Vitamin D alleviates neurotoxicity induced by propofol anaesthesia in the offspring of mice.

BACKGROUND: Maternal propofol can induce neurotoxicity in the developing brain. Vitamin D modulates neurotrophin and neuromediator synthesis, reduces neuroinflammation and neuroapoptosis. The present study examined whether vitamin D could mitigate maternal propofol induced neurotoxicity in the offspring. METHOD: Twenty pregnant mice were randomly divided into four groups. During the pregnancy, the groups designated as D and PD were given intraperitoneal vitamin D, while the groups labelled as P and C received 1 ml of saline. On the 14th day of pregnancy, groups P and PD received 200 mg kg-1 o

Cannabidiol (CBD) potentiates physiological and behavioral markers of hypothalamic-pituitary-adrenal (HPA) axis responsivity in female and male mice.

RATIONALE: Clinical literature indicates there may be a therapeutic use of cannabidiol (CBD) for stress-related disorders. Preclinical literature remains conflicted regarding the underlying neurobehavioral mechanisms, reporting mixed effects of CBD (increased, decreased, or no effect) on anxiety- and fear-related behaviors. Preclinical data demonstrated that CBD modulates hypothalamus-pituitary-adrenal (HPA) axis gene expression; it is unknown whether CBD changes HPA axis responsivity and how this relates to altered behavior. OBJECTIVES: We aimed to evaluate whether acute or chronic CBD admin

Administering MSC-Derived Exosomes After Hypoxia-Induced Seizures in Neonatal Rats Improved Cognitive Function and Delayed the Onset of Epilepsy in Adulthood, Likely by Reducing Inflammation and Oxidative Stress.

Hypoxia-induced neonatal seizures (HINSs) are a major cause of long-term cognitive deficits and heightened epilepsy risk in adulthood. Early inflammatory responses following HINS contribute to these pathological outcomes. This study examined the sustained neuroprotective benefits of exosomes derived from mesenchymal stem cells (MSC-exosomes) in a rat model of HINS, leveraging their anti-inflammatory and neuroregenerative properties. Forty-nine male and female Wistar rats were divided into four groups: (1) control + saline, (2) control + exosome, (3) hypoxia + saline, and (4) hypoxia + exosome.

Unmoving and uninflamed: Characterizing neuroinflammatory dysfunction in the Wistar-Kyoto rat model of depression.

Reductionistic research on depressive disorders has been hampered by the limitations of animal models. Recently, it has been hypothesized that neuroinflammation is a key player in depressive disorders. The Wistar-Kyoto (WKY) rat is an often-used animal model of depression, but no information so far exists on its neuroinflammatory profile. As such, we compared male young adult WKY rats to Wistar (WS) controls, with regard to both behavioral performance and brain levels of key neuroinflammatory markers. We first assessed anxiety- and depression-like behaviors in a battery consisting of the Eleva

Postweaning Iron Deficiency in Male Rats Leads to Long-Term Hyperactivity and Decreased Reelin Gene Expression in the Nucleus Accumbens.

BACKGROUND: Epidemiological research indicates that iron deficiency (ID) in infancy correlates with long-term cognitive impairment and behavioral disturbances, despite therapy. However, the mechanisms underlying these effects are unknown. OBJECTIVE: We investigated how ID affected postweaning behavior and monoamine concentration in rat brains to determine whether ID during the juvenile period affected gene expression and synapse formation in the prefrontal cortex (PFC) and nucleus accumbens (NAcc). METHODS: Fischer 344/Jcl postweaning male rats aged 21-39 d were fed low-iron diets (0.35 mg/k

Repurposing Leucovorin for Mild Traumatic Brain Injury: Evidence from Biochemical and Behavioral Outcomes in Rats.

Background/Objectives: Mild traumatic brain injury (mTBI) is common and may result in persistent cognitive and affective disturbances driven, at least in part, by delayed secondary injury mechanisms, including oxidative stress, neuroinflammation, apoptosis-related signaling, and impaired neuroplasticity. Pharmacological strategies targeting these interconnected processes remain limited. The present study investigated leucovorin, also known as folinic acid, a clinically approved reduced folate, as a potential repurposing candidate in an experimental model of mTBI. Methods: Male Wistar rats were

Alterations in stress-associated behaviors and neurochemical markers in adult rats after neonatal short-lasting local inflammatory insult.

Recently, there has been a growing interest in long-term consequences of neonatal pain because modern neonatal intensive care units routinely employ procedures that cause considerable pain and may be followed by local inflammation and hyperalgesia lasting for several hours or even days. To address this question, we developed a rat model of short lasting (<2 days) early local inflammatory insult produced by a single injection of 0.25% carrageenan (CAR) into the plantar surface of a hindpaw. Previously, we demonstrated that rats receiving this treatment within the first week after birth grow int

Sex-dependent central amygdala physiology after supradural CGRP exposure is associated with migraine-like behaviors.

BACKGROUND: Calcitonin gene-related peptide (CGRP) is a key mediator of migraine, but the mechanisms through which meningeal CGRP engages central circuits involved in the sensory and affective dimensions of migraine remain incompletely understood. Here, we tested if supradural CGRP is sufficient to induce migraine-like sensory and affective behaviors and if these effects are associated with sex-dependent changes in parabrachial-central amygdala (PB-CeA) signaling. METHODS: Adult male and female C57BL/6J mice received supradural CGRP or vehicle. Behavioral testing, whole-cell patch-clamp recor

Estradiol does not affect spasms in the betamethasone-NMDA rat model of infantile spasms.

