Xeroderma pigmentosum variant with multisystem involvement.

BACKGROUND: Xeroderma pigmentosum (XP) is a hereditary disorder characterized by recessive inheritance and elevated rates of skin carcinogenesis. There are seven complementation groups (A through G) for which the genetic defect results in a failure to repair DNA damage from UV light and sunlight; one group, the variant, fails to replicate UV-damaged DNA correctly. Patients in XP groups A, B, D, and G have associated neurologic problems, the most severe being known as the DeSanctis-Cacchione syndrome. OBSERVATIONS: We describe a patient with XP from consanguineous parents who has severe multis…

Xeroderma pigmentosum variant: response to etretinate.

We report a case of the variant form of xeroderma pigmentosum in whom tumour development was completely suppressed for 22 months by etretinate 25 mg daily.

[DNA repair defect in xeroderma pigmentosum].

Xeroderma pigmentosum (XP) is an autosomal recessive human disease, clinically characterized by the early onset of severe photosensitivity of exposed skin to sunlight, a very high incidence of skin cancers and frequent neurological abnormalities. Cells from XP patients are hypersensitive to killing by UV-light, because they have a defect in repair of UV-light induced DNA damage. Genetic complementation analysis by cell fusion has led to the identification of at least ten genetic complementation groups, designated as group A through I, and a variant. However, the genetic basis of the physiologi…

Renal leiomyosarcoma associated with xeroderma pigmentosum.

We report a case of leiomyosarcoma of the kidney, presenting in the course of xeroderma pigmentosum, which debuted clinically as Wünderlich's syndrome due to spontaneous rupture of the tumor. We describe the case history and discuss the cutaneous disorder, analyzing the clinical and etiopathogenic factors, and conclude with an attempt to establish a relationship between the two pathologies.

Antioxidant enzymes in xeroderma pigmentosum fibroblasts.

In light of recent studies implicating low catalase activities in the pathogenesis of the cancer-prone disease xeroderma pigmentosum (XP) we have measured catalase activity, protein levels, and mRNA concentrations in six XP fibroblast strains and three normal controls. Only one XP strain of complementation group A (XP1223) possessed significantly lower catalase by all three criteria. The other five XP strains (two XP variants, two strains of complementation group D, and one strain of complementation group C) possessed catalase levels which fell into the range of the interindividual variations …

Genetic complementation analysis of xeroderma pigmentosum.

To characterize the mutations in different unrelated xeroderma pigmentosum (XP) patients, a complementation test was performed using the technique of somatic cell hybridization. DNA repair following UV exposure was studied in multinuclear cells resulting from fusions between in vitro cultivated cells from different patients. The parental XP cells performed low or negligible levels of repair DNA synthesis, whereas in some combinations binuclear hybrid cells showed repair DNA synthesis as a result of complementation. The results indicate the presence of five different complementation groups in X…

Five complementation groups in xeroderma pigmentosum.

A collaborative study was undertaken to determine the relationship between the three DNA repair complementation groups in xeroderma pigmentosum found at Erasmus University, Rotterdam, and the four groups found at the National Institutes of Health, Bethesda. The results of this study reveal that there are five currently known complementation groups in xeroderma pigmentosum.
