Evaluation of a monoclonal-antibody based antigen assay for diagnosis of Wuchereria bancrofti infection in Egypt.

Conventional methods for diagnosis of Wuchereria bancrofti infection are insensitive and often impractical because of the need for night blood collections. A sensitive and specific antigen detection assay has been developed for W. bancrofti, which is based on a monoclonal antibody (AD12) that binds to a repeated epitope on a 200 kDa adult worm excretion product present in sera from infected humans. The only formal evaluation of this assay to date was performed with sera from India. In the present study, we have evaluated the performance of the AD12 antigen assay in two laboratories with sera c…

Natural infections of Wuchereria bancrofti in Aedes (Stegomyia) polynesiensis and Aedes (Finlaya) samoanus in Samoa.

Seven years after the 2nd mass treatment of the population with diethylcarbamazine, transmission of subperiodic Wuchereria bancrofti was studied in four villages in Samoa during one year by means of biting catches of Aedes polynesiensis and A. samoanus. 2 villages were coastal, one inland bush and the other an inland coconut plantation community. Overall infection and infective rates from 6702 Ae. polynesiensis were 0.84 and 0.27% respectively, and the infection rate from 2858 Ae. samoanus, collected in 10-minute catches from 24 sites, was 0.65%. No infective Ae. samoanus was found in these sa…

Wuchereria bancrofti in Tanzania: immune reactions to the microfilarial surface, and the effect of diethylcarbamazine upon these reactions.

Immune reactions to the surface of microfilariae (mff) of Wuchereria bancrofti were investigated among persons living in an endemic filariasis area of Tanzania. The sera from a proportion of mff-negative persons contained antibodies to the microfilarial sheath, and a clear correlation was seen between presence of anti-sheath antibodies and the ability of the sera to mediate leucocyte attack on the mff in vitro. The anti-sheath antibodies were mainly of the IgG class and to a lesser degree of the IgM class. Only in some sera did the binding of antibody to the microfilarial sheath lead to deposi…

Molecular xenomonitoring highlights post-MDA surveillance priorities for sustained Wuchereria bancrofti elimination in Burkina Faso.

The study by Nikièma et al. demonstrates the absence of Wuchereria bancrofti infection in Anopheles mosquitoes a decade after cessation of mass drug administration (MDA) in Burkina Faso. This rare longitudinal evidence underscores the utility of molecular xenomonitoring (MX) as a sensitive early-warning tool that complements traditional transmission assessment surveys (TAS). MX enables detection of recrudescence earlier than human-based diagnostics, particularly in low-prevalence settings, and offers opportunities for integration with malaria vector surveillance. Ethical and operational challe…

Genome-Wide Analysis Reveals Key Genes and MicroRNAs Related to Pathogenic Mechanism in Wuchereria bancrofti.

Wuchereria bancrofti is a parasite transmitted by mosquitoes and can cause a neglected tropical disease called Lymphatic filariasis. However, the genome of W. bancrofti was not well studied, making novel drug development difficult. This study aims to identify microRNA, annotate protein function, and explore the pathogenic mechanism of W. bancrofti by genome-wide analysis. Novel miRNAs were identified by analysis of expressed sequence tags (ESTs) from this parasite. Protein homology was obtained by a bidirectional best-hit strategy using BLAST. By an EST-based method, we identified 20 novel miR…

Use of high-dose, twice-yearly albendazole and ivermectin to suppress Wuchereria bancrofti microfilarial levels.

BACKGROUND: Annual mass treatment with albendazole and ivermectin is the mainstay of current strategies to interrupt transmission of Wuchereria bancrofti in Africa. More-effective microfilarial suppression could potentially reduce the time necessary to interrupt transmission, easing the economic burden of mass treatment programs in countries with limited resources. METHODS: To determine the effect of increased dose and frequency of albendazole-ivermectin treatment on microfilarial clearance, 51 W. bancrofti microfilaremic residents of an area of W. bancrofti endemicity in Mali were randomized…

Macrofilaricidal activity after doxycycline treatment of Wuchereria bancrofti: a double-blind, randomised placebo-controlled trial.

BACKGROUND: Wolbachia endosymbionts of filarial nematodes are vital for larval development and adult-worm fertility and viability. This essential dependency on the bacterium for survival of the parasites has provided a new approach to treat filariasis with antibiotics. We used this strategy to investigate the effects of doxycycline treatment on the major cause of lymphatic filariasis, Wuchereria bancrofti. METHODS: We undertook a double-blind, randomised, placebo-controlled field trial of doxycycline (200 mg per day) for 8 weeks in 72 individuals infected with W bancrofti from Kimang'a villag…

Aedes albolateralis, a potential vector of nocturnally subperiodic Wuchereria bancrofti and dengue type 2 virus.

The susceptibility of Aedes albolateralis to nocturnally subperiodic Wuchereria bancrofti and dengue type 2 virus was investigated by using artificial membrane feeding and intrathoracic inoculation techniques, respectively. The results indicated that Ae. albolateralis was susceptible to nocturnally subperiodic W. bancrofti (susceptibility rate = 9.43%) and dengue type 2 virus (susceptibility rate = 100%), suggesting the potential vector of the two pathogens.

Wuchereria bancrofti microfilarial density of autochthonous cases and natural Culex infectivity rates in northeast Brazil.

