramming of seed lipid metabolism for the tailored synthesis of valuable lipid compounds. In this review, we first summarize the core pathways of fatty acid biosynthesis and triacylglycerol assembly in seeds, together

Polyethylene Glycol (PEG)-Based Wet-Adhesive Absorbable Bone Wax for Osseous Hemostasis and Repair.

The non-absorbability and biological inertia of traditional bone wax often result in foreign body retention, inhibit bone healing, and increase the risk of infection. Herein, a novel absorbable bone wax was developed by mixing polyethylene glycol (PEG), tannic acid (TA), sodium carboxymethyl cellulose (CMC), and low-molecular-weight poly-L-lactic acid (PLLA) via one-pot method. Among them, PEG functions as the matrix, endowing it with plasticity and hydrophilicity, thereby facilitating its facile, close fit to the surface of bone defects. TA, with its profusion of phenolic hydroxyl groups, bes

Pysanky to Microfluidics: An Innovative Wax-Based Approach to Low Cost, Rapid Prototyping of Microfluidic Devices.

A wax-based contact printing method to create microfluidic devices is demonstrated. This printing technology demonstrates a new pathway to rapid, cost-effective device prototyping, eliminating the use of expensive micromachining equipment and chemicals. Derived from the traditional Ukrainian Easter egg painting technique called "pysanky" a series of microfluidic devices were created. Pysanky is the use of a heated wax stylus, known as a "kistka", to create micro-sized, intricate designs on the surface of an egg. The proposed technique involves the modification of an x-y-z actuation translation

An innovative diagnosis in gastrointestinal neuroendocrine neoplasms using Wax-Physisorption-Kinetics-based FTIR Imaging.

Neuroendocrine neoplasm (NEN) is a common gastrointestinal (GI) tract tumor divided into the neuroendocrine tumor (NET) and neuroendocrine carcinoma (NEC) according to mitosis and Ki-67 index. However, the objective discordance between interobserver may cause unsuitable diagnosis and misleading treatment. Nowadays, aberrant glycosylation of glycoconjugates inducing further populations of elongated complex oligosaccharide covalent attached to glycoconjugates anchored in the cell membrane by neo-synthesis of cancer-associated alteration of carbohydrate determinants were observed during cancer de

Dissecting the Roles of Cuticular Wax in Plant Resistance to Shoot Dehydration and Low-Temperature Stress in Arabidopsis.

Cuticular waxes are a mixture of hydrophobic very-long-chain fatty acids and their derivatives accumulated in the plant cuticle. Most studies define the role of cuticular wax largely based on reducing nonstomatal water loss. The present study investigated the role of cuticular wax in reducing both low-temperature and dehydration stress in plants using Arabidopsis thaliana mutants and transgenic genotypes altered in the formation of cuticular wax. cer3-6, a known Arabidopsis wax-deficient mutant (with distinct reduction in aldehydes, n-alkanes, secondary n-alcohols, and ketones compared to wild…

Establishing an invertebrate Galleria mellonella greater wax moth larval model of Neisseria gonorrhoeae infection.

Neisseria gonorrhoeae (gonococcus) causes the human sexually transmitted disease gonorrhea. Studying gonococcal pathogenesis and developing new vaccines and therapies to combat the increasing prevalence of multi-antibiotic resistant bacteria has made use of many ex vivo models based on human cells and tissues, and in vivo vertebrate models, for example, rodent, pig and human. The focus of the current study was to examine the utility of the invertebrate greater wax moth Galleria mellonella as an in vivo model of gonococcal infection. We observed that a threshold of ~106 - 107 gonococci/larva wa

The history of an unknown dermatological wax collection from Cluj-Napoca University "Iuliu Hatieganu", Romania.

Among the dermatological wax collections across Europe, one of the latest created is the collection from Cluj-Napoca University, Romania. The initiator was Professor Coriolan Tataru and the moulage artist employed was Dr. Richard Hoffmann. Between the years 1923 and 1928, around 200 wax moulages were made, all realised after patients hospitalised in the clinic. The majority of cases represent the dermatological infectious pathology of that time: syphilis, cutaneous tuberculosis and mycetomas. Other interesting moulages represent genodermatoses, pelagra, different cutaneous cancers, and atypica

Design of porous Eudragit® L beads for floating drug delivery by wax removal technique.

