Resonance Raman detection of carotenoid antioxidants in living human tissue.

Increasing evidence points to the beneficial effects of carotenoid antioxidants in the human body. Several studies, for example, support the protective role of lutein and zeaxanthin in the prevention of age-related eye diseases. If present in high concentrations in the macular region of the retina, lutein and zeaxanthin provide pigmentation in this most light sensitive retinal spot, and as a result of light filtering and/or antioxidant action, delay the onset of macular degeneration with increasing age. Other carotenoids, such as lycopene and beta-carotene, play an important role as well in th

Potential role of myeloid bodies in protection against photo-oxidative damage of the retinal pigment epithelium.

Myeloid bodies are organized forms of the smooth endoplasmic reticulum that occur in the retinal pigment epithelium of vertebrates. The latter is a monolayer of cells, situated between the neural retina and the chroroid vasculature, that is essential for photoreceptor homeostasis and function. Myeloid bodies operate in an environment of high oxygen tension and chronic light exposure where phagocytosis of photoreceptor outer segments takes place, resulting in photo-oxidative stress. Over time, such damage can result in pigment epithelium cell death, retinal degeneration and blindness. The purpo

Macular Xanthophylls and Markers of the Anterior Visual Pathway among Persons with Multiple Sclerosis.

BACKGROUND: Multiple sclerosis (MS) can cause retinal thinning among persons with MS with optic neuritis (MS-ON). Macular xanthophylls are carotenoids that comprise the macular pigment, filtering blue light and countering photo-oxidation. However, macular xanthophyll status and its implications for markers of neuroaxonal degeneration have not been examined in MS. OBJECTIVES: This study characterized differences in macular and serum xanthophylls, and retinal morphometry [retinal nerve fiber layer thickness at the macular (mRNFL) and optic disc (odRNFL) and total macular volume (TMV)] in indivi

Bioactive molecules and antioxidant activity in durum wheat grains and related millstream fractions.

The present study was performed to evaluate the distribution of phytochemicals in wheat grains and millstreams and the contribution of these compounds to the antioxidant activity. Bioactive molecules, although related to genotype, were highly affected by the milling procedure applied. Carotenoids were unevenly distributed in the kernel and lutein resulted to be the main carotenoid in all samples considered. The WSB method was confirmed to be a useful tool to detect yellow color intensity of semolina but it was not suitable to get information on the true content of carotenoids of the sample. Wa

Dietary carotenoids are associated with cardiovascular disease risk biomarkers mediated by serum carotenoid concentrations.

Hyperlipidemia and elevated circulating C-reactive protein (CRP) and total homocysteine (tHcy) concentrations are cardiovascular disease (CVD) risk factors. Previous studies indicated that higher serum carotenoid concentrations were inversely associated with some of these biomarkers. However, whether dietary carotenoid intake is inversely associated with these CVD risk biomarkers is not well known. We assessed the associations between individual dietary carotenoid intake and CVD risk biomarkers and tested whether the serum carotenoid concentrations explain (mediate) or influence the strength o

Chemistry, distribution, and metabolism of tomato carotenoids and their impact on human health.

Recent epidemiological studies have suggested that the consumption of tomatoes and tomato-based food products reduce the risk of prostate cancer in humans. This protective effect has been attributed to carotenoids, which are one of the major classes of phytochemicals in this fruit. The most abundant carotenoid in tomato is lycopene, followed by phytoene, phytofluene, zeta-carotene, gamma-carotene, beta-carotene, neurosporene, and lutein. The distribution of lycopene and related carotenoids in tomatoes and tomato-based food products has been determined by extraction and high-performance liquid

The effect of macular carotenoids and omega-3 supplementation on performance in precision sports: the sport nutrition intervention in performance exercise (SNIPE) study.

BACKGROUND: Macular carotenoids improve visual performance in healthy individuals, but these benefits have yet to be tested on athletes. The SNIPE study examined how the macular carotenoids [lutein (L), zeaxanthin (Z), meso-zeaxanthin (MZ)] with omega-3 fatty acids (ω3) impact visual functions and sports performance in precision sports. METHODS: In this double-blind, placebo-controlled trial, 67 athletes and players randomly received either an active supplement containing L, Z, MZ + ω3 (the formulation), or a placebo containing sunflower oil over a 9 month period. Key measurements included bl

Xanthophyll acyl esters in pepper ( Capsicum annuum ): Delineating the role of two xanthophyll acyltransferases CaPYP1 and CaPYP1-like via gene silencing and genetic complementation.

UNLABELLED: Pepper fruit accumulated xanthophyll acyl esters during ripening. To characterize the genes involved, cDNAs for two putative xanthophyll acyltransferases named CaPYP1 and CaPYP1-like were cloned. The deduced amino acid sequences of CaPYP1 and CaPYP1-like had conserved hydrolase and acyltransferase domains. Phylogenetic analyses showed that both putative acyl transferases were conserved in Viridiplantae , suggesting important functions for both. CaPYP1 and CaPYP1-like GFP-fusion proteins were localized in the plastid when expressed in leaf tissue. Gene expression analyses indicated

Preterm Pigs Fed Donor Human Milk Have Greater Liver β-Carotene Concentrations than Pigs Fed Infant Formula.

BACKGROUND: Milk carotenoids may support preterm infant health and neurodevelopment. Infants fed human milk often have higher blood and tissue carotenoid concentrations than infants fed carotenoid-containing infant formula (IF). Donor human milk (DHM) is a supplement to mother's own milk, used to support preterm infant nutrition. OBJECTIVES: We tested whether tissue and plasma β-carotene concentrations would be higher in preterm pigs fed pasteurized DHM versus premature IF. METHODS: This is a secondary analysis of samples collected from a study of the effects of enteral diet composition on n

Lutein exerts anti-inflammatory effects in patients with coronary artery disease.

BACKGROUND AND AIMS: Many coronary artery disease (CAD) patients exhibit chronic low-grade inflammation. Carotenoids are anti-oxidants with potential anti-inflammatory properties. Here, we first assessed relationships between interleukin (IL)-6 and individual carotenoids in plasma from CAD patients. Based on the results, we proceeded to assess anti-inflammatory effects of one carotenoid, lutein, in peripheral blood mononuclear cells (PBMCs) from CAD patients. METHODS: Lutein + zeaxanthin (isomers with lutein being dominant), β-cryptoxanthin, lycopene, α- and β-carotene and IL-6 were measured

A mathematical model of non-photochemical quenching to study short-term light memory in plants.

Plants are permanently exposed to rapidly changing environments, therefore it is evident that they had to evolve mechanisms enabling them to dynamically adapt to such fluctuations. Here we study how plants can be trained to enhance their photoprotection and elaborate on the concept of the short-term illumination memory in Arabidopsis thaliana. By monitoring fluorescence emission dynamics we systematically observe the extent of non-photochemical quenching (NPQ) after previous light exposure to recognise and quantify the memory effect. We propose a simplified mathematical model of photosynthesis

Analysis of geometrical isomers of dietary carotenoids.

Carotenoids are versatile isoprenoids of great importance in food science and nutrition. These compounds are natural colorants and key to combat vitamin A deficiency, a major global nutritional problem. In addition, carotenoids have health-promoting biological actions that can contribute to reduce the risk of serious diseases, including cancer, cardiovascular disease, skin, bone and eye conditions and metabolic disorders. Beneficial roles of carotenoids in cognition and fetal development are also attracting increased attention. The presence of double bonds cause carotenoids to be prone to geom