OBJECTIVE: This study attempted to validate the effects of neonatal estradiol in ameliorating the spasms in the prenatally betamethasone-primed N-methyl-d-aspartate (NMDA) model of infantile spasms in rats as shown previously in a mouse Arx gene knock-in expansion model of infantile spasms. METHODS: Neonatal rats prenatally exposed to betamethasone (on day 15 of pregnancy) were treated with subcutaneous 40 ng/g estradiol benzoate (EB) between postnatal days (P)3-P10 or P0-P5. A synthetic estrogen analogue, diethylstilbestrol, was used between P0 and P5 (2 μg per rat, s.c.). On P12, P13, and P

Fear extinction learning ability predicts neuropathic pain behaviors and amygdala activity in male rats.

Background The amygdala plays a key role in fear learning and extinction and has emerged as an important node of emotional-affective aspects of pain and pain modulation. Impaired fear extinction learning, which involves prefrontal cortical control of amygdala processing, has been linked to neuropsychiatric disorders. Here, we tested the hypothesis that fear extinction learning ability can predict the magnitude of neuropathic pain. Results We correlated fear extinction learning in naive adult male rats with sensory and affective behavioral outcome measures (mechanical thresholds, vocalizations,

Behavioral profile predicts ethanol preference in adolescent mice, but not in adults: A machine learning approach.

BACKGROUND: Clinical studies suggest that adolescents display a complex behavioral profile characterized by traits that increase their susceptibility to alcohol experimentation and impaired control over use. In the present study, we applied a machine learning model to predict the impact of diverse behavioral phenotypes on ethanol preference during adolescence and adulthood in mice. METHODS: C57BL/6 and Swiss mice were assigned to one of two age groups: adolescents (starting at PN40) or adults (starting at PN120). Over the next 3 days, novelty-seeking, anxiety-like behavior, sociability, copin

Alcohol or Stress Exposure During Late Adolescence Impairs Risk Assessment Later in Life, Disrupting the Ethological Structure of Behavior in Mice.

BACKGROUND: Decision-making relies on risk assessment as a critical process that enables organisms to evaluate threats before exploring potentially dangerous areas. While adolescence is a period of heightened vulnerability to external factors such as substance use and stress, it remains unclear how these factors disrupt risk assessment, particularly at the level of behavior structure: sequential actions, organization and transition probabilities between defensive and exploratory behaviors. We investigated whether repeated ethanol intoxication or pharmacological stress during late adolescence a

mGluR2/3 receptor agonist LY379268 alleviates PTSD-like fear memories by downregulating the CaMKII/CREB signaling pathway in the hippocampus and prefrontal cortex.

Post-traumatic stress disorder (PTSD) is a mental health condition marked by fear memories triggered by life-threatening traumatic events. The formation and consolidation of these fear memories are central to PTSD, making it important to understand the underlying neurobiological mechanisms that regulate memory plasticity. In this context, LY379268, a selective mGluR2/3 agonist, induces presynaptic long-term depression (LTD) and modulates excitatory transmission, potentially affecting memory consolidation. Propranolol, a β-adrenergic antagonist, similarly influences fear memory through synaptic

Off-targets effects of CNO on somatosensory and anxiety-related behaviors in rats.

Designer receptors exclusively activated by designer drugs (DREADDs) are a powerful chemogenetic tool for controlling targeted cell populations and manipulating brain functions. As a typical chemogenetic actuator in DREADD systems, clozapine-N-oxide (CNO) is widely utilized in neurobiological and pharmacological research. However, the acute off-target effects of CNO on somatosensory (e.g., itch, pain) stimulation-induced and anxiety-related behaviors in rodents have not been systematically investigated. Through classical behavioral paradigms, including evoked itch-scratching, noxious thermal p

[Hippocampal stereotactic injection of cerebrospinal fluid from single-prolonged stress mice induces PTSD-like behavior].

This study aimed to investigate the impact of cerebrospinal fluid (CSF) from single-prolonged stress (SPS) model mice on the neurobehavioral outcomes in normal mice, and to explore the underlying mechanisms. CSF was extracted from SPS mice (SPS-CSF), and norepinephrine (NE) levels were measured using high-performance liquid chromatography-electrochemical detection (HPLC-ECD). A cannula was used for direct hippocampal injection of SPS-CSF, followed by behavioral experiments such as the elevated plus maze and fear extinction tests to assess changes in anxiety and fear memory in SPS-CSF injected

Optogenetic inhibition of ventrolateral orbitofrontal cortex astrocytes facilitates ventrolateral periaqueductal gray glutamatergic activity to reduce hypersensitivity in infraorbital nerve injury rat model.

BACKGROUND: Trigeminal neuropathic pain (TNP) is a chronic condition characterized by heightened nociceptive responses and neuroinflammatory changes. While astrocytes are recognized as critical players in pain modulation, their specific role in influencing descending trigeminal pain pathways via ventrolateral orbitofrontal cortex (vlOFC) activity modulation remains underexplored. Therefore, we investigated the impact of optogenetic modulation of astrocytes in the vlOFC on pain hypersensitivity in a rat model of chronic constriction injury of the infraorbital nerve (CCI-ION). METHOD: Adult fem

Resolving neuroinflammatory and social deficits in ASD model mice: Dexmedetomidine downregulates NF-κB/IL-6 pathway via α2AR.

Autism spectrum disorder (ASD) is a neurodevelopmental disorder that severely affects individuals' daily life and social development. Unfortunately, there are currently no effective treatments for ASD. Dexmedetomidine (DEX) is a selective agonist of α2 adrenergic receptor (α2AR) and is widely used as a first-line medication for sedation and hypnosis in clinical practice. In recent years, there have been reports suggesting its potential positive effects on improving emotional and cognitive functions. However, whether dexmedetomidine has therapeutic effects on the core symptoms of ASD, namely so