The aim of this study was to design porous matrix beads for floating drug delivery using enteric polymer, Eudragit® L and various amounts of waxes (0, 0.1, 0.5, 1, 2 and 3% w/w). In this study, wax containing cetyl alcohol and white petrolatum was utilized to produce pores using a wax removal technique. To prepare the beads, Eudragit® L, metronidazole and wax were dissolved in acetone and then extruded into dichloromethane. The effect of the amount of wax on the floating and drug release behavior of the Eudragit® L beads was determined. After the extruded product was immersed in dichloromethan…

Wax and cutin mutants of Arabidopsis: Quantitative characterization of the cuticular transport barrier in relation to chemical composition.

Using (14)C-labeled epoxiconazole as a tracer, cuticular permeability of Arabidopsis thaliana leaves was quantitatively measured in order to compare different wax and cutin mutants (wax2, cut1, cer5, att1, bdg, shn3 and shn1) to the corresponding wild types (Col-0 and Ws). Mutants were characterized by decreases or increases in wax and/or cutin amounts. Permeances [ms(-1)] of Arabidopsis cuticles either increased in the mutants compared to wild type or were not affected. Thus, genetic changes in wax and cutin biosynthesis in some of the investigated Arabidopsis mutants obviously impaired the c

Screening of pesticide residues in honeybee wax comb by LC-ESI-MS/MS. A pilot study.

A developed multi-residue method using microflow-LC-ESI-QqQ-MS provided a wide-scope analysis for medium-polar and polar pesticide residues (120 compounds including breakdown products). Honeybee wax comb samples were extracted using a generic QuEChERS based procedure. Acceptable recoveries at concentration levels of 5 and 50 μg kg(-1) were within the 70-120% range with an associated precision RSD <20%. The LOQ values were mostly 5 μg kg(-1) for almost all pesticides. Aprox. 31 of 120 LC-amenable pesticides tested (25.8%) were detected in a pilot study of 60 samples. Pesticide residues detected

Paraffin wax emulsion for increased rainfastness of insecticidal bait to control Rhagoletis pomonella (Diptera: Tephritidae).

In regions with a humid summer climate, the use of water-soluble bait to control apple maggot is often limited by rainfall. We studied increasing the rainfastness of GF-120 fruit fly bait by adding paraffin wax emulsion. First, we verified that adding 10% wax to a mixture containing 16.7% GF-120 did not reduce the mortality of female apple maggot compared with GF-120 without wax. In addition, we determined that fly mortality caused by GF-120 plus wax subjected to simulated rain was similar to that caused by GF-120 without wax not subjected to rain. Other assays showed that higher fly mortality

Experience of bone bridge resection and bone wax packing for partial growth arrest of distal tibia.

Two patients with a partial growth arrest of the distal tibia were treated by bone bridge resection and bone wax packing technique. Their average age at the time of surgery was 7 years, both had been injured in an accident, and each had been treated with percutaneous pinning and a plaster cast. Unfortunately, partial growth arrest of the distal tibia occurred, and the ankle varus deformity gradually progressed. About 1 year after initial treatment, bone bridge resection and bone wax packing were performed. The average follow-up period was 80 months. There was no reformation of the bone bridge

Free amino-acid content of wax-stimulated human whole saliva as related to periodontal disease.

This content was analysed in patients with chronic periodontitis and in control subjects. In periodontal disease, it was characterized by higher mean concentrations of glycine, proline, tyrosine and delta-aminovaleric acid than in controls (p less than 0.001). However, the range of values varied considerably in the two groups. There were differences between periodontitis and control samples in the proportions of proline to serine (p less than 0.01) and proline to glutamic acid and glutamine (p less than 0.05). Bacterial contamination and decomposition of salivary proteins is responsible for th