Autochthonous Wuchereria bancrofti microfilaraemic cases from three neighbouring cities of Northeast Brazil (Recife, Jaboatao and Olinda) were selected from thick smear or filtration positive individuals and classified by their microfilarial (mf) densities through polycarbonate membrane blood filtration. Individuals were subdivided into two groups: autochthonous cases without any previous specific drug administrations (group A) and those with one or more previous DEC treatments (group B). In Recife, unlike Jaboatao and Olinda (P less than 0.05), the majority of autochthonous cases were over 20…

Random amplified polymorphic DNA (RAPD) for differentiation between Thai and Myanmar strains of Wuchereria bancrofti.

BACKGROUND: Lymphatic filariasis (LF) is a mosquito-borne disease caused by mosquito-transmitted filarial nematodes, including Wuchereria bancrofti and Brugia malayi. The Lymphatic Filariasis Elimination Program in Thailand has reduced the prevalence of nocturnally subperiodic W. bancrofti (Thai strain), mainly transmitted by the Ochlerotatus (Aedes) niveus group in Thailand to 0.57/100,000 population. However, it is estimated that more than one million Myanmar migrants with high prevalence of bancroftian filariasis have settled in the large urban cities of Thailand. These infected migrants ca…

[A study of ivermectin in the treatment of lymphatic filariasis due to Wuchereria bancrofti var. pacifica in French Polynesia].

Forty carriers of 20 or more W. bancrofti var. pacifica per ml were blind administered ivermectin at 50, 100, 150 or 200 mcg/kg doses. The rate of successful treatment was 100% with the 4 dosages. The percentage cure rate and the decrease percentage in the microfilarial count were significantly higher in persons treated with 100, 150 and 200 mcg/kg than in persons treated with 50 mcg/kg. Frequency and intensity of side-reactions were similar to those observed during treatment with DEC; they were more frequent and severe in persons with greatest microfilaremia but did not depend on the dosage.

In silico identification and prioritization of potential drug targets in Wuchereria bancrofti: Profilin as a promising therapeutic candidate.

Wuchereria bancrofti, a major causative agent of lymphatic filariasis, poses a significant public health burden in tropical and subtropical countries. Despite the availability of antifilarial drugs used in mass drug administration programs, issues such as non-compliance, low coverage, emerging resistance, and incomplete parasite clearance underscore the need for novel therapeutic targets. In this study, we analyzed potential drug targets identified from our previous subtractive proteomic analysis of W. bancrofti. Computational analysis of these targets highlighted Profilin as a potential candi…

Describing the interaction between Wuchereria bancrofti glutathione transferase and Bromosulphophthalein from an empirical and theoretical perspective.

Wuchereria bancrofti glutathione transferase (WbGST) is a therapeutic target in the parasitic worms that cause lymphatic filariasis. We described the molecular and structural basis of WbGST inhibition by Bromosulphophthalein (BSP) and compared it with human Mu (hGSTMu) and Pi (hGSTPi) class GSTs. BSP inhibited WbGST activity with an IC50 of 5 μM, 13.5 μM for the hGSTMu, and 110 μM for hGSTPi. Fluorescence spectroscopy showed that BSP altered polarity of intrinsic tryptophan residue environment and decreased the interaction between the enzymes and 8-Anilino-1-naphthalenesulphonate (ANS), a know…

Characterization of glycan determinants that mediate recognition of the major Wuchereria bancrofti circulating antigen by diagnostic antibodies.

The Global Program to Eliminate Lymphatic Filariasis (GPELF) relies heavily on a rapid diagnostic test (RDT) to a Wuchereria bancrofti circulating filarial antigen (Wb-CFA) to identify endemic areas and for determining when mass drug administration can stop. The antigen contains a carbohydrate epitope that is recognized by monoclonal antibody AD12. Og4C3, a monoclonal antibody that is used in a commercial ELISA for Wb-CFA recognizes the same moiety. Despite its diagnostic importance, little is known about the structure and function of this "AD12 epitope". It is also present on other W. bancrof…

Wuchereria bancrofti infection in Haitian immigrants and the risk of re-emergence of lymphatic filariasis in the Brazilian Amazon.

INTRODUCTION:: Lymphatic filariasis (LF) is a public health problem in Haiti. Thus, the emigration of Haitians to Brazil is worrisome because of the risk for LF re-emergence. METHODS:: Blood samples of Haitian immigrants, aged ≥18 years, who emigrated to Manaus (Brazilian Amazon), were examined using thick blood smears, membrane blood filtration, and immunochromatography. RESULTS:: Of the 244 immigrants evaluated, 1 (0.4%) tested positive for W. bancrofti; 11.5% reported as having received LF treatment in Haiti. CONCLUSIONS:: The re-emergence of LF in Manaus is unlikely, due to its low prev

An alternative strategy to generate coding sequence of macrophage migration inhibitory factor-2 of Wuchereria bancrofti.

BACKGROUND & OBJECTIVES: Different developmental stages of Wuchereria bancrofti, the major causal organism of lymphatic filariasis (LF), are difficult to obtain. Beside this limitation, to obtain complete coding sequence (CDS) of a gene one has to isolate mRNA and perform subsequent cDNA synthesis which is laborious and not successful at times. In this study, an alternative strategy employing polymerase chain reaction (PCR) was optimized and validated, to generate CDS of Macrophage migration Inhibitory Factor-2 (wbMIF-2), a gene expressed in the transition stage between L3 to L4. METHODS: The…
